Determination of double-and single-stranded DNA breaks in bovine sperm is predictive of their fertilizing capacity

Background:The analysis of chromatin integrity has become an important determinant of sperm quality.In frozenthawed bovine sperm,neither the sequence of post-thaw injury events nor the dynamics of different types of sperm DNA breaks are well understood.The aim of the present work was to describe such sperm degradation aftermath focusing on DNA damage dynamics,and to assess if this parameter can predict pregnancy rates in cattle.Results:A total of 75 cryopreserved ejaculates from 25 Holstein bulls were evaluated at two post-thawing periods(0-2 h and 2-4 h),analyzing global and double-stranded DNA damage through alkaline and neutral Comet assays,chromatin deprotamination and decondensation,sperm motility,viability,acrosomal status,and intracellular levels of total ROS,superoxides and calcium.Insemination of 59,605 females was conducted using sperm from the same bulls,thus obtaining the non-return to estrus rates after 90 d(NRR).Results showed an increased rate of double-stranded breaks in the first period(0-2 h:1.29±1.01%/h vs.2-4 h:0.13±1.37%/h;P0.05).Finally,whereas no correlations between bull fertility and any dynamic parameter were found,there were correlations between the NRR and the basal percentage of highly-damaged sperm assessed with the alkaline Comet(Rs=-0.563,P=0.003),between NRR and basal progressive motility(Rs=0.511,P=0.009),and between NRR and sperm with high ROS at 4 h post-thaw(Rs=0.564,P=0.003).Conclusion:The statistically significant correlations found between intracellular ROS,sperm viability,sperm motility,DNA damage and chromatin deprotamination suggested a sequen

Oleic acid reduces steroidogenesis by changing the lipid type stored in lipid droplets of ovarian granulosa cells

Background:Oleic acid is an abundant free fatty acid present in livestock that are in a negative energy-balance state,and it may have detrimental effects on female reproduction and fertility.Oleic acid induces lipid accumulation in bovine granulosa cells,which leads to a foam cell-like morphology and reduced steroidogenesis.However,why oleic acid increases lipid accumulation but decreases steroidogenesis remains unclear.This study focused on oleic acid’s effects on lipid type and steroidogenesis.Results:Oleic acid increased the lipid accumulation in a concentration-dependent manner and mainly increased the triglyceride level and decreased the cholesterol ester level.Oleic acid also led to a decline in estradiol and progesterone production in porcine granulosa cells in vitro.In addition,oleic acid up-regulated the expression of CD36 and diacylglycerol acyltransferase 2,but down-regulated the expression of 3-hydroxy-3-methylglutarylcoenzyme A reductase,scavenger receptor class B member 1 and acetyl-Coenzyme A acetyltransferase 2,as well as steroidogenesis-related genes,including cytochrome P450 family 11 subfamily A member 1,cytochrome P450family 19 subfamily A member 1 and 3 as well as steroidogenic acute regulatory protein at the mRNA and protein levels.An oleic acid-rich diet also enhanced the triglyceride levels and reduced the cholesterol levels in ovarian tissues of female mice,which resulted in lower estradiol levels than in control-fed mice.Compared with the control,decreases in estrus days and the numbers of antral follicles and corpora lutea,as well as an increase in the numbers of the atretic follicles,were found in the oleic acid-fed female mice.Conclusions:Oleic acid changed the lipid type stored in lipid droplets of ovarian granulosa cells,and led to a decrease in steroidogenesis.These results improve our understanding of fertility decline in livestock that are in a negative energy-balance state.

Follicle-stimulating hormone(FSH), as the main indicator of ovarian function, plays an important role in the clinical activities of gynecologic endocrinology. Although anti-Miillerian hormone and antral follicle count are also the indictors evaluating ovarian function, many clinicians are still relentless in their decision to impose the death penalty of ovaries when high FSH levels(especially more than 40 IU/L) are observed.We recently encountered four patients whose FSH levels were inconsistent with actual fertility because all patients had successfully conceived after treatment. In our study, we found the culprit(macro-FSH) of false-positive FSH levels by applying the polyethylene glycol protein precipitation technique. The biological functions of macro-FSH were further evaluated by using Gene Ontology and Kyoto Encyclopedia of Genes and Genomes pathway analysis. This study suggests that ovarian reserve function should be comprehensively assessed in clinic, and the causes of serum test indicators inconsistent with clinic should be identified.

MicroRNAs(miRNAs)are mediators of the aging process.The purpose of this work was to analyze the miRNA expression profiles of spermatozoa from men of different ages with normal fertility.Twenty-seven donors were divided into three groups by age(Group A,n=8,age:20–30 years;Group B,n=10,age:31–40 years;and Group C,n=9,age:41–55 years)for high-throughput sequencing analysis.Samples from 65 individuals(22,22,and 21 in Groups A,B,and C,respectively)were used for validation by quantitative real-time polymerase chain reaction(qRT-PCR).A total of 2160 miRNAs were detected:1223 were known,937 were newly discovered and unnamed,of which 191 were expressed in all donors.A total of 7,5,and 17 differentially expressed microRNAs(DEMs)were found in Group A vs B,Group B vs C,and Group A vs C comparisons,respectively.Twenty-two miRNAs were statistically correlated with age.Twelve miRNAs were identified as age-associated miRNAs,including hsa-miR-127-3p,mmu-miR-5100_L+2R-1,efu-miR-9226_L-2_1ss22GA,cgr-miR-1260_L+1,hsa-miR-652-3p_R+1,pal-miR-9993a-3p_L+2R-1,hsa-miR-7977_1ss6AG,hsa-miR-106b-3p_R-1,hsa-miR-186-5p,PC-3p-59611_111,hsa-miR-93-3p_R+1,and aeca-mir-8986ap5_1ss1GA.There were 9165 target genes of age-associated miRNAs.Gene Ontology(GO)analysis of the target genes identified revealed enrichment of protein binding,membrane,cell cycle,and so on.The Kyoto Encyclopedia of Genes and Genomes(KEGG)enrichment analysis of age-related miRNAs for target genes revealed 139 enriched pathways,such as signaling pathways regulating stem cell pluripotency,metabolic pathways,and the Hippo signaling pathway.This suggests that miRNAs play a key role in male fertility changes with increasing age and provides new evidence for the study of the mechanism of age-related male fertility decline.

Genetic structure and domestication footprints of the tusk,coat color,and ear morphology in East Chinese pigs

The domestication and artificial selection of wild boars have led to dramatic morphological and behavioral changes,especially in East Chinese(ECN)pigs.Here,we provide insights into the population structure and current genetic diversity of representative ECN pig breeds.We identify a 500-kb region containing six tooth development-relevant genes with almost completely different haplotypes between ECN pigs and Chinese wild boars or European domestic pigs.Notably,the c.195A>G missense mutation in exon 2 of AMBN may cause alterations in its protein structure associated with tusk degradation in ECN pigs.In addition,ESR1 may play an important role in the reproductive performance of ECN pigs.A major haplotype of the large lop ear-related MSRB3 gene and eight alleles in the deafness-related GRM7 gene may affect ear morphology and hearing in ECN pigs.Interestingly,we find that the two-end black(TEB)coat color in Jinhua pigs is most likely caused by EDNRB with genetic mechanisms different from other Chinese TEB pigs.This study identifies key loci that may be artificially selected in Chinese native pigs related to the tusk,coat color,and ear morphology,thus providing new insights into the genetic mechanisms of domesticated pigs.

Fixation of hybrid sterility genes and favorable alleles of key yield-related genes with dominance contribute to the high yield of the Yongyou series of intersubspecific hybrid rice

In rice,the Yongyou series of Xian-Geng intersubspecific hybrids have excellent production performance,as shown by their extremely high yield.However,the mechanisms underlying the success of these rice hybrids are unclear.In this study,three F2 populations are generated from three Yongyou hybrids to determine the genetic basis of the extremely high yield of intersubspecific hybrids.Genome constitution analysis reveals that the female and male parental lines belong to the Geng and Xian subspecies,respectively,although introgression of 20%of the Xian ancestry and 14%of the Geng ancestry are observed.Twenty-five percent of the hybrid genomes carries homozygous Xian or Geng fragments,which harbors hybrid sterility genes such as Sd,Sc,f5,and q S12 and favorable alleles of key yield-related genes,including NAL1,Ghd7,and Ghd8.None of the parents carries the S5+killer of the S5 killer-protector system.Compatible allele combinations of hybrid sterility genes ensure the fertility of these intersubspecific hybrids and overcome the bottleneck in applying intersubspecific hybrids.Additive effects of favorable alleles of yield-related genes fixed in both parents enhances midparent values.Many QTLs for yield and its key component spikelets per panicle shows dominance and the net positive dominant effects lead to heterosis.These factors result in an extremely high yield of the hybrids.These findings will aid in the development of new intersubspecific rice hybrids with diverse genetic backgrounds.

Population aging has emerged as one of the most pressing global demographic issues.Rapid population aging is being caused by a sharp drop in fertility rates over time,as well as a significant increase in life expectancy in the past decades.The World Health Organization(WHO)estimates that the proportion of those aged 60 years and older in the world’s population will almost double from approximately 12%in 2015 to 22%in 2050,with an absolute increase of 900 million to a total of 2 billion older adults(1).At the same time,the burden of global disease has changed greatly and the main diseases affecting human health have switched from acute and chronic infectious diseases to chronic non-communicable diseases(NCDs)(2).Such changing demographics and disease spectrum will profoundly affect all aspects of human society.

With a high rate of infertility, it is important to understand the context of fertility and family planning in China, to inform the necessity of supportive care. A literature review was undertaken to explore the societal constructs informing perspectives of childbearing, family planning and infertility, alongside Chinese considerations of fertility treatments, including assisted reproductive technologies and fertility counselling. In China, childbearing attitudes and behaviours are shaped by tensions between traditional cultural values of the filial piety originating from Confucianism, the history of strict family planning policy, the recent termination of one-child policy and the socioeconomic circumstance. For infertile Chinese individuals, the inability to meet these childbearing expectations gives rise to significant pressure and consequent psychological distress, particularly depressive symptoms. Demographic factors such as gender, education, income and geographical location have been found to influence prevalence and degree of depression in infertile Chinese men and women. These difficulties are compounded by barriers of cultural acceptance, legislative restrictions and availability of resources for alternative options such as adoption and surrogacy. It is important that these fertility sociocultural factors are taken into consideration when assisting Chinese patients to access and utilise fertility treatment services.

Homeostatic relevance of vitamin D in maintaining male fertility in human: Down-regulation of oxidative stress and up-regulation of anti-oxidative defense and steroidal hormones

Objective:To evaluate correlation between the levels of vitamin D and male infertility as well as to determine the efficacy of vitamin D in improving the male fertility by up-regulating the levels of testosterone and spermatogenesis.Methods: In the present study, 130 male patients (aged 25-70 years) having fertility defects were screened and 145 healthy individuals were taken as control. All human subjects were screened for 4-hydroxynonenal, isoprostane-F2α, 8-hydroxy-2′-deoxyguanosine, vitamin D, luteinizing hormone, follicle stimulating hormone, testosterones, malondialdehyde, superoxide dismutase, catalase, glutathione peroxidase, and nitric oxide.Results: The screening analysis revealed that the levels of luteinizing hormone, follicle stimulating hormone, and testosterone were lower in male infertile subjects compared to healthy subjects. Similarly, the levels of vitamin D [(17.17 ± 2.30) ng/mL] and calcium[(6.29 ± 0.31) mg/dL] were significantly lower in infertile groups compared to the normal healthy groups. Moreover, the study revealed that the levels of superoxide dismutase, catalase, and glutathione peroxidase were significantly higher in healthy subjects compared to the infertile subjects.Conclusions:Vitamin D exhibits strong relevance to male fertility by maintaining the levels of sex hormones (luteinizing hormone, follicle stimulating hormone, and testosterone), up-regulating the antioxidant defense (superoxide dismutase, catalase, and glutathione peroxidase), and down-regulating the oxidative stress (malondialdehyde, nitric oxide, and inducible nitric oxide synthase species).

Seasonal effect on physiological, reproductive and fertility profiles in breeding mithun bulls

Objective:To analyse the seasonal effect on physiological parameters, reproductive profiles andin vitro fertility in breeding mithun bulls.Methods: A total of ten adult mithun bulls age of 5 to 6 years old with good body condition (score 5-6) were selected from ICAR-NRC on Mithun, Jharnapani, Nagaland, India. The seasons were categorised into winter, spring, summer and autumn seasons based on the meteorological data and sunshine hours. The physiological parameters, reproductive profiles andin vitro fertility parameters were assessed during different seasons in mithun under the semi-intensive system of management.Results:The statistical analysis revealed that these experimental parameters were differed significantly (Pfertility for artificial breeding programme in mithun under the semi-intensive system.
