The aim of this study was to optimize a tablet formulation of dutasteride that is bioequivalent to a commercially available soft gelatin capsule(Avodart~?). The effect of cyclodextrin on enhancing the aqueous solubility of dutasteride was investigated, after which the formulation was further optimized with solubilizing polymer and surfactant. Among the cyclodextrins tested, the highest solubility was observed when dutasteride was complexed with γ-cyclodextrin. Moreover, the addition of polyvinylpyrrolidone and Gelucire/TPGS further enhanced the solubility of dutasteride. Differential scanning calorimetry(DSC) and powder X-ray diffraction(pX RD) studies demonstrated that dutasteride existed in the amorphous form in the complex. Optimized dutasteride complexes were selected after a pharmacokinetic study in rats, and film-coated tablets were prepared by the direct compression method. In vitro dissolution profiles for the tablets of dutasteride complexes were similar to those of the reference. Moreover, pharmacokinetic parameters including the C max and AUC values after oral administration in beagle dogs were not significantly different from those of the reference with a relative bioavailability of 92.4%. These results suggest the feasibility of developing a tablet formulation of dutasteride using cyclodextrin complex in addition to a solubilizing polymer and surfactant.

We previously determined "Tableting properties" by using a multi-functional single-punch tablet press(GTP-1). We plotted "Compactability" on the x-axis against "Manufacturability"on the y-axis to allow visual evaluation of "Tableting properties". Here, we examined whether this evaluation method can be used in the formulation design of tablets prepared by wet granulation. We used the GTP-1 to measure "Tableting properties" with different amounts of binder, disintegrant, and lubricant, and compared the results with those of tableting on a commercial rotary tableting machine. Tableting failures(capping and binding in particular) occurred when samples that had been evaluated as having poor "Compactability" or"Manufacturability" on the GTP-1 were compressed on the rotary tableting machine. Thus,our evaluation method predicted tableting failure at the commercial scale. The method will prove useful for scaling up production.

Objective:To study the jelly formulation produced by Musa acuminata Colla(AAA Group) peels and evaluate its antioxidant properties which are related to the product ***:The formulations of peel jelly were established under two-level full factorial designs within two blocks and one center *** response optimizer,the amount of sugar and citric acid was obtained;hence,the peel jellies were *** evaluation of antioxidant properties was conducted by using total phenolic content(TPC)assay and 1,1 diphenyl-2-picrylhydrazyl(DPPH) free radical ***:The TPC of peel powder varied from 91.8 to 602.26 mg gallic acid equivalents/100 g dry weight,and 5%-7% peel jellies had phenolic content ranging from 29.38 to 48.31 mg gallic acid equivalents/100 g dry *** results of DPPH test indicated that at 10 mg/mL,the peel powder showed 89% DPPH inhibition,while 7% peel jelly prominently exhibited 84% DPPH *** correlation between DPPH IC50 value and TPC of peel powder as well as peel jelly was quite reasonably high with correlation coefficient ranging from 0.843 7 to ***:TPC can be used as an indicator in assessing the antioxidant activity of fruits and *** present investigation reveals that TPC is mainly responsible for DPPH free radical scavenging capacity.

Accurate real-time truck simulation via semirecursive formulation and Adams-Bashforth-Moulton algorithm

In this paper, a tailored four-step Adams-Bashforth-Moulton (ABM) algorithm is applied to a semirecursive formulation to perform a real-time simulation of a semitrailer truck. In the ABM algorithm, each integration step involves two function evaluations, namely predictor and corrector. This is fundamentally different when compared to the classic fourth-order Runge-Kutta (RK) integrator approach that contains four function evaluations. A semitrailer truck under investigation is modeled in term of a semirecursive method and simulated by using the presented ABM algorithm. The results show that the four-step ABM method can reduce CPU time almost 50% for solving the truck dynamics with very similar accuracy, in comparison to the fourth-order RK method. The presented ABM algorithm could be used in the semirecursive formulation to carry out accurate real-time simulation of medium-large vehicle systems.

Axially variable-length solid element of absolute nodal coordinate formulation

An axially variable-length solid element with eight nodes is proposed by integrating the arbitrary Lagrangian-Eulerian (ALE) formulation and the absolute nodal coordinate formulation (ANCF). In addition to the nodal positions and slopes of eight nodes, two material coordinates in the axial direction are used as the generalized coordinates. As a consequence, the nodes in the ALE-ANCF are not associated with any specific material points and the axial length of the solid element can be varied over time. These two material coordinates give rise to a variable mass matrix and an additional inertial force vector. Computationally efficient formulae of the additional inertial forces and elastic forces, as well as their Jacobians, are also derived. The dynamic equation of a flexible multibody system (FMBS) with variable-length bodies is presented. The maximum and minimum lengths of the boundary elements of an FMBS have to be appropriately defined to ensure accuracy and non-singularity when solving the dynamic equation. Three numerical examples of static and dynamic problems are given to validate the variable-length solid elements of ALE-ANCF and show their capability.

Second generation(2G)high-temperature superconductor(HTS)(RE)Ba2Cu3Ox(REBCO)shows a great potential in building high field magnets beyond 23.5 *** electromagnetic modelling is vital for the design of HTS magnet,however,this always suffers the challenge of huge computation for high field magnets with large number of *** study presents a novel electromagnetic modelling based on T-A formulation for REBCO magnets with thousands of *** equivalent turn method is proposed to reduce the number of turns in calculation,so that the computation cost can be reduced significantly,and meanwhile the key electromagnetic behaviour of HTS magnet can be simulated with enough *** ramping operation of a fully HTS magnet with 12,000 turns are analysed using both the original T-A model with actual turns and improved T-A model with equivalent *** two models show a good agreement on the key electromagnetic behaviours of the magnet:distribution of current density,magnetic fields,screen current induced field and magnetisation loss,so that this improved T-A model using equivalent turns is *** T-A modelling of REBCO magnet is a powerful tool for the electromagnetic analysis of industry-scale high field magnets.

Efficacy of Lubeikangru formulation in mice with hyperplasia of the mammary glands induced by estrogen and progesterone

OBJECTIVE: To evaluate the protective effects of Lubeikangru formula(LF) on hyperplasia of the mammary glands(HMG) induced by estrogen and progesterone in ***: Female mice were divided randomly into five groups: normal, model, tamoxifen(3 mg/kg),Rupixiao(900 mg/kg) and LF(900 mg/kg). All mice except those in the normal group were treated sequentially with estradiol and progesterone to induce HMG. From the tenth day of induction, mice in normal and model groups received distilled water and mice in the other groups were given the corresponding drugs by gavage, once a day, for 30 *** the end of treatment, the mammary glands, ovaries, hypothalamus, and serum was collected for whole-mount and hematoxylin and eosin(HE)staining, enzyme-linked immunosorbent assays(ELISAs), or western ***: Whole-mount and HE staining of mammary glands showed that LF rescued(at least in part) the hyperplasic morphology of the mammary glands, and the number of branch points decreased after LF treatment(P < 0.05). ELISAs revealed that levels of estrogen and progesterone were decreased following LF treatment, whereas levels of gonadotropin-releasing hormone, follicle-stimulating hormone, and luteinizing hormone were increased in serum and tissues. Western blotting confirmed that LF treatment led to a reduction in expression of phosphorylated(p)-Erk, p-p38 and p-c-Jun N-terminal kinase. LF was also confirmed to be safe by acute-toxicity ***: LF can protect the mammary glands of mice from estrogen-and progesterone-induced hyperplasia by adjusting hormone levels and regulating the mitogen-activated protein kinase pathway.
