food allergy and related risk factors in 2540 preschool children: an epidemiological survey in Guangdong Province, southern China

Background:Although the number of studies on allergic diseases in the general population of southern China is increasing,only a few have addressed food allergy(FA)in children in this *** present study aimed to investigate the prevalence,clinical manifestations,spectrum of allergens,and related risk factors of FA in preschool children in Guangdong Province,southern ***:A random cluster-sampling method was used to select 24 kindergartens from 12 cities in Guangdong *** parents or guardians of the children were requested to complete a questionnaire on general information and data regarding FA diagnosis and symptoms in the children and their first-degree ***,the Chi-square test,multivariate regression analysis,and Spearman's rank-order correlation coefficient analysis were performed to identify statistically significant ***:Analysis of 2540 valid questionnaires revealed an FA prevalence rate of 4%.Adverse food reactions were due to the consumption of shrimp(4.4%),crab(3.2%),mango(2.3%),cow's milk and dairy products(1.9%),and eggs(1.4%).Logistic regression analysis indicated that a history of FA and a history of allergic rhinitis in the first-degree relatives were the major factors leading to FA in ***:The incidence of FA in children in Guangdong Province is higher than that commonly *** individual's genetic background is an important risk factor for ***,mitigation of the impact of lifestyle and environmental factors should be carefully considered to reduce the incidence of childhood FA.

Cutaneous exposure to food allergens through a disrupted skin barrier is recognized as an important cause of food allergy,and the cutaneous sensitized mouse model has been established to investigate relevant allergic ***,the role of different genetic backgrounds of mice on immune responses to food allergens upon epicutaneous sensitization is largely *** this study,two strains of mice,i.e.,the BALB/c and C57BL/6 mice,were epicutaneously sensitized with ovalbumin on atopic dermatitis(AD)-like skin lesions,followed by intragastric challenge to induce IgE-mediated food *** outcomes were measured as clinical signs,specific antibodies and cytokines,and immune cell subpopulations,as well as changes in intestinal barrier function and gut *** showed that both strains of mice exhibited typical food-allergic symptoms with a Th2-skewed *** C57BL/6 mice,rather than the BALB/c mice,were fitter for establishing an epicutaneously sensitized model of food allergy since a stronger Th2-biased response and severer disruptions in the intestinal barrier and gut homeostasis were *** study provides knowledge for selecting an appropriate mouse model to study food-allergic responses associated with AD-like skin lesions and highlights the role of genetic variations in the immune mechanism underlying pathogenesis of food allergy.

Preventive effects of Bifidobacterium lactis Probio-M8 on ovalbumin-induced food allergy in mice

food allergy is a significant public health concern *** probiotics have been found to enhance food allergy by regulating immune-microbe interactions in animal models and ***,the effects of Bifidobacterium lactis Probio-M8 on food allergy have not been thoroughly *** present study examined the anti-allergic properties of Probio-M8,particularly in relation to immune response and gut microbiota *** demonstrate that oral administration of Probio-M8 effectively mitigated the allergy symptoms triggered by ovalbumin(OVA)by ameliorating the morphological damage in the jejunum,reducing OVA-specific IgE and histamine levels in the serum,and suppressing Th2 cytokines(interleukin(IL)4 and IL-13)while increasing Th1 cytokines(interferon(IFN)γ)and regulatory T(Treg)cytokines(IL-10 and transforming growth factor(TGF)β1)in the culture supernatants of splenic ***,Probio-M8 effectively altered the diversity and composition of gut microbiota,particularly the relative abundances of Akkermansia_muciniphila in OVA-induced *** to the OVA group,the Probio-M8 group showed a decrease in the relative abundance of Akkermansia_*** conclusion,Probio-M8 demonstrates the potential to alleviate food allergy by regulating the Th1/Th2 response and modulating gut microbiota,thereby offering a novel therapeutic strategy for patients with food allergy.

Oral administration of Lactiplantibacillus plantarum 22A-3 exerts anti-allergic activity against intestinal food allergy mouse models sensitized and challenged with ovalbumin

In recent years,researches on food components with anti-allergic effects have been gathering attention,because of the expectation for the establishment of a safe and effective treatment for food *** studies have reported that Lactiplantibacillus plantarum 22A-3 (LP22A3) inhibited degranulation of mast cells and reduced IgE *** have developed a gastrointestinal allergy system in which mice are sensitized by intraperitoneal and oral administration of OVA and then challenged by oral administration of high doses of *** a result,an increase in the amount of IgE in the blood and a decrease in the temperature of the colon were confirmed,and it was clarified that food allergy was induced by oral administration of high dose of OVA as the *** administration of LP22A3 ameliorated allergic responses significantly by reducing the amount of IgE in the blood and to recovered the decrease of rectal ***,LP22A3 did not affect the intestinal barrier *** LP22A3 significantly suppressed mRNA expression of OX40L and *** results suggested that LP22A3 suppressed Th2 differentiation and IL-4 production via downregulation of OX40L,and consequently suppressed IgE ***22A3 might provide a safe and effective treatment for allergic diseases due to ability modulating intestinal immune system.

food allergy (FA) is one of the global human health problems, affecting about 1 in 12 children and 1 in 10 adults worldwide. Probiotics have alleviating effects on FA, but the mechanisms have not been fully understood. We sought to explore the therapeutic potential of probiotics Bifidobacterium longum CECT7894 (B. longum CECT7894) on anaphylaxis in the ovalbumin (OVA)-induced FA model using an integrated multiomics approaches. The results revealed treatment with B. longum CECT7894 relieved OVA-induced allergic symptoms such as patho-logical changes in small intestine to some extent. Furthermore, probiotics utilization also reduced IgE levels. The transcriptomic data demonstrated that the expressions of genes associated with sphingolipid metabolism were altered in the group received B. longum CECT7894 compared to the sensitized group, including Sgpl1, Trpc1 and Prss8. In addition, the abundances of Sphingobacterium at the genus level, Sphingobacterium sp. 21 and B. longum at the species level, which might related with sphingolipid metabolism, were significantly altered by B. longum CECT7894 when compared to the OVA group. Employed the metabolomics analysis, phytosphingosine, C16 sphinganine, sphinganine and sphingosine were identified as significantly changed metabolites and sphingolipid metabolism pathway was significantly enriched by pathway enrichment analysis. The Spearman's correlation analysis further demonstrated that there were strong correlations between serum biochemical indicators, tran-scripts, gut microbiota and metabolites. Based on the findings described above, from the perspective of genes, gut flora and metabolites changes, it can be concluded that B. longum CECT7894 might suppress OVA-induced FA by regulating the sphingolipid metabolism pathway.

Probiotics' Preventive Effect on Pediatric food allergy: A Meta-analysis of Randomized Controlled Trials

Objective To investigate the preventive effect of probiotics on pediatric food allergy. Methods From MEDLINE bibliographical database, we searched and reviewed all randomized controlled trials on the preventive effects of probiotics on pediatric food allergies up to September 2013 and excluded the studies that do not meet inclusion criteria and extracted the data. Meta-analysis for the results of homogenous studies was performed using RevMan 5.0 and the co-effect was pooled by using fixed-effects model of relative risk(RR) ratios. Results Ten trials published between 2007 and 2012 including 2701 cases were included. Meta-analysis based on included data showed that the preventive effect of prenatal and postnatal probiotic supplementation on food allergies was not significant with the RR=0.88(95% CI: 0.76-1.03). Conclusion Present evidences cannot show in unequivocal terms that prenatal and postnatal probiotic supplementation will prevent food allergic diseases.

food allergy is a common and increasing problem worldwide. The newly-found knowledge might provide novel experimental strategies, especially for laboratory diagnosis. Approximately 20% of the population alters their diet for a perceived adverse reaction to food, but the application of double-blind placebo-controlled oral food challenge, the ＂gold standard＂ for diagnosis of food allergy, shows that questionnaire-based studies overestimate the prevalence of food allergies. The clinical disorders determined by adverse reactions to food can be dassified on the basis of immunologic or nonimmunologic mechanisms and the organ system or systems affected. Diagnosis of food allergy is based on clinical history, skin prick tests, and laboratory tests to detect serum-food specific IgE, elimination diets and challenges. The primary therapy for food allergy is to avoid the responsible food. Antihistamines might partially relieve oral allergy syndrome and IgE-mediated skin symptoms, but they do not block systemic reactions. Systemic corticosteroids are generally effective in treating chronic IgE-mediated disorders. Epinephrine is the mainstay of treatment for anaphylaxis. Experimental therapies for IgE-mediated food allergy have been evaluated, such as humanized IgG anti-IgE antibodies and allergen specific immunotherapy.
