The 21st century ＂Maritime Silk Road＂ strategy is a significant part of the belt and road initiatives of China. The cognition and investigation of ocean environment is essential and necessary in these regions which will provide scientific reference for many fields such as navigation, ocean engineering, and disaster prevent and reduction. A high-resolution cross-calibrated multi-platform wind product is used to analyze gales over the Maritime Silk Road. The yearly mean speed and space distribution of gale, and the frequencies and trends of gale and extreme wind speed are analyzed. The results show that relatively high pools of gale are mainly located in the waters of the Arabian Sea, the Somali Sea, Indo-China Peninsula sea area, and Bay of Bengal in the summer. The gale frequency of the Somali Sea is more than 90%. Overall, the gale days increase year by year in the majority of the South China Sea and the northern Indian Ocean, especially in the autumn and the winter.

In this study, coastal gales and rainfall attributed to the landfall of Typhoon Soudelor(2015) are analyzed based on observational dense automatic weather stations data, advanced scatterometer-retrieved 10-m ocean surface wind data and simulations using the Weather Research and Forecast(WRF) model. This study focuses on gale bands in the right-front quadrant of the typhoon and associated coastal winds over Zhejiang and Fujian Provinces in China before the landfall of the typhoon. The results are summarized as follows.(1) 10-m surface wind data from automatic weather stations over land and islands, advanced scatterometer-retrieved 10-m ocean surface wind data, and the WRF simulation indicate similar mesoscale offshore gales.(2) The model simulation with a 333-m grid mesh indicates a gale zone over the right-front quadrant of the typhoon; the gale is "broken" over the coastal areas, and formed an inhomogeneous gale band.(3) The model-simulated winds agree well with the island observations.(4) Non-uniform gales over boundary layers result in horizontal wind-speed gradients and strong convergence that favors the development of convection and the maintenance of ocean surface gales.

宿主天然免疫系统可以识别病原菌并诱导炎症应答,从而清除病原菌,这是宿主抵御微生物入侵的重要防线。炎症小体是国内外热门的研究领域,其中研究得最广泛的是 NLRC4（NLR family CARD domain-containing protein4）炎症小体和 NLRP3（NLR family pyrin domain-containing 3）炎症小体,它们在抵御病原微生物感染的过程中发挥重要作用。炎症小体是由模式识别受体（PRRs）,ASC接头蛋白和pro-Caspase-1组装形成的多蛋白复合物,pro-Caspase-1会发生自裂解成为活化形式,活化的Caspase-1能够促进细胞因子IL-1β和IL-18的成熟和分泌,并诱导细胞焦亡（Pyroptosis）。沙门菌（Salmonella）是一种重要的人兽共患病原菌,在感染宿主的过程中,细菌可以利用复杂的策略逃避免疫系统的清除,得以在宿主细胞内存活和复制,进而形成持续性感染。然而,沙门菌如何抑制炎症小体的活化逃避宿主免疫系统的清除在很大程度上仍然是未知的。基于此,本研究以肠炎沙门菌C50336ΔfliC（以下简称ΔfliC）为研究对象,构建转座子突变体文库,筛选参与调控炎症小体活化的基因,并探究其调控炎症小体活化的机制,为肠炎沙门菌新型防控技术和措施的建立提供新的思路。1肠炎沙门菌抑制炎症小体活化基因的筛选本研究以ΔfliC为亲本株,构建了肠炎沙门菌转座子随机插入失活突变体文库。以J774A.1细胞为感染模型,通过计算Z值高通量筛选诱导乳酸脱氢酶（LDH）释放水平具有显著性差异的突变株。共筛选了 1843株,诱导LDH释放水平显著性上调的有25株（Z ≥ 2）,显著性下调的有47株（Z ≤-2）。第二轮筛选对这些突变株在巨噬细胞中诱导炎症小体激活的能力进行了检测,结果显示,831号和1552号突变株在J774A.1细胞上稳定诱导更强的LDH释放、IL-1β分泌和Caspase-1活化水平。利用两轮半PCR对转座子插入序列进行定位,结果显示,这两株突变株的转座子分别插入在dinJ和gale的ORF中。dinJ编码沙门菌Ⅱ型毒素-抗毒素系统（TA）RelB/DinJ家族抗毒素,gale编码一种UDP-葡萄糖-4-差向异构酶。这些结果表明,肠炎沙门菌可能通过DinJ和gale抑制炎症小体的活化。分别构建了dinJ和gale的基因缺失突变株和回补株,并对这些菌株的生长特性和在J774A.1细胞上的黏附侵袭率进行了测定。结果显示,各缺失株和回补株之间的生长速率和J774A.1细胞上的黏附侵袭率均无显著差异。说明dinJ或gale的缺失不影响肠炎沙门菌的生长速率,也不影响其在J774A.1细胞上的黏附和侵袭能力。这些结果也表明肠炎沙门菌DinJ蛋白和gale蛋白抑制炎症小体的活化与黏附侵袭无关。2肠炎沙门菌DinJ蛋白抑制NLRP3炎症小体活化为了探究dinJ基因在调控炎症小体活化中发挥的作用,本研究用dinJ突变株和回补株感染J774A.1细胞,测定炎症小体活化的各项指标。结果显示,与ΔfliC亲本株相比,ΔfliCΔdinJ和ΔfliCΔdinJ::Vector感染的J774A.1细胞中检测到更强的LDH释放和IL-1β分泌,并且Caspase-1的活化也显著增强,ΔfliCΔdinJ::dinJ回补株感染组恢复到与ΔfliC亲本株相似的水平。野生型小鼠原代骨髓来源巨噬细胞（WT BMDMs）感染模型中也得到了相同的结果,ΔfliCΔdinJ缺失株诱导WT BMDMs的LDH释放、IL-1β分泌和Caspase-1活化水平显著高于ΔfliC亲本株感染组。这些结果表明肠炎沙门菌DinJ能够抑制炎症小体活化。此外,炎症小体各组分NLRP3、NLRC4和ASC蛋白的表达在各感染组之间无显著差异,表明DinJ抑制炎症小体的活化不是由抑制炎症小体组分的表达介导的。随后分别制备Casp1-/-、Nlrc4-/-和Nlrp3-/-BMDMs进一步探究DinJ蛋白抑制炎症小体的类型。在各菌株感染的Casp1-/-BMDMs中,检测不到IL-1β的分泌,LDH释放水平也降低,说明IL-1β的分泌依赖于Caspase-1。在Nlrc4-/-BMDMs感染组中,ΔfliC和ΔfliCΔdinJ::dinJ诱导的LDH释放、IL-1β分泌和Caspase-1活化均显著性低于ΔfliCΔdinJ,而在Nlrp3-/-BMDMs感染组中,dinJ缺失株和回补株诱导炎症小体活化的能力与亲本株相比均无明显差异。表明DinJ能够特异性抑制NLRP3依赖型炎症小体的激活,从而阻断炎性细胞因子的释放。本研究构建了 ΔfliC-pCX340和ΔfliC-pCX340-dinJ菌株,利用荧光共振能量转移（FRET）实验检

探讨甲状腺微小乳头状癌中半乳糖凝集素3(galectin-3)、细胞角蛋白19(CK19)表达变化及其意义。2013~2015年手术治疗的105例甲状腺肿瘤标本,其中49例甲状腺微小乳头状癌,31例结节性甲状腺肿不伴乳头状增生,25例结节性甲状腺肿伴乳头状增生,应用免疫组织化学方法检测galectin-3和CK19的表达。结果显示,微小乳头状癌galectin-3阳性表达率为93.88%,明显高于结节性甲状腺肿不伴乳头状增生的6.45%和结节性甲状腺肿伴乳头状增生的20.00%(Pgalectin-3敏感性为97.96%,特异性为89.29%;CK19敏感性为93.88%,特异性为87.50%;两者联合检测敏感性为84.20%,特异性为88.22%。结果表明,galectin-3和CK19联合检测可用于鉴别诊断甲状腺微小乳头状癌与良性病变,有重要的临床应用价值。

Based on the gale observation data in 5 observation stations in Jinzhou area from 1973 to 2007,the gale trend and the periodic change in Jinzhou area were analyzed by using the linear trend estimation method and the spectral analysis method. Meanwhile,the circulation situation characteristics and the spatial-temporal distribution characteristics of gale in four seasons were discussed by surveying generally the weather charts. The results showed that the gale in Jinzhou area presented the fluctuation decline trend and had 3.5,7.0 years periodic changes. The gale in spring is the most and in summer was the least. It was less in winter and autumn. Seen from the analysis on the circulation situation,the gale circulation situation in Jinzhou area was similar to in Liaoning. The circulation situation in spring was basically consistent with in autumn,winter. The main situations had 3 kinds:north high south low(west high east low) ,two high clipping low,south high north low(east high west low) . In summer,the regional gale weather was mainly caused by the meso-micro scale system.

Extreme Wind Characteristics of Coastal Communities in Bayelsa State: Implications for Community Planning and Development in Nigeria

The study examined extreme wind characteristics of the coastal communities in Bayelsa State, Nigeria for possible community planning and development. To achieve this aim, data on wind speed were sourced from the Nigerian Meteorological Agency (NIMET). Personal interview and focused group discussions were done with the aid of well structured questionnaire in the various coastal communities sampled to identify impact and coping strategies from extreme winds. The Beaufort Winds Scale and regression analysis were the statistical tools used for the analysis of the data in order to achieve the objectives of the study. The results indicate that, the return period to obtain maximum 1-year wind speed value of 29.3 m/s (violent storm), 27.8 m/s (storm), 24.3 m/s (strong gale) and 20.6 m/s (fresh gale) will be 17 years, 5.7 years, 2.8 years and 1.4 years respectively. Fresh gale characterized the extreme wind events in the area. Result further showed that out of 19 occurrences of wind events, 11 were extreme cases, while 2 occurred as violent storm of 29.3 m/s. Findings also showed that extreme winds occurred more (8 cases) during the early part (March-April) of the raining season when the area is under the influence of maritime moisture laden air mass than the dry season which is dried and dusty. Using a combination of return period of the magnitude of extreme wind and the log of wind speed for the 16 years a model predicting the incidence of extreme wind was done. Awareness on the dangers of wind hazard increases while early warning systems are advocated to mitigate the associated dangers with extreme wind events in the study area.

Physical model of self-sustained infrasonic air oscillations related to interaction of fresh gale with choppy sea surface is proposed. It is shown that air infrasonic oscillations are expected inside moving 3D cavities in sea surface generated by gale and detected far from its region. Interaction of wind with moving sea wave crests is shown to be of weaker impact on oscillations in far field. For wind velocity in the range from 10 to 40 m/s deepest cavities acquire resonance frequencies in the range of 3.0 - 0.7 Hz, i.e. frequencies much lower than their quarter wavelength resonance frequencies. In the course of oscillations effective wind velocity applied to cavities can achieve value from 0.4 to 0.6 of wind velocity, while air self-sustained oscillations velocity amplitude can run up in the range from 0.2 to 0.3 of wind velocity. Wind intensification leads to oscillations frequency decrease and oscillation energy losses increase with wind velocity cubed. Cavities natural frequencies are transformed due to air attached mass and volume elasticity additional transformation under wind influence in the range from 1.05 to 1.9 with respect to resonance frequencies at rest. Amplitude of self-sustained oscillation in atmosphere is expected to increase with wind velocity cubed, while cavity air oscillation velocity-linear with wind velocity. Wind velocity threshold of an order of 25 - 30 m/s overcome is necessary to observe effect. Spectral peaks on resonance frequencies in the range 0.7 - 2.5 Hz are expected in effect observation. Infrasonic signals observable far from whole gale in atmosphere, sea water thickness and earth crust on self-sustained oscillation frequency and its harmonics frequencies beginning from third harmonic 2.1 - 7.5 Hz are regarded as phenomenon signs.
