BACKGROUND fanlian huazhuo formula(FLHZF)has the functions of invigorating spleen and resolving phlegm,clearing heat and purging *** has been identified to have therapeutic effects on type 2 diabetes mellitus(T2DM)in clinical ***-alcoholic fatty liver disease(NAFLD)is frequently diagnosed in patients with ***,the therapeutic potential of FLHZF on NAFLD and the underlying mechanisms need further *** To elucidate the effects of FLHZF on NAFLD and explore the underlying hepatoprotective mechanisms in vivo and in *** HepG2 cells were treated with free fatty acid for 24 hours to induce lipid accumulation cell ***,experiments were conducted with the different concentrations of freeze-dried powder of FLHZF for 24 hours.C57BL/6 mice were fed a high-fat diet for 8-week to establish a mouse model of NAFLD,and then treated with the different concentrations of FLHZF for 10 *** FLHZF had therapeutic potential against lipid accumulation and abnormal changes in biochemical indicators in vivo and in *** experiments verified that FLHZF alleviated abnormal lipid metabolism might by reducing oxidative stress,regulating the AMPKα/SREBP-1C signaling pathway,activating autophagy,and inhibiting hepatocyte *** FLHZF alleviates abnormal lipid metabolism in NAFLD models by regulating reactive oxygen species,autophagy,apoptosis,and lipid synthesis signaling pathways,indicating its potential for clinical application in NAFLD.

The prevalence of metabolic dysfunction-associated steatotic liver disease(MASLD)has increased significantly in recent decades and is projected to increase further due to the rising obesity *** patients are at higher risk of developing advanced liver diseases“cirrhosis and hepatocellular carcinoma”as well as liver-or cardiovascular-related *** lipid-lowering therapies failed to reduce the risk of mortality in these ***,there is an urgent need for pharmacotherapies that can control and even reverse this *** huazhuo formula(FLHZF)is a combination herbal preparation,and its various individual constituents regulate hepatic lipid metabolism,adipose tissue inflammation,and gut ***,these useful effects,limited information is available on its benefits in diet-induced *** this article,we discuss the research findings recently published about the therapeutic effects of FLHZF in suppressing MASLD development and underlying *** a series of in vitro and in vivo experiments,the authors demonstrated for the first time that FLHZF suppresses MASLD in male mice possibly by inhibiting hepatic de novo lipogenesis pathways and reducing hepatocyte *** study paves the way for future investigations aimed at investigating FLHZF’s role in inhibiting lipogenesis particularly using radioactively-labeled glucose and acetate,and governing hepatocyte mitochondrial function,gut microbiome profile,and its effects in other models of MASLD,and female mice.
