The Chinese population is entering a new stage of development,marked by the Fertility intentions of women of childbearing age continuing to decline and concerns over their consistently low Fertility rates.The number of newborns has been decreasing annually,with 2022 figures falling below 10 million for the first time since the founding of the People’s Republic of China,resulting in negative population growth(1).Currently,China’s population over the age of 60 years is nearing 300 million and is projected to surpass 20%of the total population during the 14th Five-Year Plan period.

Background MicroRNAs(miRNAs)are small,single-stranded,non-coding RNA molecules of 22–24 nucleotides that regulate gene expression.In the last decade,miRNAs have been described in sperm of several mammals,including cattle.It is known that miRNAs can act as key gene regulators of early embryogenesis in mice and humans;however,little is known about the content,expression,and function of sperm-borne miRNAs in early bovine embryo.In this study,total sperm RNA was isolated from 29 cryopreserved sperm samples(each coming from a separate bull)using a RNeasy kit and treatment with DNase I.RNA concentration and purity were determined through an Epoch spectrophotometer and an Agilent Bioanalyzer.The expression of 10 candidate miRNAs in bovine sperm(bta-miR-10a,bta-miR-10b,bta-miR-138,bta-miR-146b,bta-miR-19b,bta-miR-26a,bta-miR-34a,bta-miR-449a,bta-miR-495 and btamiR-7),previously identified in testis and/or epididymis,was evaluated with RT-qPCR.The cel-miR-39-3p was used as a spike-in exogenous control.Nonparametric Mann–Whitney tests were run to evaluate which miRNAs were differentially expressed between bulls with high Fertility[HF;non-return rates(NRR)ranging from 39.5 to 43.5]and those with subFertility(SF;NRR ranging from 33.3 to 39.3).Several sperm functionality parameters(e.g.,viability,membrane stability or oxygen consumption,among others)were measured by multiplexing flow cytometry and oxygen sensing technologies.Results RNA concentration and purity(260/280 nm ratio)(mean±SD)from the 29 samples were 99.3±84.6 ng/μL and 1.97±0.72,respectively.Bioanalyzer results confirmed the lack of RNA from somatic cells.In terms of the presence or absence of miRNAs,and after applying the Livak method,8 out of 10 miRNAs(bta-miR-10b,-138,-146b,-19b,-26a,-449a,-495,-7)were consistently detected in bovine sperm,whereas the other two(bta-miR-10a,and-34a)were absent.Interestingly,the relative expression of one miRNA(bta-miR-138)in sperm was significantly lower in the SF than in the HF group(P=0.038).In addition to being associated to

OBJECTIVE:To evidently assess the applicability of regulate menstrual cycle(MC)characteristics in Traditional Chinese Medicine(TCM)as an indicator for Fertility.METHODS:A community-based prospective cohort study was conducted in China.Between January 2010 and December 2012,women who were willing to conceive within 2 years were enrolled in the study.Other than the MC length pattern,a well-adopted MC characteristic,menstrual blood color and clots were specifically concerned for women enrolled.All participants were followed up in 2 years by trained nurses.Pregnancy rate,fecundability odds ratio(FOR)and risk of miscarriage were assessed as Fertility outcomes.RESULTS:A total of 2109 women were effectively included in this cohort for analysis.Results show that women with irregular MC length were less likely to achieve conception(FOR_(irregular)=0.59;95%CI=0.45-0.77,PFertility outcomes(FOR=1.02,95%CI=0.83-1.25,P=0.88;OR=1.2695%CI=0.77-2.07,P=0.35).CONCLUSIONS:MC characteristics can be an effective and simple indicator for women’s Fertility.Increasing the knowledge of MC characteristics for women in reproductive ages would bring great benefits to their preconception health conditions.

Background:Identification of relevant in vivo biomarkers for Fertility remains a challenge for the livestock industry.Concentrations of the small peptide hormone oxytocin(OXT),involved in male reproductive function and present in the seminal plasma(SP)of several species could be a robust one.This study characterized concentrations of SPOXT in ejaculates from boars used in artificial insemination(AI)programs aiming to evaluate its relationship with sperm quality variables and in vivo Fertility of their liquid-stored AI-semen.Seminal OXT concentrations(ng/mL)were measured in 169 ejaculates from 61 boars of the Duroc,Pietrain,Landrace and Large White breeds using a direct competitive immunoassay test based on AlphaLISA®technology.Ejaculate(ejaculate volume,sperm concentration,total sperm count)and sperm parameters(motility,viability,intracellular generation of reactive oxygen species,plasma membrane fluidity)were assessed at 0 h and 72 h in AI-semen samples stored at 17℃.In vivo Fertility included only 18 Large White and Landrace boars whose AI-semen was used to inseminated>100 sows and evaluated both farrowing rate and litter size of 3,167 sows.Results:The results showed that SP-OXT differed between boars and between ejaculates within boar(P<0.05)but not between breeds(Duroc,Pietrain,Landrace and Large White).Ejaculates with higher SP-OXT concentration/mL(hierarchically grouped;P<0.001)had larger volume and came from younger boars(P<0.05).Ejaculates of boars showing positive farrowing rate deviation exhibited higher(P<0.05)SP-OXT concentration/mL than those with negative farrowing rate deviation.Conclusion:The SP concentrations of OXT are boar,ejaculate and age dependent,and positively related with ejaculate volume and farrowing rates of liquid-stored semen AI-doses.

Background:Genetic selection in cattle has been directed to increase milk production.This,coupled to the fact that the vast majority of bovine artificial inseminations(AI)are performed using cryopreserved sperm,have led to a reduction of Fertility rates over the years.Thus,seeking sensitive and specific sperm biomarkers able to predict Fertility rates is of vital importance to improve cattle reproductive efficiency.In humans,sperm chromatin condensation evaluated through chromomycin A3(CMA3)has recently been purported to be a powerful biomarker for sperm functional status and male inFertility.The objectives of the present study were:a)to set up a flow cytometry method for simultaneously evaluating chromatin condensation and sperm viability,and b)to test whether this parameter could be used as a predictor of in vivo Fertility in bulls.The study included pools of three independent cryopreserved ejaculates per bull from 25 Holstein males.Reproductive outcomes of each sire were determined by non-return rates,which were used to classify bulls into two groups(highly fertile and subfertile).Results:Chromatin condensation status of bovine sperm was evaluated through the combination of CMA3 and Yo-Pro-1 staining and flow cytometry.Sperm quality parameters(morphology,viability,total and progressive motility)were also assessed.Pearson correlation coefficients and ROC curves were calculated to assess their capacity to predict in vivo Fertility.Sperm morphology,viability and total motility presented an area under the ROC curve(AUC)of 0.54,0.64 and 0.68,respectively(P>0.05),and thus were not able to discriminate between fertile and subfertile individuals.Alternatively,while the percentage of progressively motile sperm showed a significant predictive value,with an AUC of 0.73(P=0.05),CMA3/Yo-Pro-1 staining even depicted superior results for the prediction of in vivo Fertility in bulls.Specifically,the percentage of viable sperm with poor chromatin condensation showed better accuracy and precision to predict in vivo Fertility,wi

Dear Editor,Utilization of heterosis is fundamental to modern agricultural improvement and contributes significantly to food security and agricultural development(Ouyang et al.,2022).However,heterosis of F,hybrids cannot be stably inherited.Apomixis,or asexual reproduction through seeds,offers the potential for the fixation of heterosis in F,hybrids and the formation of clonal offspring,which could have considerable implications for plant breeding and agriculture(Schmidt,2020).But naturally occurring apomixis has never been found in major crops.Recently,synthetic apomixis has been successfully generated by inducing incomplete meiotic development combined with parthenogenesis development to produce cloned rice(Oryza sativa)seeds(Khanday et al.,2019;Wang et al.,2019;Vernet et al.,2022;Wei et al.,2023).

Gardaneh Salavat porphyry copper system is located in the northwest of Iran. It comprises a quartz diorite to diorite porphyritic intrusion that created extensive alteration haloes in the host rocks. Despite all technical considerations, the exploration activities did not yield to ore discovery. As the matter of fact, all of the geological,geophysical, and geochemical characteristics of a porphyry Cu-Au deposit exist but there is no economic mineralization. Detailed examinations suggest that the only reasonable explanation can be related to the inFertility or low metal endowment of the porphyritic intrusion. The reasons include(1) anomalous ranges of Cu, Au, and Mo rather than economic ore grades,(2) weakly development of potassic alteration,(3) probably sulfur saturation of magma that occurred before intrusion emplacement in the host rocks, and(4) probably low metal endowment in the regional scale and the lack of known porphyry copper deposits in close neighborhoods. It is tried to test the proposed hypothesis with the introduced Fertility indicators based on major and trace element geochemistry. However,current Fertility indicators could not prove or reject the proposed hypothesis for Gardaneh Salavat porphyritic intrusion because of the high-K calc-alkaline to shoshonitic nature of the intrusive rocks together with the unavailability of relatively fresh/unaltered samples. This case study is notable to examine because(1) it shows that characterization of fertile from barren intrusions seems to be as important as vectoring towards concealed porphyry copper deposits, especially while dealing with deep exploration targets that are associated with high financial risks, and(2) it highlights the inefficiencies and uncertainties in introduced magma Fertility indicators for porphyry copper intrusions. As a result, it is of high importance to conduct an overarching investigation on other possible Fertility indicators as a fundamental need for the future of the exploration industry.

The two bay scallop subspecies,Argopecten irradians irradians(NN)and A.i.concentricus(SS),are fast growing and major cultured bivalves in China.However,their relatively small sizes and decreasing production traits caused by long-term inbreeding have been major concerns to the industry in the last two decades.Hybridization between the two bay scallop subspecies may provide a new approach to breed a new variety with superior production traits for the industry.For this end,in this study,we hybridized the two bay scallop subspecies in order to obtain a new strain that incorporates the genes of both subspecies.No significant difference was found in fertilization rate,hatching rate and metamorphosis rate between the purebred and crossbred cohorts(NN♀×SS♂,denoted as NS;SS♀×NN♂,denoted as SN).Both mating strategy(intra-vs.inter-population crosses)and egg origin had significant effects on growth and survival at the larval stage.Heterosis was observed in the crossbred and was more pronounced in older stages.Genetic diversity of the reciprocal hybrids,especially that of SN,was increased compared with the purebred cohorts.Almost all hybrids were completely fertile and able to reproduce by selffertilization or by backcrossing with either parent.Apparently,male sterile individuals whose gonads were fully occupied by the ovary part at mature stage were found in the hybrids for the first time.The hybrids,especially SN,may provide precious germplasm resources for the production of ternary hybrids with the Peruvian scallop,A.purpuratus.

Small noncoding RNAs (sn RNAs) such as short interfering RNAs (si RNAs), micro RNAs (mi RNAs), and PIWI-interacting RNAs (pi RNAs) play important roles in diverse cellular processes. These small regulatory RNA molecules exert key regulatory functions by forming complexes with different Argonaute proteins, including the Argonaute(AGO) and PIWI subfamilies (Peters and Meister, 2007).AGO subfamily proteins are ubiquitously expressed and interact with mi RNAs or si RNAs, while PIWI subfamily proteins are expressed mainly in germline cells and interact specifically with pi RNAs (Bartel, 2018).

Rapeseed(Brassica napus,AACC)was formed by hybridization between progenitor species Brassica rapa(AA)and Brassica oleracea(CC).As a result of a limited number of hybridization events between specific progenitor genotypes and strong breeding selection for oil quality traits,rapeseed has limited genetic diversity.The production of resynthesized B.napus lines via interspecific hybridization of the diploid progenitor species B.rapa and B.oleracea is one possible way to increase genetic variation in rapeseed.However,most resynthesized lines produced so far have been reported to be meiotically unstable and infertile,in contrast to established B.napus cultivars.This hinders both maintenance and use of this germplasm in breeding programs.We characterized a large set of 140 resynthesized lines produced by crosses between B.rapa and B.oleracea,as well as between B.rapa and wild C genome species(B.incana,B.hilarionis,B.montana,B.Bourgeaui,B.villosa and B.cretica)for purity(homozygosity),Fertility,and genome stability.Self-pollinated seed set,seeds per ten pods as well as percentage pollen viability were used to estimate Fertility.SNP genotyping was performed using the Illumina Infinium Brassica 60K array for 116 genotypes,with at least three individuals per line.Most of the material which had been advanced through multiple generations was no longer pure,with heterozygosity detected corresponding to unknown parental contributions via outcrossing.Fertility and genome stability were both genotypedependent.Most lines had high numbers of copy number variants(CNVs),indicative of meiotic instability,and high numbers of CNVs were significantly associated with reduced Fertility.Eight putatively stable resynthesized B.napus lines were observed.Further investigation of these lines may reveal the mechanisms underlying this effect.Our results suggest that selection of stable resynthesized lines for breeding purposes is possible.
