1-DE/MS Analysis of Proteins Related to the Spathe color Variation in an Anthurium andraeanum mutant

[Objectives] This study was conducted to explore the color variation mechanism of Anthurium andraeanum spathe at the protein level. [Methods]The differential proteins of wild type and its white mutant were separated and identified by using one-dimensional gel electrophoresis and mass spectrometry( 1-DE/MS). [Results] Compared with leaves and spadices,the 1-DE patterns of two kinds of spathe proteins were significantly different,and two different bands were detected in wild type spathes and mutant spathes respectively. The four significantly differential bands were selected and analyzed by mass spectrometry,and 138,111,70 and 427 proteins were identified respectively. The results of GO functional annotation analysis showed that the molecular functions of the proteins were mainly catalytic activity and binding,and the main biological processes involved were cellular process and metabolic process. Many proteins involved in the synthesis of anthocyanins and flavonoids,sugar metabolism and some resistance proteins were screened,indicating that the spathe color difference of A. andraeanum‘Pink champion'is not only related to anthocyanin anabolism,but also regulated by various metabolic pathways. [Conclusions]The study provides a new experimental basis for elucidating the molecular mechanism of the regulation of A. andraeanum flower color.

Cultivated Pelargonium spp.,also known as ’house geranium’ belonging to the family of Geraniaceae,is one of the most economical and important bedding and pot plants in North *** color is the most important economic character of Pelargonium,and its color variation mainly arises from anthocyanin *** study of anthocyanidins biosynthesis pathway and regulation mechanism will contribute to plant color improvement and ornamental value *** unravel the complex metabolic networks and gene expression differences between Pelargonium andits anthocyaninloss mutant induced by carbon ions,we collected petals of wild-type and mutant in fully expanded stage and extracted total RNAs *** samples were assessed by Illumina HiSeq2000,a total of 116,114 uni-transcripts were assembled from 45,451,718 and 49,504,188 sequencing raw reads of wild-type and *** 89817 unigenes were obtained with an average length of 751.45 bp and an N50 of 1249 ***,33,630 and 47,933 unigenes had significant hits to known proteins in the Swiss-Protand ***,34,320 unigenes had significant similarity to existing sequences in the GO *** pathway analysis revealed that the differentially expressed unigeneswere significant enrichment in flavonoid biosynthesis *** total,1503 differentially expressed cDNA clones(fold change >2 and FDR mutant,and more genes were down-regulated in the *** significantly suppressed expression of F3 ’5 ’H and ANS might well cause the anthocyanin-loss *** sequence data obtained by the de novo sequencing of Pelargonium and its flower color mutant provided new clues to aid in the understanding of flower mutation induced by carbon ions and establisheda platform for future molecular biological research on Pelargonium.

Heavy ion mutation technology as a unique and efficient mutagen has been widely used in germplasm innovation and plant breeding. flower color as the important qualitative character and economic indicator for ornamental plants become the primary target for ornamental breeding. Here we report one flower color mutant obtained using carbon ion irradiation and attempt to elucidate underlying mutation mechanism. The petal color of the mutant changed from orange-red to light pink,and colors of peduncle,pistil and stamen also displayed significant differences with wild type. Anatomical structure observations showed that the salmon pigments fully filled with the epidermal cells of wild type,while only a small number of pale pink pigments in mutant. HPLC results indicated that the main pigments determined the flower color of wild type were pelargonidin,cyanidin and delphinidin. On the contrary,the unique pigment detected in mutant was cyanidin. The core reason of depigmentation in mutant was the absence of pelargonidin and delphinidin. Quantitative real-time PCR analysis suggested that the significantly down-regulated transcriptional level of early genes CHS,CHI and suppressed expression of gene ANS blocked the biosynthesis of anthocyanin,and that was the crucial reason for the deficiency of anthocyanin which lead to the depigmentation of mutant. These results explained the mechanism of coloration change in mutant induced by carbon ions and provided the research foundation for the follow-up design and color improving in geranium.
