Bioinformatic identification of key genes and pathways that may be involved in the pathogenesis of HBV-associated acute liver failure

In order to explore the molecular mechanisms behind the pathogenesis of acute liver failure(ALF)associated with hepatitis B virus(HBV)infection,the present study aimed to identify potential key genes and pathways involved using samples from patients with HBV-associated *** GSE38941 array dataset was downloaded from the Gene Expression Omnibus database,and differentially expressed genes(DEGs)between 10 liver samples from 10 healthy donors and 17 liver specimens from 4 patients with HBV-associated ALF were analyzed using the Linear Models for Microarray Data *** Ontology and KEGG pathway enrichment analyses of the DEGs were performed,followed by functional annotation of the genes and construction of a proteineprotein interaction(PPI)*** modules were subsequently identified and *** total,3142 DEGs were identified,of which 1755 were upregulated and 1387 were *** extracellular exosome,immune response,and inflammatory response pathways may potentially be used as biomarkers of ALF *** total,17 genes(including CCR5,CXCR4,ALB,C3,VGEFA,and IGF1)were identified as hub genes in the PPI network and may therefore be potential marker genes for HBV-associated ALF.

Bioinformatics prediction of potential mechanisms and biomarkers underlying dilated cardiomyopathy

BACKGROUND Heart failure is a health burden responsible for high morbidity and mortality worldwide, and dilated cardiomyopathy(DCM) is one of the most common causes of heart failure. DCM is a disease of the heart muscle and is characterized by enlargement and dilation of at least one ventricle alongside impaired contractility with left ventricular ejection fraction enrichment analyses. Moreover, a protein-protein interaction network(PPI) was constructed to identify the hub genes and modules. The MicroRNA Database was applied to predict the microRNAs(miRNAs) targeting the hub genes. Additionally, immune cell infiltration in DCM was analyzed using *** In total, 97 DEGs(47 upregulated and 50 downregulated) were identified. GO analysis showed that the DEGs were mainly enriched in “response to growth factor,” “extracellular matrix,” and “extracellular matrix structural constituent.” KEGG pathway analysis indicated that the DEGs were mainly enriched in “protein digestion and absorption” and “interleukin 17(IL-17) signaling pathway.” The PPI network suggested that collagen type Ⅲ alpha 1 chain(COL3A1) and COL1A2 contribute to the pathogenesis of DCM. Purpose:Wnt signaling is closely related to colorectal cancer,high Wnt activity sustains colorectal cancer stem cell(CSC) precursors during the transition from colitis to cancer,leading the development and progression of colorectal ***,it’ s necessary to identify differentially expressed genes(DEGs) of different Wnt activity and to find the potential targets of colorectal CSCs for overcoming colorectal ***:DEGs of different Wnt activity were screened out by GE02 ***,gene ontology(GO) and pathway enrichment analysis were carried out by DAVID ***,the protein-protein interaction network was constructed by STRING *** plug-in Cyto-hubba was used to get the algorithms of PPI network in *** then chose proteins with higher algorithm scores among 12 algorithms and use Funrich to get the common hub genes including FGA,A2 M,IGF1,TGFB2,PDGFB,and ***,by different analysis between colorectal cancer and normal tissues,we got the common genes relative mRNA expression from Oncomine ***:Among three databases GSE32408,GSE17375 and GSE33112,a total of 102 common DEGs,such as TIAM1,A2 M and MMP13 were screened *** functional enrichment analysis showed that the genes were associated with immunity,extracellular items,and binding-related *** pathway analysis was related to Pathways in cancer,Gap junction,and Proteoglycans in ***-protein interaction network construction revealed that common hub genes,including FGA,A2 M,IGF1,TGFB2,PDGFB,and *** their mRNA expression levels were significantly higher in colorectal cancer than in normal colon tissues,which revealed they may play important role in the progression of colorectal ***:In summary,we found 102 DEGs and hub genes that might explain the mechanisms of different Wnt activity as well as the characteristics of CSCs,which may play important role in the progression of colorectal cancer.
