Antimicrobial resistance characteristics and phylogenetic relationships of pleuromutilin-resistant enterococcus isolates from different environmental samples along a laying hen production chain

Antimicrobial resistance in the laying hen production industry has become a serious public health problem.The antimicrobial resistance and phylogenetic relationships of the common conditional pathogen enterococcus along the laying hen production chain have not been systematically clarified.105 enterococcus isolates were obtained from 115 environmental samples (air,dust,feces,fies,sewage,and soil) collected along the laying hen production chain (breeding chicken,chick,young chicken,and commercial laying hen).These enterococcus isolates exhibited resistance to some clinically relevant antibiotics,such as tetracycline (92.4%),streptomycin (92.4%),and erythromycin (91.4%),and all strains had multidrug resistance phenotypes.Whole genome sequencing characterized 29 acquired antibiotic resistance genes (ARGs) that conferred resistance to 11 classes of antibiotics in 51pleuromutilin-resistant enterococcus isolates,and lsa(E),which mediates resistance to pleuromutilins,always co-occurred with lnu(B).Alignments with the Mobile Genetic Elements database identified four transposons (Tn554,Tn558,Tn6261,and Tn6674) with several ARGs(erm(A),ant(9)-la,fex(A),and optrA) that mediated resistance to many clinically important antibiotics.Moreover,we identified two new transposons that carried ARGs in the Tn554 family designated as Tn7508 and Tn7492.A complementary approach based on conventional multi-locus sequence typing and whole genome single nucleotide polymorphism analysis showed that phylogenetically related pleuromutilin-resistant enterococcus isolates were widely distributed in various environments on different production farms.Our results indicate that environmental contamination by antimicrobial-resistant enterococcus requires greater attention,and they highlight the risk of pleuromutilin-resistant enterococcus and ARGs disseminating along the laying hen production chain,thereby warranting effective disinfection.

Background:This study aimed to investigate the effects of oral administration of enterococcus faecium NCIMB10415(E.faecium)on intestinal development,immunological parameters and gut microbiota of neonatal piglets challenged with enterotoxigenic Escherichia coli K88(ETEC).A total of 961-day-old sow-reared piglets were randomly assigned to 2 groups,with 48 piglets in each group.The piglets were from 16 litters(6 piglets each litter),and 3 piglets each litter were allocated to the E.faecium-supplemented(PRO)group,while the other 3 piglets were allocated to the control(CON)group.After colostrum intake,piglets in the PRO group were orally administrated with 3×10~9 CFU E.faecium per day for a period of one week.On day 8,one piglet per litter from each group was challenged(CON+ETEC,PRO+ETEC)or not(CON-ETEC,PRO-ETEC)with ETEC in a 2×2 factorial arrangement of treatments.On day 10(2 days after challenge),blood and tissue samples were obtained from piglets.Results:Before ETEC challenge,there were no significant differences for the average daily gain(ADG)and fecal score between the two groups of piglets.After ETEC challenge,the challenged piglets had greater fecal score compared to the non-challenged piglets,whereas E.faecium administration was able to decrease the fecal score.Piglets challenged with ETEC had shorter villous height,deeper crypt depth,and reduced number of goblet cells in the jejunum and decreased m RNA abundance of claudin-1 in the ileum,whereas increased the percentage of lymphocytes,concentrations of IL-1βin the plasma and TNF-αin the ileal mucosa,as well as increased the m RNA abundances of innate immunity-related genes in the ileum tissue.These deleterious effects caused by ETEC were partly alleviated by feeding E.faecium.In addition,piglets in PRO-ETEC group had decreased the percentage of CD8^+T cells of the peripheral blood when compared to those in CON-ETEC group.Moreover,E.faecium administration increased Verrucomicrobia at phylum level and decreased Bilophila at genus level.Conclusions:These resul

Exogenous endophthalmitis caused by enterococcus casseliflavus:A case report

BACKGROUND Endophthalmitis caused by Enterococci is rare,and cases involving vancomycinresistant enterococci are even rarer.We report the first case of enterococcus casseliflavus endophthalmitis associated with injury caused by a pig.We also review reported cases of exogenous endophthalmitis caused by enterococcus casseliflavus and discuss the clinical management and prognosis of this disease.CASE SUMMARY A 43-year-old man with no previous visual problems complained of endophthalmitis in his left eye following injury caused by a pig.Visual acuity was light perception and B-ultrasonography revealed vitreous opacities with retinal detachment.He was treated with intravitreal vancomycin and ceftazidime after refusing vitrectomy.However,the vitreous opacities and retinal detachment deteriorated and he underwent vitrectomy 5 d post-injury.Intraoperatively,advanced rhegmatogenous and tractional retinal detachment with proliferative vitreoretinopathy were observed.On postoperative day 5,vitreous cultures grew Gram-positive cocci identified as enterococcus casseliflavus.A 2-wk course of intravenous ampicillin and dexamethasone was commenced.On postoperative day 14,visual acuity improved to hand movement.At 6 mo post-injury,visual acuity improved to 20/667,but optic atrophy was present.CONCLUSION Systemic administration of linezolid in the treatment of enterococcus casseliflavus endophthalmitis can improve visual acuity.However,intravitreal amikacin should be considered despite concerns of toxicity when oral linezolid fails to prompt improvement.

Antimicrobial activity of some essential oils against oral multidrugresistant enterococcus faecalis in both planktonic and biofilm state

Objective:To evaluate some essential oils in treatment of intractable oral infections,principally caused by hiofilm of multidrug-resistant enterococcus faecalit(E.faecalis),such as persistent endodontic infections in which their treatment exhibits a real challenge for dentists.Methods:Ten chemically analyzed essential oils by gas chromatography-mass spectrometry were evaluated for antimicrobial activity against sensitive and resistant clinical strains of E.faecalit in both planktonic and hiofilm state using two methods,disk diffusion and broth microdilution.Results:Studied essential oils showed a good antimicrobial activity and high ability in E.faecalit biofilm eradication,whether for sensitive or multidrug-resistant strains,especially those of Origanum glandulosum and Thymbra capitata with interesting minimum inhibitory concentration,biofilm inhibitory concentration,and biofilm eradication concent ration values which doesn't exceed 0.063%,0.75%,and 1.5%,respectively.Conclusions:Findings of this study indicate that essential oils extracted from aromatic plants can be used in treatment of intractable oral infections,especially caused by biofilm of multidrugresistant E.faecalis.

Repeated hydrogen peroxide dosing briefly reduces cyanobacterial blooms and microcystin while increasing fecal bacteria indicators in a eutrophic pond

This study explored the effects of H_(2)O_(2)on Cyanobacteria and non-target microbes using fluorometry,microscopy,flow cytometry,and high throughput DNA sequencing of the 16S rRNA gene during a series of mesocosm and whole-ecosystem experiments in a eutrophic pond in NY,USA.The addition of H_(2)O_(2)(8 mg/L)significantly reduced Cyanobacteria concentrations during a majority of experiments(66%;6 of 9)and significantly increased eukaryotic green and unicellular brown algae in 78%and 45%of experiments,respectively.While heterotrophic bacteria declined significantly following H_(2)O_(2)addition in all experiments,bacteria indicative of potential fecal contamination(Escherichia coli,enterococcus,fecal coliform bacteria)consistently and significantly increased in response to H_(2)O_(2),evidencing a form of‘pollution swapping’.H_(2)O_(2)more effectively reduced Cyanobacteria in enclosed mesocosms compared to whole-ecosystem applications.Ten whole-pond H_(2)O_(2)applications over a twoyear period temporarily reduced cyanobacterial levels but never reduced concentrations below bloom thresholds and populations always rebounded in two weeks or less.The bacterial phyla of Cyanobacteria,Actinobacteria,and Planctomycetes were the most negatively impacted by H_(2)O_(2).Microcystis was always reduced by H_(2)O_(2),as was the toxin microcystin,but Microcystis remained dominant even after repeated H_(2)O_(2)treatments.Although H_(2)O_(2)favored the growth of eukaryotic algae over potentially harmful Cyanobacteria,the inability of H_(2)O_(2)to end cyanobacterial blooms in this eutrophic waterbody suggests it is a non-ideal mitigation approach in high biomass ecosystems and should be used judiciously due to potential negative impacts on non-target organisms and promotion of bacteria indicative of fecal contamination.

Study on tongue coating microbes of bitter taste,sticky and greasy taste in chronic atrophic gastritis

OBJECTIVE:To objectively reveal the relationship between tongue coating microbes and bitter taste,sticky and greasy taste in chronic atrophic gastritis(CAG)patients.METHODS:16S rRNA high-throughput sequencing was used to detect bacterial diversity and community composition of tongue coating microbes from samples of CAG patients.LEfSe algorithm was used for discovering the different tongue coating microbes in CAG patients with or without bitter taste,also that in CAG patients with or without sticky and greasy taste.RESULTS:We respectively compared the features of tongue coating microbes in bitter taste,sticky and greasy taste of CAG patients.At the genus level,25 tongue coating microbes were significantly different in CAG patients with bitter taste or without bitter taste;17 tongue coating microbes were significantly different in CAG patients with sticky and greasy taste or without sticky and greasy taste.Campylobacter and Rothia were closely related to CAG patients with bitter taste.enterococcus,Serratia,Leptotrichia and Selenomonas were closely related to CAG patients with stick and greasy taste.CONCLUSION:Campylobacter and Rothia possibly contribute to bitter taste of CAG patients,and enterococcus,Serratia,Leptotrichia and Selenomonas contribute to stick and greasy taste of CAG patients,which is potential for the diagnosis and treatment of CAG.

The rough crystal of exopolysaccharide produced by enterococcus durans, a strain of lactic acid bacteria screened from the intestine of a cock in our laboratory, was purified by CM-cellulose column chromatography, DEAE-Sephadex A-25 ionexchange and Sephadex G-100 gel chromatography to give EPS-Ⅰ. The EPS-Ⅰ was eluted as a single peak in HPLC analysis, indicating the homogeneity of EPS-Ⅰ and free from low-molecular-weight polysaccharides. The molecular weight of the EPS-Ⅰ was determined as 42 000 by the light scattering method. The result of its elemental analysis was C 41.08% and H 7.23% without the elements of N, P and S. Monosaccharide analysis showed that it was composed of Glc and Man in a molar ratio of 4∶1. Sugar composition analysis, methylation analysis and 1H and 13C NMR spectroscopy revealed that the EPS-Ⅰ was composed of pentasaccharide repeating units. The sequence of sugar residues was determined by using two-dimensional NMR, including heteronuclear multiple-bond correlation(HMBC) and nuclear overhauser effect spectroscopy(NOESY). The structure of the pentasaccharide repeating unit of EPS-Ⅰ was given, a new excellular polysaccharide from lactic acid bacterium compared with other EPSs was reported.

Virulence factors of enterococcus strains isolated from patients with inflammatory bowel disease

AIM: To determine the features of enterococcus that contribute to the development and maintenance of the inflammatory process in patients with inflammatory bowel disease (IBD). METHODS: Multiplex polymerase chain reaction (PCR) was applied to assess the presence of genes that encode virulence factors [surface aggregating protein (asa1), gelatinase (gelE), cytolysin (cylA), extracellular surface protein (esp) and hyaluronidase (hyl)] in the genomic DNA of 28 strains of enterococcus isolated from the intestinal tissues of children with IBD (n =16) and of children without IBD (controls; n = 12). Additionally, strains with confirmed presence of the gelE gene were tested by PCR for the presence of quorum sensing genes (fsrA, fsrB, fsrC) that control the gelatinase production. Gelatinase activity was tested on agar plates containing 1.6% gelatin. We also analysed the ability of enterococcus strains to release and decompose hydrogen peroxide (using Analytical Merckoquant peroxide test strips) and tested their ability to adhere to Caco-2 human gut epithelium cells and form biofilms in vitro. RESULTS: A comparison of the genomes of enterococcus strains isolated from the inflamed mucosa of patients with IBD with those of the control group showed statistically significant differences in the frequency of theasa1 gene and thegelE gene. Furthermore, the cumulative occurrence of different virulence genes in the genome of a single strain ofenterococcus isolated from the IBD patient group is greater than in a strain from the control group, although no significant difference was found. Statistically significant differences in the decomposition of hydrogen peroxide and adherence to the Caco-2 epithelial cell line between the strains from the patient group and control group were demonstrated. The results also showed that profuse biofilm production was more frequent amongenterococcus strains isolated from children with IBD than in control strains. Virulence determinants and biofilm formation in clinical isolates of enterococcus: A cross-sectional study

Objective:To evaluate the relationship between biofilm formation and incidence of virulence determinants in clinical isolates of enterococcus.Methods:In this cross-sectional study,the clinical isolates of enterococcus strains were collected from the university teaching hospitals in Ahvaz,Iran from June 2017 to June 2018.Then,the prevalence of enterococcus species,antibiotic resistance,virulence factors,and biofilm-producing ability were determined.Results:Of the 119 tested isolates,17(14.3%)were enterococcus faecalis,72(60.5%)were enterococcus faecium and 30(25.2%)were other enterococcus spp.Gelatinase was detected in 97(81.5%)isolates,enterococcal surface protein in 41(34.5%)isolates,serine protease in 39(32.8%)cases,accessory colonization factor in 111(93.3%)cases and pathogenicity islands in 17(14.3%)cases.The biofilm formation ability was observed in 75(63.0%)of all isolates and the association between the presence of enterococcal surface protein gene and biofilm formation was statistically significant.Higher resistance to vancomycin,gentamycin,and teicoplanin was indicated in enterococcus faecium with 81.8%,58.4%,and 85.7%resistance rate,respectively.All enterococcus faecalis isolates were sensitive to teicoplanin and vancomycin.Conclusions:The presence of antibiotic-resistance with several virulence factors in enterococcus spp has become a concern.High prevalence of enterococcal surface protein gene among biofilm-producing isolates suggests a potential relation between biofilm formation and the enterococcal surface protein gene,and further studies are needed to identify the mechanism of biofilm inhibition.

Hospital-Adapted Clonal Complex 17 <i>enterococcus Faecium</i>Found among Sand Enterococcal Isolates

Though poorly studied, sand is an environment with an extended degree of interaction with man. Enterococcal strains can be found in sand but we do not know to what extent these ubiquitous opportunistic nosocomial pathogens isolated from sand carry antimicrobial resistances and virulence traits. In an attempt to fill in this knowledge gap, two distinct types of sand (beach and children playground) were examined concerning composition in enterococcal species, genetic diversity of isolates and abundance of resistance to antimicrobials and virulence traits. Five different species were found, namely enterococcus faecium, enterococcus faecalis, enterococcus hirae, enterococcus flavescens and enterococcus casseliflavus. Although genetic diversity was evident, two different E. faecium clones, common to the two types of sand, were detected, suggesting the existence of clones well adapted to this specific environment or from a common source. E. faecium was associated with multiple antibiotic resistances, including to fluoroquinolones and tetracycline that are commonly used by veterinarians and clinicians. Among the multiresistant E. faecium strains from beach sand, two were from sequence type (ST) 442, which belongs to the wide-spread Hospital-adapted clade CC17. They both carried the esp gene and the genomic island associated with CC17. The other virulence factors screened were disseminated among E. faecalis strains, but seldom detected in the other species, evidencing the existence, in these environments, of E. faecalis strains carrying the same virulence factors as the clinical ones. The present work thus stresses the need to follow-up the presence and characterization of enterococcal strains from both beach and children playground sands and of including these environments in the epidemiological global analysis of enterococcal isolates.
