Scope: A previous study found interactions between sterol regulatory element binding protein gene(SREBF1) gene polymorphism and dietary cholesterol intake on serum lipids. This study aims to confirm and extend these findings by characterizing the effects of cholesterol from egg yolks on lipid profiles in healthy young adults with different SREBF1 genetic makeup. Methods Methods: A randomized two-period crossover intervention to serve two egg yolks/day was designed for 32 rs2236513/rs2297508/rs4925119 C/G/G carriers and 32 AA/CC/AA homozygotes. However, uneven egg sizes during period 1 made the intervention unable to achieve the protocol’s recommended cholesterol intake level, and the intervention was modified in period 2 to be 0.8 g egg yolk/day/kg body weight. Results Results: The intervention increased serum lipids in period 2 but not in period 1. Furthermore, intervention effects on LDL-C and LDL-C/HDL-C among C/G/G carriers in period 2 were two times and eight times larger, respectively, than those among AA/CC/AA homozygotes. In contrast, the intervention effect on HDL-C in period 2 among AA/CC/AA homozygotes was sevenfold that of C/G/G carriers. Conclusion Conclusion: These findings indicate a diminished ability among C/G/G carriers to regulate excess dietary cholesterol, suggesting they may need a more restrictive egg yolk intake compared with AA/CC/AA homozygotes.

Influence of Dietary Isoflavonoids on Stability of egg yolk Lipids

An experiment was conducted to investigate the influence of dietary tea polyphenol (TP) and daidzein (DA) on lipid stability of egg yolk. Two-hundred and seventy 27-week-old Hisex Brown laying hens were divided into nine groups, fed a diet supplemented with 0, 5, 10, 20, 40 mg kg-1 TP and 5, 10, 20, 40 mg kg-1 DA respectively for a period of 8 weeks ad libitum. The dietary addition of TP and DA both could reduce egg yolk LPO content significantly at the end of 4-week experiment (P egg yolk lipid stable at the supplemental dose of 10 mg kg-1 diet respectively. A significant liner reduction in egg yolk LPO content was found with the increasing of dietary TP and DA. The egg yolk LPO content was decreased by 23.1 and 27.0% (Pegg yolk were decreased significantly with the dietary addition of TP or DA respectively (P < 0.05). There was no significant difference between the effects of TP and DA on improvement of layers' antioxidative capacity. The dietary TP and DA both could improve the bird's antioxidative level directly irrelevant to SOD and GSH-Px activities.

Astaxanthin(Ax)and lutein are important fat-soluble pigments and essential nutrients for human and animal health.Haematococcus pluvialis microalga and Phaffia rhodozyma yeast are ideal species for commercial Ax production.Marigold flowers are a main source of commercial lutein.Dynamics of dietary Ax and lutein in the gastrointestinal tract are similar to lipids,but their activities are tremendously challenged by many physiological and dietary factors;few data are available about these in poultry.Dietary Ax and lutein have insignificant effects on egg production and egg physical properties,but have pronounced effects on yolk color,nutrition,and functionality.The two pigments can also enhance antioxidative capacity and immune function of laying hens.A few studies have shown that Ax and lutein can improve fertilization and hatchability of laying hens.Considering the pigmentation and health benefits of Ax and lutein from hen feed to human food,the commercial availability,chicken yolk improvement,and immune function of Ax and lutein are the focuses of this review.The potential roles of carotenoids in the cytokine storm and gut microbiota are also briefly presented.The bioavailability,metabolism,and deposition of Ax and lutein in laying hens are suggested for future research.

Deinagkistrodonacutus caused a large number of snakebite incidents related to human in southern China.The intravenous administration of conventional antivenoms produced in horses or other mammal is the mainstay and the only specific treatment of snakebite.Avian antibodies（IgY）,isolated from chicken egg yolks,are regarded as a new alternative to be applied for the snake envenomations.Based on the reasons,we produced IgY antibodies against D.acutus venom and assessed its protective capacity.In our studies,two 24-week-old laying white leghorn chickens were immunized intramuscularly in the breast region at multiple sites with 0.293 mg of D.acutus venoms to generate IgY antibodies against the venom antigen.After the first immunization,Chickens received three boost at 21 days apart to raise the level of specific antibodies in egg yolk.After being extracted by caprylic acid and ammonium sulphate precipitation,the crude extracts of antibodies were further purified by affinity chromatography.The purity and molecular weight of IgY antibodies was determined by SDS-PAGE and the specificity of antibodies was studied by immunodiffusion assay.Indirect ELISA and Western blot analysis were used to determine the titer and antigen recognition repertoire of IgY antibodies respectively.The median effective dose（ED50） of anti-Deinagkistrodonacutusvenoms IgY antibodies was 14.14 mg IgY/ kg body weight with the challenge dose as 3LD50.150 ug of IgY antibodies was able to neutralized 50%of 3MHD of venom.The results indicate that antibodies generated in chicken may probably to be a new star which is superior alternative then conventional immunoglobulins in respect of the treatment for snake envenomation and in the future.

Vitellogenin content in fat body and ovary homogenates of workers and queens of Melipona quadrifasciata anthidioides during vitellogenesis

Vitellogenin （Vg） is an egg yolk protein that is produced primarily in the fat body of most female insects. In the advanced social structure of eusocial honeybees, the presence of the queen inhibits egg maturation in the workers＇ ovaries. However in the stingless bee Melipona quadrifasciata, the workers always develop ovaries and lay a certain amount of eggs while provisioning the brood cells with larval food during what is known as the worker nurse phase. The present work is a comparative study of the presence of Vg in homogenates of the fat bodies and ovaries of the nurse workers, and the virgin and physogastric queens of M. quadrifasciata. The presence of Vg was determined by sodium dodecyl sulfate polyacrylamide gel electrophoresis and irnmunoblotting using Apis mellifera anti-egg antibody. Vg was not detected in the fat bodies or ovaries of the workers, but it was found in the ovaries of virgin and physogastric queens and in the fat body ofphysogastric queens. The results are discussed, taking into account the reproductive state of the individuals and the other possible roles of Vg, such as a storage protein for metoabolism of other organs.

Investigation of Two Precipitation Methods for Extracting Immunoglobulin Y(IgY)from egg yolks

Two groups of hens(control and immunization group)were arranged in an experimental design with an immunization schedule of 3 injections of BSA antigen.IgY antibodies were extracted from egg yolks by two precipitation processes(chloroform and polyethylene glycol precipitates)and quantified using a standard curve of protein concentration.The purification of IgY was confirmed by SDS-PAGE.Total protein extracted from egg yoks were less contaminated with yellow pigments(lutein and zeaxanthin)nd by using chloroform precipitate.The 2 week post-immunization,IgY-1 concentration increased respectively to 3903±726μg.ml(chloroform-1 extraction process)and 2937±294μg.ml(PEG extraction process)(P

IgY technology: Methods for developing and evaluating avian immunoglobulins for the in vitro detection of biomolecules

The term“IgY technology”was introduced in the literature in the mid 1990s to describe a procedure involving immunization of avian species,mainly laying hens and consequent isolation of the polyclonal IgYs from the“immune”egg yolk(thus avoiding bleeding and animal stress).IgYs have been applied to various fields of medicine and biotechnology.The present article will deal with specific aspects of IgY technology,focusing on the currently reported methods for developing,isolating,evaluating and storing polyclonal IgYs.Other topics such as current information on isolation protocols or evaluation of IgYs from different avian species are also discussed.Specific advantages of IgY technology(e.g.,novel antibody specificities that may emerge via the avian immune system)will also be discussed.Recent in vitro applications of polyclonal egg yolk-derived IgYs to the field of disease diagnosis in human and veterinary medicine through in vitro immunodetection of target biomolecules will be presented.Moreover,ethical aspects associated with animal well-being as well as new promising approaches that are relevant to the original IgY technology(e.g.,development of monoclonal IgYs and IgY-like antibodies through the phage display technique or in transgenic chickens)and future prospects in the area will also be mentioned.
