Influence of ellagic acid on prostate cancer cell proliferation:A caspase-dependent pathway

Objective:To evaluate the effect of allagic acid treatment on the cell viability of human prostate cancer ***:ellagic acid(10-100 mol/L) treatment(48 h) of human prostate carcinoma PC3 cells was found to result in a dose-dependent inhibition of cell growth and apoptosis of PC3 cells as assessed by MTT assay,western *** cytometry and confocal ***:We observed that ellagic acid treatment of PC3 cells resulted in a dose dependent inhibition of cell growth/cell *** ellagic acid caused cell growth inhibition was found to be accompanied by induction of apoptosis,as assessed by the cleavage of poly (ADP-ribose) polymerase(PARP) and morphological ***,induction of apoptosis accompanied a decrease in the levels of antiapoptotic protein Bcl-2 and increase in proapoptotic protein *** shifting the Bax:Bcl-2 ratio in favor of *** acid treatment of PC3 cells was also found to result in significant activation of caspases,as shown by the dose dependent decrease in the protein expression of procaspase-3,-6.-8 and -*** ellagic acid-mediated induction of apoptosis was significantly(80%-90%) inhibited by the caspase inhibitor N-benzyloxycarbonyl-Val-Ala-Asp(OMe)-fluoromethylketone(Z-VAD-FMK).Thus these data suggested an essential role of caspases in ellagic acid-mediated apoptosis of PC3 ***:It is tempting to suggest that consumption of tropical pigmented fruits and vegetables could be an effective strategy to combat prostate cancer.

OBJECTIVE Astroglia support neurons by providing substrates for neuronal metabolism, glutamate clearance and anti-oxidant protection. Nuclear factor erythroid 2-related factor 2(Nrf2) is the main switch of intracellular antioxidant defense system. Also, Nrf2 signaling is recognized to activate the neurotrophic pathway to replace/protect damaged organelles. ellagic acid(EA), an excretion component of fruits and nuts, displays anti-oxidant, cardioprotective and antiinflammatory activities. However, few studies have been focused on the neurotrophic properties of EA. Our study investigated whether EA could enhance astroglia neurotrophic effects to support neurons and the underlying mechanisms as well. METHODS Primary neuron-enriched cultures, primary astroglia-enriched cultures and primary neuron-astroglia co-cultures were applied to detect whether pharmacological regulation of astroglia function by EA could be utilized to overcome neuronal death. RESULTS This study indicated that EA promoted neuronal survival. Furtherly, astroglia Nrf2 participate in EA-elicited neuronal survival with the following scenarios. First, EA induced astroglia proliferation, neurotrophic factors release and Nrf2 activation. Second, astroglia-targeted Nrf2 si RNA inhibited EA-mediated astrogliosis,neurotrophic factors excretion and neuronal survival. CONCLUSION EA mediated astroglia Nrf2 activation to enhanced neurotrophic effects on neurons, and these findings might provide new strategies for neurotrophic factor-based treatment of neurodegenerative disease.

Pomegranate leaf attenuates lipid absorption in the small intestine in hyperlipidemic mice by inhibiting lipase activity

Pomegranate leaf(PGL)has a definite role in regulating lipid ***,pharmacokinetic results show the main active ingredient,ellagic acid,in PGL has lower oral bioavailability,suggesting that the lipid-lowering effect of PGL may act through inhibiting lipid absorption in the small *** results demonstrated that pomegranate leaf and its main active ingredients(i.e.,ellagic acid,gallic acid,pyrogallic acid and tannic acid)were capable of inhibiting pancreatic lipase activity in *** computational molecular docking,the four ingredients had good affinity for pancreatic *** lipid overload experiments showed that a large dosage of PGL significantly reduced serum total cholesterol(TG)and triglycerides(TC)levels in addition to inhibiting intestinal lipase activity,which demonstrated that PGL could inhibit lipase activity and reduce the absorption of *** also found that PGL could reverse the reduced tight-junction protein expression due to intestinal lipid overload,promote Occludin and Claudin4 expression in the small intestine,and enhance the intestinal mucosal *** conclusion,we demonstrated that PGL can inhibit lipid absorption and reduce blood TG and TC by targeting pancreatic lipase,promoting tight-junction protein expression and thereby preventing intestinal mucosa damage from an overload of lipids in the intestine.

Phosalone-induced inflammation and oxidative stress in the colon: evaluation and treatment

AIM: To investigate the side effects of phosalone on intestinal cells and to evaluate benefits of ellagic acid(EA) as a ***: In order to conduct an in vivo study, a rat model was used. The rats were divided into ten groups based on the materials used in the experiment and their dosage. The first group was fed normally. The second group was administered EA through gavage. Next Four groups were given(1/3, 1/5, 1/10, 1/20) LD50 phosalone; an organophosphorus compound. The last four groups received(1/3, 1/5, 1/10, 1/20) LD50 phosalone and of EA. After one month, the rats were sacrificed and their colon cells were examined to evaluate the level of inflammation, proteins and oxidative stress ***: The results of this research show that phosalone elevates oxidative stress and changes the level of tumor necrosis factor-a(TNF-α), interlukin-6β(IL-6β) and nuclear factor(NF)-κB proteins. EA administration reduced phosalone toxicity and changed oxidative stress and inflammatory markers for all phosalone doses. Overall changes in reduction of TNF-α(230.47 ± 16.55 pg/mg protein vs 546.43 ± 45.24 pg/mg protein, P < 0.001), IL-6β(15.85 ± 1.03 pg/mg protein vs 21.55 ± 1.3 pg/mg protein, P < 0.05), and NF-κB(32.47 ± 4.85 pg/mg protein vs 51.41 ± 0.71 pg/mg protein, P < 0.05) manifest that the efficacy of EA is more viable for 1/3 LD50 dose of phosalone. Furthermore, EA is effective to counteract the negative outcomes of oxidative stress. When EA was used to treat 1/3 LD50 of phosalone's side effects, it improved the level of ACh E activity(48.5% ± 6% vs 25% ± 7%, P < 0.05), TTM(0.391 ± 0.008 mmol/L vs 0.249 ± 0.032 mmol/L, P < 0.05), FRAP(46.04 ± 5.005 μmol/L vs 18.22 ± 1.9 μmol/L, P < 0.01) and MPO(0.222 ± 0.019 U/mg protein vs 0.387 ± 0.04 U/mg protein, P < 0.05). CONCLUSION: This research highlights that EA is effective to alleviate the side effects of phosalone by reducing the level of oxidative stress and inflammatory proteins.
