Variations in Reproductive Hormones during estrus cycle in Local Beef Cattle from Jilin of China

[ Objective] To provide a theoretical basis for improving reproductive efficiency of domestic beef cattle in Jilin of China. [ Method] China Red Steppe cattle, Yanbian yellow cattle and Simmental cattle in Jilin Province were used for detection. The levels of progesterone （P4）, estradiol （ E2 ）, follicle-stimulating hormone （FSH） and luteinizing hormone （LH） in serum and urine during estrus cycle were determined. [ Result] The levels of P4 and E2 were significantly different between pregnant cattle and non-pregnant cattle on Day 21 post estrus （ P 〈 0.01 ）. The level of LH on estrus day was significantly different from that on the other days of estrus cycle （ P 〈 0.05）. [ Conclusion] It is feasible to detect P4 level for diagnosis of estrus and eady pregnancy in beef cattle.

Relationship between estrus cycles and behavioral durations of captive female alpine musk deer

Owing to poor breeding success in captive alpine musk deer, an understanding of the behavioral patterns of muskdeer in captivity is important. This study was conducted from June 2004 to January 2005 at the Xinglongshan MuskDeer Farm, which is located within Xinglongshan National Nature Reserve, Gansu Province, China. Focal samplingand continuous recording were used to observe the behaviors of 51 female alpine musk deer (Moschus sifanicus),42 of which had completed a single estrus cycle and nine of which had completed two or more estrus cycles. Allanimals were adults that had been born and raised in captivity. The durations of 12 behaviors, including environmentalsniffing, moving and feeding, were recorded during the non-breeding seasons and behavioral patterns werecompared. The behavioral patterns of females that had completed a single estrus cycle and females that hadcompleted multiple estrus cycles were compared to assess potential behavioral differences. The results showed thatfemales who had only one complete estrus cycle demonstrated more resting behavior, but less feeding and locomotorbehavior than females who had completed multiple estrus cycles. Furthermore, single estrus cycle femalesdemonstrated tail-rubbing during the breeding season. The results may yield useful information that can be used indeveloping better musk deer farming practices.

Combined effects of Gymnema sylvestre and Pergularia daemia on letrozole-induced polycystic ovarian syndrome in rats

Aim:To investigate the role of hypothalamus nociceptin/orphanin FQ（OFQ）andits endogenous receptor,the opioid receptor-likel receptor（ORL1 receptor）in theestrus cycle of female ***:Radioimmunoassay was used to detect theeffect of the intracerebroventricular（icy）administration of OFQ and/or the ORL1receptor antagonist[Nphe1]Nociceptin（1-13）NH2,that is,NC13 on luteinizinghormone（LH）levels of estrogen- and progesterone（EBP）-primed,ovariectomized（OVX）rats（EBP-primed OVX rats）.RT-PCR,Western blotting,and immunohis-tochemistry techniques were adopted to observe the changes of OFQ and theORL1 receptor in the pre-optic area（POA）and the medial basal hypothalamus（MBH）of the estrus cycle of female ***:Pre-ovulatory LH surges in EBP-primed,OVX rats were significantly reduced by icv administration of 20 and 200nmol OFQ（Pestrus waslowered markedly compared to diestrus and estrus（Pestrus ***,the number of OFQ-immunoreac-tive neurons in the medial POA,ventromedial hypothalamus,and the arcuatenucleus on pro-estrus was significantly decreased compared to diestrus and es-tins（Pestrus sug-gests that it might play a negative modulatory role in the estrus cycle.

Antifertility Activity of Hydroalcoholic Extract of Ocimum Basilicum Linn. Leaves on Female Wistar Rats

Objective To determine the effect of hydroalcoholic extract of Ocimum basilicum Linn. leaves on ovulation, implantation and maintenance of pregnancy in adult female cyclic Wistar rats. Methods Ocimum basilicum leaves extract （364 mg/kg and 624 mg/kg） was screened for its antiovulatory, antiimplantation and abortifacient activities. Vaginal smear of each rat was monitored daily during the 15-day treatment of the first experiment. Estimation of cholesterol in ovarian tissue and its histology was carried out to confirm its antiovulatory activity. In the second and third experiments, animals received treatment from 1st to 7th and 8th to 16th day of pregnancy, respectively. The number of implantations and litter size were determined through laparotomy on l Oth and 20th day of pregnancy, respectively in two tests. The treated groups were compared with pain control. Results A significant increase in duration of estrus cycle and diestrus phase was observed （P〈O.O01）. A significant decrease in ovarian weight and notable increase in ovarian tissue cholesterol level was also observed （P〈O. 001）. Weight of uterus in treated group was found to be decreased. Histological report showed presence of large corpora lutea in ovarian parenchyma. Neither antiimplantation nor abortifacient effect was observed in rats treated with both the doses of extract. Conclusion Hydroalcoholic extract of Ocimum basilicum interferes normal ovulation by disrupting the estrus cycle and prolonging diestrus phase thus, has a potential of being developed into a female contraceptive.

Functional Transcriptomic Analysis of Female Rat Hippocampus via RNA Seq in Response to Estrogen

In the hippocampus, estradiol modulates a wide range of neural functions related to neurotransmission, synaptic plasticity, and neurogenesis. Previous studies showed that synaptic plasticity of female hippocampus cyclically fluctuates across the estrus cycle, but the key genes responsible for mediating these changes are still unknown. Here, we explored the hippocampal transcriptome via RNA-seq using proestrus（PE） and estrus（ES） stage female rats. At standard fold change selection criteria, 253 genes were differentially expressed in PE vs ES group, and 15 transcripts were identified as the most significant genes during the estrus cycle. The gene ontology（GO） analysis revealed the distinct features of biological adhesion, immune system process, developmental process, biological regulation, response to stimulus, signaling, cellular component organization or biogenesis, metabolic process, and cellular process including neurological system process were enriched and down regulated during PE vs ES stage. Next, we treated ovariectomized（OVX） rats with E2 and analyzed the transcriptome. Total 1903 transcripts were identified as DEGs in OVX and OVX plus E2 treated groups. The significantly upregulated genes were successfully expressed and identified in OVX+E2 group as the plasma E2 level restored to PE stage. This further confirmed the validity of our RNA-seq result in PE vs ES group. All alterations identified were confirmed by realtime PCR. Altogether, the results suggest that E2 upregulates the expression of genes involved in regulating neurite outgrowth, nerve regeneration process, promote synaptic plasticity, neuroprotection;maintaining normal cognitive process, enhancing spatial and reference memory, neural resilience against aging, oligodendrocyte maturation and myelination of the CNS;mediating retinoid signaling, involved in AKT/ERK signaling pathways and neurological disorders.

Objective Sex hormones in the brain play an important role in regulating stress *** gonads produce the most sex hormones that are lipophilic steroids which can pass through the blood brain barrier,we aimed to study whether and how the alterations in the peripheral sex hormone levels affect the stress-related molecules in the *** Female SD rats were sacrificed in different stages of estrous cycle（n = 12 per stage for proestrus,estrus and diestrus） *** addition,15 rats were ovariectomized（OVX） and compared with 15sham-operated female ***,10 rats were castrated（CAS） and compared with 10 sham-operated male *** the operated rats got recovery for 3 *** blood and hypothalami were *** and hypothalamic levels of estradiol（E2）,testosterone（T） were measured by *** corticosterone（CORT） levels were measured by *** of estrogen receptor-α（ER-α）-,ER-β-,androgen receptor-,aromatase-,mineralcorticoid receptor-,glucocorticoid receptor-,corticotropin releasing hormone-,vasopressin- and oxytocin-mRNA in the hypothalamus were determined by real-time quantitative ***（1） In normally cycling female rats,although the blood levels of E2 and T were significant higher among proestrus than estrus（P ＜ 0.0001,P ＜0.0001） and diestrus（P ＜ 0.0001,P ＜ 0.0001） stages,there were no significant changes in stress-related molecules in the hypothalamus.（2） The OVX group showed significantly lower plasma levels of E2 and T（P ＜ 0.0001 and P ＜ 0.01）but a trend of increased E2 in the hypothalamus（P = 0.055）.None of the stress-related molecules showed significant changes in the hypothalamus（P ≥ 0.142） except for a significantly increased ERα-mRNA（P=0.038）.（3） The CAS group showed significantly lower T levels both in the plasma（P＜ 0.01） and in the hypothalamus（P ＜ 0.05）,but plasma CORT increased（P ＜ 0.05）.None of the stress-related molecules showed significant changes in the hypothalamus（P ≥0.121） exc
