Laminarin oligosaccharides(LOSs)with a specific degree of polymerization prepared through the laminarin degradation via laminarinase present more significant nutritional functions and application *** intestinal bacteria are promising potential producers of novel carbohydrate-active enzymes with unique ***,a novel glycoside hydrolase family 128(GH128)laminarinase OUC-BsLam26 from the intestinal bacterium Bacteroides ***7301 was heterologously expressed and *** recombinant OUC-BsLam26 with a molecular mass of 49.86 kDa exhibits highest activity(6.60 U/mg)at 45℃ and pH 6.0,which shows noticeable temperature and pH *** purified OUC-BsLam26 could degrade laminarin via an endo-type mode with the generation of laminaripentaose,laminaritetraose,laminaritriose,and laminaribiose,among them,laminaritetraose is the principal product,which accounts for 45.25% of the total products,which is significantly different from the reported GH128 *** minimum recognition substrate of OUC-BsLam26 is ***,OUC-BsLam26 also could catalyze the transglycosylation process with the production of some novel ***,the intestinal bacterium Bacteroides ***7301 contains laminarinase with unique product composition and OUC-BsLam26 is a hopeful bio-catalyst with the potential to produce laminaritetraose and some novel glycosides.

High expression of INF2 Predicts Poor Prognosis and Promotes Hepatocellular Carcinoma Progression

The vaccinia viral vector containing T7 promoter was used to construct the expression plasmids carrying HCV structural genes of C, E1 and E2/NS1. These genes were transiently expressed in mammalian cells in the presence of the T7 RNA polymerase which was provided by the recombinant vaccinia virus vTT7. expression of mature core protein, envelope protein E1 and E2 was detected by Western blot using HCV patient sera as the primary antibodies. It was found that the sera from different HCV patients reacted differently with the expressed products, so did the sera collected at different times from the same patient, from whom the HCV structural genes were isolated. Among six mammalian cell lines, Vero and HeLa were the most suitable for the expression of C, E1 and E2. The recombinant vaccinia viruses have been constructed to constantly produce the C, E1 and E2 proteins for further research.

expression of the Tribolium castaneum (Coleoptera: Tenebrionidae) hsp83 gene and its relation to oogenesis during ovarian maturation

Heat shock proteins （HSP） can protect organisms and cells from thermal damage. In this study, we cloned the full length cDNA encoding the HSP83 protein （the homologue of HSP90） of Tribolium castaneum（red flour beetle）. The isolated cDNA contains the full coding sequence, a partial 5＇ untranslated region of 55 bp and the complete 3＇ untranslated region. We found the hsp83 gene is located on chromosome 5 of the T. castaneum genome. The predicted HSP83 protein sequence has a high similarity （on average 86.77%） with that of other insect species. The expression of the hsp83 gene in the whole body and in the ovary could be induced with heat stress （40℃ for 1 h） in newly hatched （within 3 h post emergence） and mature （10 days post emergence） beetles. Under normal conditions, the hsp83 expression in the ovary is about 3-fold higher than in the whole body at both stages. No significant difference in hsp83 expression was observed between the two ovarian developmental stages regardless if the beetles were treated with heat shock or not. The expression of the HSP83 protein in the whole body could also be induced with heat stress in newly hatched and mature beetles. However, in the ovary, HSP83 was only expressed in the follicle cells of mature beetles and not in newly hatched beetles, regardless if the beetles were treated with heat shock or not. Furthermore, the females were not able to produce mature oocytes after knock-down of the hsp83 expression by injecting dsRNA. These results suggest that the HSP83 protein is involved in protection against heat stress and could be involved in oogenesis during ovarian maturation of T. castaneum.

ZNE191 is a new zine finger gene which has a 1107 bp open reading frame (ORF) and encodes a 368amino acid protein including a putative DNA-binding domain of four Cys2His2, zine finder motifs at its C-terminal region. The ZNF191 eDNA is located in the chromosome 18q12.1 region where it is known that a variety of hereditary diseases arc related to. Probably, this protein has potential function of stimulating the gene transcription.In order to examine the function and structure of ZNF191 protein, the OHK of ZNf191 and its zinc finger region ZNF191 (243-368) genes were inserted into PTSA-18 expression vector by PCR amplification, then the constructed genes were expressed in the PTSA-18/B121 (DE3) system. The two proteins were purified by DEAE-52, CM-23 and Heparin-Sepharose 4B columns. The pooled proteins showed a single band as assayed by Coomasie Brillian Blue Staining of an SDS/polyacryamide gel.

expression profiling of the BpIAA gene family and the determination of IAA levels in Betula platyphylla tetraploids

Betula platyphylla Sukaczev tetraploids have significantly larger leaf, fruit and stoma (gigantic phenotype) than diploids of the same species;however, the mechanism underlying this difference remains unclear. Tetraploid B. platyphylla transcriptome data have indicated that the expression of genes related to indole-3-acetic acid (IAA) biosynthesis and signal transduction was altered after genome duplication. IAA exerts pleiotropic effects on growth and development by inducing the expression of Aux/IAA. We identified 20 Aux/IAA genes (BpIAA1– BpIAA20) in B. platyphylla distributed across 10 chromosomes. Multiple alignment and motif analyses revealed that nine BpIAA proteins shared all four conserved domains. Phylogenetic analysis indicated that Aux/IAA families were divided into four subfamilies and that there were two pairs of BpIAA sister genes. The BpIAAs were differentially expressed in diploids and tetraploids. Moreover, the expression levels of the nine BpIAA genes were specifically up-regulated in tetraploids from June to September compared with May (except August 5th) in tetraploids, while they were down-regulated in diploids. IAA levels were more than twofold higher in tetraploids than diploids during the vegetative season. These results indicate that genome duplication of B. platyphylla caused the up-regulated of genes involved in IAA synthesis, and the increased concentration of IAA may induce the constitutive expression of 20 BpIAA genes. Therefore, the significant changes in the expression patterns of the BpIAAs contributed to the gigantic phenotype of tetraploids to some extent. Our research sheds light on the phenotypic variations observed in B. platyphylla tetraploids.
