Effect of gangliosides on the Adhesion of Neuroblastom Cells to Collagen

Objective To study the effect of endogeneous ganglioside (GS) on the adhesion of LA-N5 neuroblastoma cells to collagen. Methods LA-N5 cells were cultured in the presence of D-threo-1-phenyl-2-decanolamino-3-morphinoline-1-propanol (D-PDMP), an inhibitor of glucosylceramide synthase. The adhesion of the LA-N5 cells to immobilized collagen was tested. Results After 6 days, endogenous GS was reduced by 98%. No change in the cell morphology, viability, proliferation rate or percentage of apoptotic cells was observed. The adhesion to collagen of cells exposed to D-PDMP was reduced by 65% compared to the control LA-N5 cells: OD 570 0.07± 0.01 vs 0.21± 0.030(P 0.05). Similarly, the pre-incubation of GS-depleted cells with purified tumor GS GD 2, the most abundant GS in LA-N5 cells, restored adhesion. Conclusions Endogenous tumor GS regulates neuroblastoma cell adhesion to collagen, suggesting that it may play a role in tumor cell migration, invasion and metastasis.

Guillain-Barrésyndrome(GBS)is often triggered by a preceding bacterial or viral ***,it has been observed in association with acute hepatitis A,B and C,and three cases have been previously described in India in which GBS was associated with acute hepatitis E.A molecular mimicry mechanism is supposed to be involved in the pathogenesis of GBS triggered by infectious agents,although the nature of the shared epitopes has not been characterized in most instances,including that in the case of hepatotropic *** report a case of GBS following acute hepatitis E in a European *** presence of antiganglioside GM2 antibodies in this patient suggested molecular mimicry involving ganglioside GM2 in the pathogenesis of GBS associated with hepatitis E.

Can Campylobacter jejuni play a role in development of celiac disease? A hypothesis

Celiac disease （CD） is an entropathy with malabsortive condition in which an allergic reaction to the cereal grain-protein （gluten） causes small intestine rnucosal injury. CD is a multifactorial disorder in which both genetic and environmental factors contribute to the disease development. Mechanisms have been described to explain the pathology of CD. T cells specific for multiple gluten peptides are found in virtually all patients. Generation of such a broad T cell response may be a prerequisite for disease development. CD is associated with multiple extraintestinal presentations, including neurological deficits. Recent studies have shown a significant correlation between anti-ganglioside antibodies and neurological disorders in patients with underlying CD. gangliosides are glycosphingolipids which are abundant in nervous system and in other tissues including gastrointestinal tract. It is not known what triggers the release of anti-ganglioside antibodies in people with gluten sensitivity. But, the mechanism is likely to involve the intestinal immune system response to ingested gliadin, a component of wheat gluten. Studies showed that mechanisms different from gluten exposure may be implicated in antibody formation, and other environmental factors may also exist. In addition, considering the fact that genetic predisposition dysregulating mucosal immune responses in the presence of certain environmental triggers like gastrointestinal infections may be strong etiological factors for developing chronic intestinal inflammation including CD, the hypothesis raised in our mind that antiganglioside antibody formation in CD may play a role not only in development of neurological complications in celiac patients, but also in development of CD itself. GM1 is a common ganglioside pentasaccharide present on mammalian cell *** has been shown to play important roles in cellular communications and initiation of β-amyloid *** order to synthesize GM1,an efficient synthetic route was developed via a [3+2] *** GM3 trisaccharide acceptor bearing an azido propyl group at the reducing end was prepared using the traditional acetamide protected sialyl thioglycosyl donor,which gave better stereoselectivity than sialyl donors protected with trichloroacetamide or *** glycosylation of the axial 4-hydroxyl group of GM3 by the disaccharide donor was found to be highly dependent on donor protective *** bearing the more rigid benzylidene group gave low glycosylation *** the benzylidene with acetates led to productive coupling and formation of the fully protected GM1 *** of the pentasaccharide produced GM1 functionalized with the aminopropyl side chain,which will be a valuable probe for biological studies.

Objective: To observe the effects of ganglioside GM1 on reduction of brain edema and amelioration of cerebral metabolism after traumatic brain injury (TBI). Methods: An acute experimental closed TBI model in rats was induced by a fluid percussion brain injury model. At five and sixty minutes after TBI, the animals were intraperitoneally injected by ganglioside GM1 (30 mg/kg) or the same volume of saline. At the 6th hour after TBI, effects of ganglioside GM1 or saline on changes of mean arterial pressure (MAP), contents of water, lactic acid (LA) and lipid peroxidation (LPO) in the injured cerebral tissues were observed. Results: After TBI, MAP decreased and contents of water, LA and LPO increased in brain injury group; however, MAP was back to normal levels and contents of water, LA and LPO decreased in ganglioside GM1 treated group, compared with those in brain injury group (P 0.05 ) was ***: Ganglioside GM1 does have obvious neuroprotective effect on early TBI.
