Social dysfunction is a risk factor for several neuropsychiatric illnesses.Previous studies have shown that the lateral septum(LS)-related pathway plays a critical role in mediating social behaviors.Howeve r,the role of the connections between the LS and its downstream brain regions in social behavio rs remains unclea r.In this study,we conducted a three-chamber test using electrophysiological and chemogenetic approaches in mice to determine how LS projections to ventral CA1(vCA1)influence sociability.Our res ults showed that gamma-aminobutyric acid(gaba)-e rgic neuro ns were activated following social experience,and that social behavio rs were enhanced by chemogenetic modulation of these neurons.Moreover,LS gabaergic neurons extended their functional neural connections via vCA1 glutamatergic pyramidal neurons,and regulating LSgaba→vCA1Gluneural projections affected social behaviors,which were impeded by suppressing LSprojecting vCA1 neuronal activity or inhibiting gabaAreceptors in vCA1.These findings support the hypothesis that LS inputs to the vCA1 can control social prefe rences and social novelty behaviors.These findings provide new insights rega rding the neural circuits that regulate sociability.

Novel alternative splicing of gaba receptor RDL exon 9 from Laodelphax striatellus modulates agonist potency

The resistance to dieldrin gene(RDL)encodes the primary subunit of the insect ionotropic y-aminobutyric acid(gaba)receptor(gabaR),which is the target of phenylpyrazole and isoxazoline insecticides.The splice variants in exons 3 and 6 of RDL,which have been widely explored in many insects,modulate the agonist potency of the homomeric RDL gabaR and potentially play an important role in the development of insects.In the present study,four splice variants of exon 9 were identified in RDL of the small brown planthopper,Laodelphax striatellus(LsRDL),resulting in LsRDL-9sl,LsRDL-92l\LsRDL-9b,and LsRDL-9c.LsRDL-9a has one more amino acid(E,glutamic acid)compared with LsRDL-9a\and LsRDL-9b lacked two amino acids and had seven different amino acids compared with LsRDL-9c.Two-electrode voltage-clamp recording on LsRDLs expressed in Xenopus oocytes showed that alternative splicing of exon 9 has significant impact on LsRDL sensitivity to the agonists gaba and 0-alanine,whereas no significant difference was observed in the potencies of the non-competitive antagonists(NCAs)ethiprole and fluralaner on the splice variants.Our results suggest that alternative splicing of RDL exon 9 broadens functional capabilities of the gabaR in L.striatellus by influencing the action of gaba.

Preliminary Study on γ-Aminobutyric Acid (gaba_A) Like receptor in Rat Testis

For making it clear whether gabaA-like receptor is in cells or rat testis and how itsgene expresses in tissue cells, the mRNA of rat testis was microinjected into Xenopusoocyte. The membrane electrobiological results showed that an inward 30 nA micro-currentwas elicited, under two-electrode voltageclamped configuration, by extracellular gaba(500 μmol/L), and both Bicuculline and Picrotoxin could inhibit this effect. It is an indication that the mRNA of rat testis can express gaba receptor in the membrane of Xenonpus oocyte. Using Muscimol as a excitant of gaba receptor type A, the micro--currentwas also elicited to generate; however, using Baclofen as a excitant of gaba receptortype B, the micro-current could not be induced. These results indicate that gaba receptorexpressed in Xenopus oocyte membrane is not type B but for type A. On the basis of the research result above, 24-mer oligonucleotides to be complementary to the high conservativeregion of the cDNAs of neur-gaba receptors were synthesized as probes to hybridize respectively with the RNAs Of brain and testis of adult rat, and of rat testes of the variousages. The dot hybridizations showed that the hybrid signal of rat testis was stronger thanthat of rat brain, meaning that, compared with rat brain, the RNA of rat testis has morehomologous regions with probes, and also implying that the gaba receptor from theRNA Of rat testis may be similar to neural-gaba receptor. This receptor therefore iscalled as gaba_A-like receptor. The dot blot analysis above also showed the testis gaba_Alike receptor. The dot blot analysis above also showed testis gaba_A like receptor mRNAcontent changed with the develOPmental age of rats. There is a very low level of gene expression in the rat testis on day 5 postnatal; there is the highest expression of gaba_A-likereceptor gene in rat testes on day 30to 100 after birth; the expression begins to drop on day200 after birth.

Polyacetylenes are widely present in food and herbs,while these compounds have specific neurotoxicity,and even cause death.The toxicity study of these compounds is still inadequate.Target site of toxicity is not clear,and the mechanisms of toxicity are poorly understood.In order to clarify the neurotoxicity of these compounds,we selected bupleurotoxin isolated from B.longiradiatum as a neurotoxicity-inducing compound.17 toxicity biomarkers were identified using global metabolomics,4 of them were then verified by targeted metabolomics.Bioinformatics analysis using the Ingenuity Pathway Analysis（IPA） software found a strong correlation between the gaba receptor and these metabolites.A validation test using a rat hippocampal neuron cell line was designed to confirm that founding and the results confirmed that BETX inhibited gaba-induced currents（Igaba） in a competitive manner.In summary,our study illustrated the molecular mechanism of the toxicity of polyacetylenes.

Design and Synthesis of Photoaffinity Probe Candidates for the gaba-gated Chloride Channel

In order to characterize binding sites of insecticidal compounds on gaba gated chloride channel, new photoaffinity probe candidates based on 5e-t-butyl-2e-[4-（substituted-propynyl）phenyl]-1,3-dithiane for the noncompetitive blocker （NCB） site of the γ-aminobutyric acid （gaba）-gated chloride channel were designed and synthesized, and their potency as an inhibitor on NCB was measured by 4＇-ethynyl-4-n-[2,3-^3H2]-propylbicycloorthobenzoate （^3H EBOB） assay. The synthesized compounds showed high inhibition activities with half maximum inhibition concentrations （IC50） of lower than 35 nmol/L and were very stable in binding conditions as well photoreacted quickly at 300 nm light. These new compounds are expected to be good photoaffinity labeling probes if radioisotope iodine is incorporated.

Objective Neonatal hypoxic-ischemic encephalopathy（NHIE） is one of the most prevalent causes of death or lifelong disability in children. Thus, it is urgent to seek for new and more effective neuroprotective therapy to minimize the consequences of HIE. Previous studies have shown that oxytocin can improve learning and memory ability after ischemic stroke of adult rats. Here, we ask whether oxytocin has neuroprotective effects on neonatal rats with hypoxic-ischemic brain injury. Methods Brain slices of 350 μmthick from 7-10 days old Sprague-Dawely rats were used. Visualized whole-cell patchclamp recordings were obtained from hippocampal CA1 pyramidal neurons. In-vitro model of hypoxic-ischemic cell injury was used by exposing the brain slices to the oxygen-glucose deprivation（OGD） solution for 10 min or longer. Results In 12 out of 16 neonatal CA1 pyramidal neurons, bath application of oxytocin（0.1 μM） induced an inward current（16.11 ± 1.98 pA） at a holding potential of-70 m V under voltage-clamp recording, which suggest a probable effect of exogenous oxytocin on CA1 pyramidal neurons. Therefore, when switched to current-clamp（I=0）, we observed that oxytocin significantly prolonged the onset time of anoxic depolarization from 13.44 ± 1.84 min to 23.19 ± 2.04 min after the superfusion of OGD solution. Interestingly, both oxytocin receptor antagonist d VOT and gaba receptor antagonist bicuculline blocked this effect. Moreover, oxytocin increased both the amplitude and frequency of spontaneous inhibitory postsynaptic currents（s IPSCs） in a concentration-dependent manner but had no significant influence on spontaneous excitatory postsynaptic currents（s EPSCs） in CA1 pyramidal neurons. In addition, the facilitation effect of oxytocin on IPSCs was blocked by either tetrodotoxin or d VOT. Conclusion Oxytocin exerts a neuroprotective effect by enhancing the inhibitory synaptic transmission through oxytocin receptors in neonatal CA1 pyramidal neurons. Objective:There is substantial evidence supporting the notion that the primary somatosensory cortex (S1) is an important structure involved in the perceptional component of pain.Moreover,neuronal circuit and synaptic plasticity occurring within the S1 after injury has been proposed to contribute substantially to the pathophysiogenesis of chronic pain.However,few reports have been conducted to simultaneously investigate synaptic plasticity in the S1 in response to persistent pain at cellular and network levels.The present study was thus undertaken to evaluate,at the local network level,potential changes in synaptic connection and transmission in the S1 following peripheral persistent nociception in rats.Methods:Periphery subcutaneous injection of bee venom (BV) was used to mimic the state of persistent pain.An initial probe of underlying mechanisms was attempted at the neuronal circuit level by using multi-electrode array recording(MED-64).In electrophysiological experiments, typical neocortical neurons characterized by morphology and action potential were applied to obtain the spontaneous and miniature excitatory and inhibitory post-synaptic currents(EPSC/IPSC).Furthermore,immunohistochemistry and Western blotting were performed to investigate the changes of AMPA and gaba receptors in the state of peripheral persistent pain.Results:By using MED-64 coupled with two-dimensional current source density imaging, we found that persistent pain produced enlargement of synaptic connection size in the S1 slice,reflecting increased spatial plasticity by pain.Meanwhile,compared with saline control,significant increase was also found in the temporal plasticity,namely long-term potentiation.Further slice patch clamp recordings on identified S1 neurons demonstrated that both of the amplitude and frequency of spontaneous and miniature EPSC were greatly increased, while the amplitude of spontaneous and miniature IPSC was markedly decreased after BV injection.Moreover,the half-width of EPSC and IPSC was not changed following persi
