Analgesic-like activity of perillyl acetate:In vivo and in silico studies

Objective:To evaluate the antinociceptive activity of perillyl acetate in mice and in silico simulations.Methods:The vehicle,perillyl acetate(100,150 and/or 200 mg/kg,i.p.),diazepam(2 mg/kg,i.p.)or morphine(6 mg/kg,i.p.)was administered to mice,respectively.Rotarod test,acetic acidinduced abdominal writhing,formalin-induced nociception,hot plate test,and tail-flick test were performed.Opioid receptorsinvolvement in perillyl acetate antinociceptive effect was also investigated.Results:Perillyl acetate did not affect the motor coordination of mice.However,it reduced the number of acetic acid-induced abdominal twitches and licking times in the formalin test.There was an increase of latency time in the tail-flick test of 30 and 60minutes.Pretreatment with naloxone reversed the antinociceptive effect of perillyl acetate(200 mg/kg).In silico analysis demonstrated that perillyl acetate could bind toμ-opioid receptors.Conclusions:Perillyl acetate has antinociceptive effect at the spinal level in animal nociception models,without affecting the locomotor integrity and possibly throughμ-opioid receptors.In silico studies have suggested that perillyl acetate can act as aμ-opioid receptor agonist.

Chemical composition and phytotoxic activity of the volatile oil of invasive Xanthium italicum Moretti from Xinjiang,China

Aerial parts of Xanthium italicum in an air tight container greatly inhibited root elongation of radish, implying that this invasive plant could release biologically active volatile organic compounds （VOCs） into the environment to affect other plants＇ growth. This phenomenon was further studied by evaluating the phytotoxic effects of X. italicum essential oil against two dicot plants, amaranth （Amaranthus mangostanus L.） and lettuce （Lectuca sativa L.）, and two monocot plants, wheat （Triticum aestivum Linn） and ryegrass （Lolium multiforum）, and analyzing the chemical composition of the oil. Among the 4 test species, amaranth was the most sensitive plant, 0.5μl/mL essential oil application resulted in a 50% reduction on root elongation, and 2.5 μl/mL essential oil almost completely inhibited its seedling growth. Wheat was the least sensitive species, whose root growth was reduced to 36% of control by 5 μl/mL essential oil. The essential oil exerted moderate inhibitory effect on both lettuce and ryegrass. Compared to a commercial herbicide-Harness, X. italicum oil exhibited stronger phytotoxicity on amaranth, lettuce and wheat, but weaker activity on ryegrass. The chemical composition of the essential oil isolated by hydrodistillation from the aerial parts of X. italicum Moretti was analyzed by GC/MS. Thirty two compounds were identified, representing 94.89% of total oil, which was found to be rich in monoterpene hydrocarbons （60.71%）. The main constituents of the oil were limonene （51.61%）, germacrene B （6.98%）, 6-cadinol （5.94%）, β-pinene （5.23%）, α-caryophyllene （5.1%） and bornyl acetate （3.15%）. Bioassay revealed the dominant constituent-limonene, was unlikely the responsible phytotoxic compound due to its low biological activity; rather, there might be other oil constituent（s） that either act alone, or work together, and possibly assisted by synergistic effect, to display the phytotoxic activity. Our results suggested that X. italicum might produce allelopathic VOCs to facilitate its invasion success. This is

Ethnopharmacological relevance: Elsholtzia densa Benth. is widely used as a folk medicine in China. Elsholtzia species’chemical constituents can be divided into essential oil, flavonoids, lignanoids, coumarins, steroids and fatty acids. essential oils are known to possess antimicrobial activity against a wide spectrum of bacteria and fungi. Aim of the study: In the present study the Chemical composition, in vitro antibacterial, antifungal and antiviral activities of the essential oil of Elsholtzia densa Benth. were evaluated. Material and methods: The chemical composition of the essential oil of Elsholtzia densa Benth. has been analyzed by Gas chromatography-mass spectrometry （GC-MS）. Furthermore, the essential oil was tested against 3 bacteria and one fungi at different concentrations using minimum inhibitory concentration （MIC）. Then use MDCK cells culture method on the essential oil against influenza A and B viruses. By observing cytopathic effect （CPE） and calculating the selectivity index （SI） to evaluate for its anti-influenza activity in vitro. Results: Twenty-eight compounds, constituting about 77.36% of the total oil, were identified. The most abundant oil components are Heptenol, Naphthalene, Azulene, cyclohexane, Patchoulene, alpha-Ylangene, Azulenemethanol and Caryophyllene. Results showed that the essential oil exhibited antibacterial and antifungal activity. Although the essential oil exhibited activity against influenza A virus it did not show any antiviral activity against influenza B virus. Conclusion: The essential oil of the aerial parts of Elsholtzia densa Benth .displays antibacterial, antifungal and antiviral activities in a certain degree in vitro and is worthy of further study.

So the author prepared the essential oil of orange peel. It is helpful to the health of mankind.

Although there are normal cognitive changes that take place as a person becomes older,ageing in humans is generally associated with a deterioration of cognitive performance,in particular of learning and memory.There are a number of herbal medicines that are reported to improve brain function and intelligence.In the present study,the ameliorating effects of an essential oil extracted from Acori graminei rhizoma on learning and memory in aged,dysmnesia rats and mice were determined using the step-down passive avoidance test and Y maze.Oral administration of the essential oil(0.02,0.04 and 0.08 g kg) to rats for 30 days and to mice for 15 days improved the latency and number of errors in aged,dysmnesia rats and mice.The cerebral neurotransmitters in aged rats given the essential oil(0.02,0.04,0.08 g kg) for 30 days were also investigated,and increased levels of norepinephrine,dopamine and serotonin,and decreased levels of acetylcholinesterase activity were found.The results suggest that the essential oil improves cognitive function in aged animals possibly by increasing norepinephrine,dopamine and serotonin relative levels,and by decreasing the activity of acetylcholinesterase in the cerebra.

The purpose of this study was to find alternatives to chemical fungicides to control vegetables and fruits postharvest diseases.The essential oil(EO) of Origanum vulgar was isolated by hydro distillation and analyzed with gas chromatograph coupled to a quadrupole mass spectrometer(GC-MS) and gas chromatography flame ionization detector(GC-FID).The major compounds of O.vulgar EO were methyl eugenol(16.46 %),carvacrol(14.97 %),thymol(9.82%).The EO and its major constituents were tested in vitro and in vivo antifungal activities against Botrytis cinerea.Overall,the compounds showed better antifungal activity in volatile activity assay than poisoned food technique.In volatile activity assay,the EC50 values of the EO,thymol,carvacrol,methyl eugenol inhibited the mycelia growth were 16.44,0.93,1.61,67.57 mg/L,respectively.The EC50 values ranged from 17.56 to 112.43 mg/L for poisoned food technique.Thymol and carvacrol had very good antifungal activities against spore germination in volatile activity assay and poisoned food technique.Results from the in vivo antifungal tests demonstrated that when the concentration of thymol and carvacrol were 62.5 mg/L,and more than 90 % decay area of tomatoes could be inhibited by the two compounds.Thus,O.vulgare EO,thymol and carvacrol can be considered as potential source for the development of natural fungicides to control vegetables and fruits postharvest diseases.

Gastric motor dysfunction induced by psychological stress results in many symptoms of functional dyspepsia (FD).There are a number of herbal medicines that are reported to improve gastrointestinal motor.However,the mechanisms of considerable herbal medicines are not explicit.In the present study,the effects of an essential oil(EO) extracted from Atractylodes lancea on delayed gastric emptying,gastrointestinal hormone and hypothalamic corticotrophin-releasing factor (CRF) abnormalities induced by restraint stress in rats were investigated and the mechanism of the EO is also explored.Oral administration of EO for 7 days did not affect normal gastric emptying,but accelerated delayed gastric emptying induced by restraint stress in rats.The EO significantly increased the levels of motilin(MTL) and gastrin(GAS) and decreased the levels of somatostatin(SS) and CRF.The EO did not modify gastric emptying in vagotomized rats that underwent restraint stress, but antagonized delayed gastric emptying induced by intracisternal injection of CRF.These results suggest that the regulative effects of the EO on delayed gastric emptying are preformed mainly via inhibition of the release of central CRF and activation of vagal pathway,which are also involved in the release of gastrointestinal hormones such as MTL,GAS and SS.

The antifungal activity against Botryosphaeria dothidea from postharvest kiwifruits were screened from 33 essential oil monomers and the action mode of active compound were further evaluated.The results showed antifungal activity of 33 tested essential oil monomers were greatly affected by their chemical structures and carvacrol was the strongest one with EC of 12.58 μL and EC of22.08 μL/L.Furthermore,carvacrol exhibited significantly inhibitory effects on the kiwifruit pathogen in vivo and in vitro.Carvacrol could evidently alter the hyphal morphology of B.dothidea and severely damaged cell membrane with the reduction of its lipid components.Cell membrane permeability were increased and intracellular homeostasis including ion and biomacromolecules were destroyed by carvacrol.Carvacrol significantly inhibited the mitochondrial activity and the respiration rates were decreased,which induced the cell death of B.dothidea.The results from the research suggest that carvacrol had great potential for controlling postharvest rot soft of kiwifruit.

Natural deep eutectic solvents(NADESs) as functional liquid media have received increasing attention in recent years, which are directly composed of natural products. [1-3] A novel method based on natural deep eutectic solvent coupled with microwave-assisted hydrodistillation was used for the extraction of Cuminum cyminum L. essential oil. Several experimental parameters, such as NADESs type, NADESs/water ratio, NADESs/power ratio, sample pretreatment stage of microwave power, set-up temperature, duration, and hydrodistillation duration were all studied. Under the optimal conditions, the NADESs composed by choline chloride and L-lactic acid(molar ratio with 1:3) obtained the highest EO yield of 2.2%. Also, the EOs were analyzed by using gas chromatography-mass spectrometry(GC-MS), with a total of 108 compounds being identified. In this case, microwave helps to reduce the time for extraction and NADESs can effectively improve the EO yield. Especially, compared with binary DES system, NADESs obviously brought about more identified compositions. Hence, the NADESs-based approach has the potential of being extended to the analysis of low content of volatile compounds from other plant resources.
