Genome wide association analysis identifies candidate genes for fruit quality and yield in Actinidia eriantha

quality and yield are the primary concerns in kiwifruit breeding,but research on the genetic mechanisms of fruit size,shape,and ascorbic acid(ASA)content is currently very limited,which restricts the development of kiwifruit molecular *** this study,we obtained a total of 8.88 million highly reliable single nucleotide polymorphism(SNP)markers from 140 individuals from the natural hybrid offspring of Actinidia eriantha cv.‘White’using whole genome resequencing technology.A genome-wide association study was conducted on eight key agronomic traits,including single fruit weight,fruit shape,ASA content,and the number of inflorescences per branch.A total of 59 genetic loci containing potential functional genes were located,and candidate genes related to single fruit weight,fruit length,ASA content,number of inflorescences per branch and other traits were identified within the candidate interval,such as AeWUSCHEL,AeCDK1(cell cycle dependent kinase),AeAO1(ascorbic oxidase)and AeCO1(CONSTANS-like 4).After constructing an RNAi vector for AeAO1 and injecting it into the fruit of cv.‘Midao 31’to interfere with the expression of the AeAO1 gene,the results showed that the activity of ascorbic oxidase in the fruit of‘Midao 31’significantly decreased,while the content of ASA significantly *** study provides valuable insights into the genetic basis of variation in *** fruit traits,which may benefit molecular marker-assisted breeding efforts.

Influences of open-central canopy on photosynthetic parameters and fruit quality of apples(Malus×domestica)in the Loess Plateau of China

Although open-central canopy(OCC)is popular in apple(Malus×domestica)orchards in Loess Plateau of China,its relevant photosynthetic mechanisms have not been *** this study,changes in photosynthetically active radiation(PAR),gas exchange,chlorophyll fluorescence quenching and fruit quality in apple trees were measured in OCC and compared with those in the conventional condensed round and large canopy(RLC).Results showed that light intercepted at different orientations was 44%higher by OCC than that by *** improved light environment within OCC remarkably increased leaf maximum net photosynthetic rate(Pnmax)and significantly decreased stomatal *** high light,the ratio of photorespiratory rate to gross photosynthetic rate(Pr/Pg)in OCC was higher than that in ***,reversible component in non-photochemical quenching(r(qE))was increased,while irreversible component(r(qI))was decreased in OCC than in *** a result,the fruit quality in OCC was greatly boosted as evidenced by the significantly increased single fruit weight,fruit flesh firmness and fruit soluble solid contents and the sharply decreased fruit titratable acid *** intercepted by the canopy and the fruit soluble solid contents,leaf Pnmaxor single fruit weight were positively correlated,while PAR or Pnmaxwas negatively correlated with the fruit titratable acid ***,the improved crown light environment and the enhanced leaf photosynthetic performance and photoprotective capacity in OCC led to the boosted fruit quality.

In this study,an integrated transcriptome and metabolome analysis was used to explore the molecular foundation of fruit quality in two parent lines of Citrullus lanatus with distinct flesh characteristics,including ‘14-1’(sweet,red,and soft)andW600'(bitter,light yellow,and firm),as well as the corresponding F,population(bitter,light yellow,and firm).Numerous differentially expressed genes(DEGs)were identified in the fruit samples:3766 DEGs for ‘14-1’vs.W600,2767 for ‘14-1’vs.F_(1),and 1178 for F_(1) vs.‘W600’at the transition stage;and 4221 for ‘14-1’vs.W6002447 for ‘14-1’vs.F,and 446 for F,vs.‘W600’at the maturity *** gene co-expression network analysis(WGCNA)revealed that a gene module including 1111 DEGs was closely associated with flesh taste and color,and another gene module including 1575 DEGs contributed significantly to flesh *** metabolomic results showed that there were 447 differential metabolites(DMs)for ‘14-1’vs.‘W600’fruits,394 for ‘14-1’vs.F,and 298 for F,vs.‘W600’at the maturity *** WGNCA and metabolomic results,several DEGs and DMs were further identified as hub players in fruitqualityformation:6DEGs with 4DMs for flesh sweetness;6DEGs with 13DMs forbitterness;9DEGs with 10 DMs for flesh color;and 9 DEGs with 4 DMs for flesh ***,these observations not only expand our knowledge of the molecular basis of fruit quality in watermelon,but also provide potential targets for future watermelon improvement.

Integrated Use of Organic and Bio-fertilizers to Improve Yield and fruit quality of Olives Grown in Low Fertility Sandy Soil in an Arid Environment

Olive productivity should be improved through stimulating nutrition,particularly under poor fertility ***,the objective of this study was to assess the efficacy of applying organic and bio-fertilizers on the physiological growth,yield and fruit quality of olive trees under newly reclaimed poor-fertility sandy soil in an arid *** a field experiment carried out at El-Qantara,North Sinai,Egypt over two consecutive seasons(2019-2020 and 2020-2021),olive Kalamata trees were evaluated under three organic fertilizer treatments alone or in combination with three bio-fertilizers *** fertilizer was applied as goat manure(16.8 kg/tree/year),or olive pomace(8.5 kg/tree/year)in mid-December of each season *** *** bio-fertilizers were applied as N-fixing bacteria(150 g/tree)was inculated in early March of each season,or amino acid mixture(1.5%)was applied three times,at 70%of full bloom,21 days after full bloom,and a month later in comparison to a non-fertilized trees(control).The cultivar used was Kalamata,a dual-purpose cultivar for oil and table olives whose value increases when processed as table *** results indicated that the goat manure followed by olive pomace significantly enhanced photosynthetic pigments(chlorophyll a,b,and carotenoids),leaf mineral contents(N,P,K,Ca,Mg and Fe),tree canopy volume,number of flowers per inflorescence,number of inflorescences per shoot,initial fruit set,fruit *** fruit quality,fruit length and width,fruit weight,and total fruit yield was increased compared to the non-fertilized ***,The bio-fertilizer N-fixing bacteria followed by the amino acid mixture significantly improved all of the aforementioned ***,it is recommended,both environmentally and economically to utilize organic and bio-fertizers,particularly goat manure combined with N-fixing bacteria,in low-fertility soil to sustain olive production as well as reducing mineral ferti

The Physiological and Molecular Responses of Exogenous Selenium to Selenium Content and fruit quality in Walnut

To study the effect of exogenous selenium on fruit quality in walnut(Juglans regia L.),8-year-old walnut(Qingxiang)was taken as the research *** the fruit expansion stage,300 mg/L of sodium selenate,yeast selenium and sodium selenite solutions were applied on the leaf of walnut,and the selenium levels in leaves,pericarp and kernel were determined at the ripening *** fruit quality,mineral nutrient content,antioxidant enzyme activity,and related genes’expression were *** results showed that the three exogenous selenium increased the selenium levels in leaves,pericarp and kernel of *** also significantly increased fruit and kernel weights,and kernel linoleic acid,but markedly decreased kernel crude fat and saturated fatty *** spraying promoted the absorption of mineral nutrients such as potassium and zinc,but inhibited the absorption of calcium,and had no significant effect on iron and magnesium in the *** exogenous selenium increased the activities of superoxide dismutase(SOD),peroxidase(POD),ascorbate peroxidase(APX),and catalase(CAT)significantly in the *** for sodium selenate treatment significantly reduced malondialdehyde(MDA)content in the kernel,the other two selenium sources treatments had no significant effect on MDA and hydrogen peroxide(H2O2)*** also increased the expression of JrCu/Zn-SOD,Jr2-Cys POD,JrCyt-APX and JrCAT in kernel,to different *** implies that,in the walnut fruit enlargement period,the foliar spraying selenium could increase the selenium content of walnut,affect the mineral nutrient absorption,improve the antioxidant capacity and related genes’expression,and reduce the degree of peroxide,and then improve the quality of ***,yeast selenium showed the comprehensive effect of the best.

Prediction of Apple fruit quality by Soil Nutrient Content and Artificial Neural Network

As the most valuable organ of tomato plants,fruit has attracted considerable attention which most focus on its quality formation during the ripening process.A considerable amount of research has reported that fruit quality is affected by metabolic shifts which are under the coordinated regulation of both structural genes and transcriptional *** recent years,with the development of the next generation sequencing,molecular and genetic analysis methods,lots of genes which are involved in the chlorophyll,carotenoid,cell wall,central and secondary metabolism have been identified and confirmed to regulate pigment contents,fruit softening and other aspects of fruit flavor ***,both research concerning the dissection of fruit quality related metabolic changes,the transcriptional and post-translational regulation of these metabolic pathways are ***,a weighted gene correlation network analysis of representative genes of fruit quality has been carried out and the potential of the combined application of the gene correlation network analysis,fine-mapping strategies and next generation sequencing to identify novel candidate genes determinants of fruit quality is discussed.

Blueberry fruits from 45 commercial cultivars (39 northern highbush and 6 half highbush blueberry) grown in Suwon,Korea were analyzed for fruit size,soluble solids content,titratable acidity,total anthocyanin content,total phenolic content,and antioxidant *** characteristics varied widely among the 45 blueberry *** weight ranged from 0.9 to 3.6 g,soluble solids content from 8.3 to 14.3 °Brix,and titratable acidity from 0.8% to 3.6%.Antioxidant activity ranged from 0.7 to 2.1 mg of quercetin equivalents per gram of fresh berries in different blueberry *** the 45 blueberry cultivars,high amounts of anthocyanins and polyphenols,and high antioxidant activity were observed in 'Elliott','Rubel','Rancocas',and 'Friendship'.

Over-expression of GhDWF4 gene improved tomato fruit quality and accelerated fruit ripening

Brassinosteroids（BRs）, a class of steroidal phytohormones are essential for many biological processes in plant. However, little is known about their roles in fruit development. Tomato is a highly valuable vegetable and has been adopted as the model species for studying fruit growth, development, and ripening. To understand the role of endogenous BRs in the development of tomato fruit, the expression patterns of three homologues of DWF4 gene were investigated and the transgenic tomato plants were generated in which the Gh DWF4 gene from upland cotton（Gossypium hirsutum L.） was ectopically expressed. The contents of main quality components were analyzed in fruits of transgenic tomato line and non-transgenic line（control plant, CP） when the fruit was mature. Sl CYP90B3 that possesses high homology with Gh DWF4 preferentially expressed in mature fruit. Significantly higher contents of soluble sugar, soluble proteins, and vitamin C were obtained in fruit of transgenic tomato lines compared with those in the CP. Furthermore, overexpressing Gh DWF4 promoted fruit growth and ripening. The weight per fruit was increased by about 23% in transgenic lines. In addition, overexpressing Gh DWF4 promoted the germination of transgenic tomato seeds and hypocotyl elongation of seedlings. These results indicated that overexpressing Gh DWF4 gene in tomato could increase the contents of many nutrients in fruit and accelerate fruit ripening. It is suggested that increased endogenous BRs in fruit affect the growth and development of tomato fruit and therefore improved the nutrient quality of tomato.
