This study investigated the effect of salicylic acid(SA) and sodium nitroprusside(SNP; NO donor) on nickel(Ni) toxicity in germinating finger millet seedlings. Fourteen-day-old finger millet plants were subjected to 0.5 mmol L^(-1) Ni overload and treated with 0.2 mmol L^(-1)salicylic acid and 0.2 mmol L^(-1)sodium nitroprusside to lessen the toxic effect of *** Ni overload led to high accumulation in the roots of growing plants compared to shoots, causing oxidative stress. It further reduced root and shoot length, dry mass,total chlorophyll, and mineral content. Exogenous addition of either 0.2 mmol L^(-1)SA or0.2 mmol L^(-1)SNP reduced the toxic effect of Ni, and supplementation with both SA and SNP significantly reduced the toxic effect of Ni and increased root and shoot length,chlorophyll content, dry mass, and mineral concentration in Ni-treated plants. The results show that oxidative stress can be triggered in finger millet plants by Ni stress by induction of lipoxygenase activity, increase in levels of proline, O_2^(·-) radical, MDA, and H_2O_2, and reduction in the activity of antioxidant enzymes such as CAT, SOD, and APX in shoots and roots. Exogenous application of SA or SNP, specifically the combination of SA + SNP,protects finger millet plants from oxidative stress observed under Ni treatment.

Flexible triboelectric nanogenerators (TENGs)-based pressure sensors are very essential for the wide-range applications, comprising wearable healthcare systems, intuitive human-device interfaces, electronic-skin (e-skin), and artificial intelligence. Most of conventional fabrication methods used to produce high-performance TENGs involve plasma treatment, photolithography, printing, and electro-deposition. However, these fabrication techniques are expensive, multi-step, time-consuming and not suitable for mass production, which are the main barriers for efficient and cost-effective commercialization of TENGs. Here, we established a highly reliable scheme for the fabrication of a novel eco-friendly, low cost, and TENG-based pressure sensor (TEPS) designed for usage in self-powered-human gesture detection (SP-HGD) likewise wearable healthcare applications. The sensors with microstructured electrodes performed well with high sensitivity (7.697 kPa^-1), a lower limit of detection (~ 1 Pa), faster response time ( 4,000 compression-releasing cycles. The proposed method is suitable for the adaptable fabrication of TEPS at an extremely low cost with possible applications in self-powered systems, especially e-skin and healthcare applications.

ZNE191 is a new zine finger gene which has a 1107 bp open reading frame (ORF) and encodes a 368amino acid protein including a putative DNA-binding domain of four Cys2His2, zine finder motifs at its C-terminal region. The ZNF191 eDNA is located in the chromosome 18q12.1 region where it is known that a variety of hereditary diseases arc related to. Probably, this protein has potential function of stimulating the gene transcription.In order to examine the function and structure of ZNF191 protein, the OHK of ZNf191 and its zinc finger region ZNF191 (243-368) genes were inserted into PTSA-18 expression vector by PCR amplification, then the constructed genes were expressed in the PTSA-18/B121 (DE3) system. The two proteins were purified by DEAE-52, CM-23 and Heparin-Sepharose 4B columns. The pooled proteins showed a single band as assayed by Coomasie Brillian Blue Staining of an SDS/polyacryamide gel.

Prolonged activation of nuclear factor(NF)-кB signaling significantly contributes to the development of colorectal cancer(CRC).New therapeutic opportunities are emerging from targeting this distorted cell signaling ***,we discovered the critical role of RING finger 138(RNF138)in CRC tumorigenesis through regulating the NF-кB signaling,which is independent of its Ubiquitin-E3 ligase activity involved in DNA damage ***138^(−/−) mice were hyper-susceptible to the switch from colitis to aggressive malignancy,which coincided with sustained aberrant NF-кB signaling in the colonic ***,RNF138 suppresses the activation of NF-кB signaling pathway through preventing the translocation of NIK and IKK-Beta Binding Protein(NIBP)to the cytoplasm,which requires the ubiquitin interaction motif(UIM)*** importantly,we uncovered a significant correlation between poor prognosis and the downregulation of RNF138 associated with reinforced NF-кB signaling in clinical settings,raising the possibility of RNF138 dysregulation as an indicator for the therapeutic intervention targeting NF-кB *** the xenograft models built upon either RNF138-dificient CRC cells or the cells derived from the RNF138-dysregulated CRC patients,we demonstrated that the inhibition of NF-кB signaling effectively hampered tumor ***,our work defined the pathogenic role of aberrant NF-кB signaling due to RNF138 downregulation in the cascade events from the colitis switch to colonic neoplastic transformation and progression,and also highlights the possibility of targeting the NF-кB signaling in treating specific subtypes of CRC indicated by RNF138-ablation.

Currently, most efficient algorithms for force-closure analysis and dynamic force distribution utilize linear programming, but friction models are nonlinear. Substituting polyhedral cones for circular cones of Coulomb friction realizes the linearization of the frictional point contact constraint. So far, however, there is no approach to soft finger contact. This paper present such an approach. Then the foregoing algorithms can be extended to grasping with soft finger contact. Herein an optimal force distribution algorithm for soft multifingered grasps is developed with an illustrative example.

Nearly half of the world population suffers from micronutrient malnutrition,particularly Zn *** is important to understand genetic variation for uptake and translocation behaviors of Zn in relevant crop species to increase Zn concentration in edible *** the present study,genetic variation in grain Zn concentration of 319 finger millet genotypes was *** genetic variation was found among the genotypes,with concentrations ranging from 10 to 86 μg g^(-1)*** and translocation studies with Zn/^(65) Zn application in 12 selected low-Zn genotypes showed wide variation in root uptake and shoot translocation,with genotypes GEC331 and GEC164 showing greater uptake and *** GEC164 and GEC543 showed increased grain Zn *** GEC331 and GEC164 also showed improved yield under Zn *** variation in grain Zn concentration among finger millet genotypes found in this study offers opportunities to improve Zn nutrition through breeding.

Background: The mitogen-activated extracellular signal-regulated kinase 1/2(MEK1/2) inhibitor trametinib has shown promising therapeutic effects on melanoma, but its efficacy on colorectal cancer(CRC) is limited. Synthetic lethality arises with a combination of two or more separate gene mutations that causes cell death, whereas individual mutations keep cells alive. This study aimed to identify the genes responsible for resistance to trametinib in CRC cells,using a synthetic lethal short hairpin RNA(shRNA) screening ***: We infected HT29 cells with a pooled lentiviral shRNA library and applied next-generation sequencing to identify shRNAs with reduced abundance after 8-day treatment of 20 nmol/L trametinib. HCT116 and HT29 cells were used in validation studies. Stable ring finger protein 183(RNF183)-overexpressing cell lines were generated by pcDNA4-myc/his-RNF183 transfection. Stable RNF 183-knockdown cell lines were generated by infection of lentiviruses that express RNF183 shRNA, and small interference RNA(siRNA) was used to knock down RNF183 *** real-time PCR was used to determine the mRNA expression. Western blotting, immunohistochemical analysis, and enzyme-linked immunosorbent assay(ELISA) were used to evaluate the protein abundance. MTT assay,colony formation assay, and subcutaneous xenograft tumor growth model were used to evaluate cell ***: In the primary screening, we found that the abundance of RNF183 shRNA was markedly reduced after treatment with trametinib. Trametinib induced the expression of RNF183, which conferred resistance to drug-induced cell growth repression and apoptotic and non-apoptotic cell deaths. Moreover, interleukin-8(IL-8) was a downstream gene of RNF183 and was required for the function of RNF183 in facilitating cell growth.
