Effects of virus infection on expression of cell cycle regulatory proteins in the unicellular marine algae emiliania huxleyi

The authors have investigated the biochemical events by which marine algal virus infection induces cell cycle arrest. The key G 2 /M-phase regulatory proteins are analyzed by immunobloting in unicel-lular emiliania huxleyi,suggesting that virus induced cell cycle arrest is related with virus＇s effect on cyclins and cyclin dependent kinases. E. huxleyi virus（EhV） represses Cdc2/cyclinB complex activity by inhibiting the activity of Cdc2 kinase in a phosphorylation-related manner,blocking host cells G 2 /M checkpoint. Dephosphorylated /inactive Cdc25C combined with up-regulation of Wee1 expression at early infect period appears to be important mechanisms by which EhV represses Cdc2/cyclinB complex activity that is required for entry into M phase. This study has allowed us to confirm that algal virus infection leads to selective activation or inhibition of certain cell-cycle factors,which may play a significant role in establishing a more efficient environment for viral gene expression and DNA replication.

The interaction between the bloom-forming alga emiliania huxleyi(Prymnesiophyceae) and its specific large dsDNA virus(EhV) is a major factor determining the fate of carbon in the *** induces a metabolic switch in various host metabolic pathways,and particularly enhances its replication and survival by modulating host cell lipid *** regulation can occur at both transcriptional and epigenetic(microRNA) levels,though combined transcriptomic and microRNAomic responses to viral infection are still largely unexplored for marine *** combined transcriptomic,microRNAomic and lipidomic analyses to understand the molecular mechanisms to virus-driven lipid metabolism *** show that EhV induced profound transcriptome remodeling targeted toward lipid *** sequencing revealed 26 microRNAs potentially regulating 13560 protein-coding *** enrichment analysis of five differentially expressed microRNA-targeted genes predicted suppression in FAs hydrolysis and sphingolipids synthesis during viral ***,we detected a significant increase in FAs(including simple FAs and OAHFAs) content and decreases in their biodegradation gene expression,supporting viral replication and *** contrast,sphingolipids(including Cer and CerGl) showed a nearly complete decrease and coincided with down-regulation of microRNA-targeted sphingolipid synthesis-related genes(such as DCDS and CERS),indicating the competitive inhibition of host de novo sphingolipid synthesis during lytic viral infection.

The bloom-forming coccolithophore emiliania huxleyi and its specific coccolithoviruses(*** virus,EhV) plays an important role in the oceanic carbon cycles and climate *** have developed a novel "virocell metabolism" model to meet their metabolic ***,the regulatory mechanism of this metabolism is still not fully ***,nontargeted lipidomics analysis was performed to explore the cellular sphingolipid metabolic pathways mediating the interaction between *** BOF 92 and its lytic virus EhV 99 *** show that thirteen significantly changed sphingolipids(including Cer and CerGl) were identified during viral *** lipidomics profiling revealed a nearly complete decrease of differential sphingolipids at both early(6 hpi) and later stages(45 hpi) as lytic infection proceeded and coincided with downregulation of host denovo sphingolipid genes,suggesting the competitive inhibition of host sphingolipid synthesis to produce the required building blocks for viral production replication and ***,biomarker candidates Cer38:1;2 and Cer 40:2;2 isomer 1 may indicate the general dysfunctions in *** in response to viral *** study provides insight into how EhV depends on the hijacking of host sphingolipid metabolism during the chemical "arms race" in the ocean.

The interaction between emiliania huxleyi and its specific large dsDNA virus(*** virus,EhV) has been recognized as a major ecological and Biogeochemistry factor in the marine environment During co-evolution,EhV "hijacked" a group of key enzyme genes in the de novo biosynthesis pathway of sphingolipid from the host genome through gene horizontal transfer The function of these genes is not very clear so ***,viral genes encode sterol desaturase(EhV-SD) and fatty acid desaturase(EhV-FAD) genes in EhV99 B1 genome were cloned into Saccharomyces cerevisiae with the two corresponding gene-deficient strains and induced to *** comparing the lipid composition and content of recombinant yeast and deficient yeast cells and constructing a yeast lipid metabolic pathway map based on differential lipid *** results reveal that EhV-SD and EhV-FAD deeply rewired lipid metabolism in yeast cells,and lipid composition and content in the recombinants changed *** EhV-SD regulates the contents of various sterols,which indirectly affected the metabolism of ceramide and neutral *** EhV-FAD effected the contents of sphingolipids and fatty acid,and changes in the ratio of saturated to unsaturated fatty *** findings provide a biochemical basis for exploring the function of unique genes of EhV and new insights into how viruses in the ocean reconstruct host metabolism and life cycle.

The distribution and abundance of emiliania huxleyi（E. huxleyi） assemblages in the marine sediments of the Aravonitsa Plateau, Greece, and from the eastern Mediterranean are used（1） to evaluate the calcareous nannoplankton NN21 a and NN21 b biozones and the NN21a/NN21 b boundary, and（2） to analyze the palaeoenvironmental and palaeoclimatic conditions prevailing in this interval. The sediment succession displays varied E. huxleyi assemblages and these are interpreted as reflecting climatic variability during marine isotope stages MIS 1-8.

大气CO浓度升高，使得溶于海洋的CO增加，改变表层海洋碳酸系统的平衡，降低海水pH(海水酸化)和CaCOB3B饱和度。这种海洋酸化现象会影响钙化生物的钙化作用以及相关生态过程。颗石藻类作为主要的浮游钙化生物，在海洋碳循环过程中起着重要的作用。为此，本文探讨了CO浓度升高(海水酸化)对赫氏颗石藻(emiliania huxleyi)的影响，并揭示了其在酸化状态下对阳光紫外辐射(UVR)的生理响应。主要结果如下：通过调整培系统中CO分压(pCO为350，780，1460 ppmv)，调控培养液中的pH水平分别为8.2，7.9和7.6。在半连续培养过程中，我们检测了海水酸化对颗石藻生长、形态、光合和钙化固碳及其UVR敏感性的影响。在不同pH条件下，该藻的生长速率在0.7-1.1d之间变动，经一定时间(8天左右)的适应后，受pH变化的影响不显著(维持在0.9 d左右)。海水酸化对光合作用的影响也不显著，然而，其对钙化作用的抑制作用特别明显，使其降低42.3-89.4％。为此，颗石层(钙壳)厚度相应变薄21-33％。在酸化状态下生长的细胞，经受UVR辐射处理时，无论是钙化还是光合固碳速率，UVR导致的抑制作用均明显增大：在pH为8.2时，钙化作用、光合作用受UVR的抑制分别为5％和45％，pH下降到7.6时，抑制率分别增加到了15％和90％。由于UVR对钙化固碳的抑制率偏高，使得无机与有机固碳比值(钙化固碳C/光合固碳P)随酸化急剧降低。酸化状态下UVR导致的较高抑制率，与钙化壳变薄有关;颗石层能使细胞少接受20-25％的UVR。颗石藻的钙化壳(颗石层)，起着滤除或减少有害紫外辐射的作用，这对其应付高强度的阳光辐射，减少光抑制非常重要。大气CO浓度升高导致的海洋酸化，减少颗石藻的钙化量，会使细胞接受UVR的辐射量增加，UVR导致的损伤增大，PIC/POC值降低。为此，与颗石藻类相关的生物地球化学及生态学过程，会受到海洋酸化的影响。
