Polysaccharide extracts of Astragalus membranaceus and Atractylodes macrocephala promote intestinal epithelial cell migration by activating the polyamine-mediated K^+ channel

Astragalus membranaceus(Radix Astragali, RA) and Atractylodes macrocephala(Rhizoma Atractylodis Macrocephalae, RAM) are often used to treat gastrointestinal diseases. In the present study, we determined the effects of polysaccharides extracts from these two herbs on IEC-6 cell migration and explored the potential underlying mechanisms. A migration model with IEC-6 cells was induced using a single-edged razor blade along the diameter of cell layers in six-well polystyrene plates. The cells were grown in control--media or media containing spermidine(5 μmol·L^(-1), SPD), alpha-difluoromethylornithine(2.5 mmol·L^(-1), DFMO), 4-Aminopyridine-(40 μ-mol·L^(-1), 4-AP), the polysaccharide extracts of RA or RAM(50, 100, or 200 mg·L^(-1)), DFMO plus SPD, or DFMO plus polysaccharide extracts of RA or RAM for 12 or 24 h. Next, cytosolic free Ca^(2+)([Ca^(2+)]cyt) was measured using laser confocal microscopy, and cellular polyamine content was quantified with HPLC. Kv1.1 mRNA expression was assessed using RT-qPCR and Kv1.1 and RhoA protein expressions were measured with Western blotting analysis. A cell migration assay was carried out using Image-Pro Plus software. In addition, GC-MS was introduced to analyze the monosaccharide composition of both polysaccharide extracts. The resutls showed that treatment with polysaccharide extracts of RA or RAM significantly increased cellular polyamine content, elevated [Ca^(2+)]cyt and accelerated migration of IEC-6 cells, compared with the controls(P extracts not only reversed the inhibitory effects of DFMO on cellular polyamine content and [Ca^(2+)]cyt, but also restored IEC-6 cell migration to control level(P < 0.01 or < 0.05). Kv1.1 mRNA and protein expressions were increased(P < 0.05) after polysaccharide extract treatment in polyamine-deficient IEC-6 cells and RhoA protein expression was increased. Comparative analysis of chemical constituents, antimicrobial and antioxidant activities of ethylacetate extracts of Polygonum cuspidatum and its endophytic actinomycete, Streptomyces sp. A0916

The present study investigated the chemical composition of ethylacetate extracts from an endophytic actinomycete Streptomyces sp. A0916 and its host Polygonum cuspidatum. A comparative analysis of the antimicrobial and antioxidant properties of the extracts was also conducted. 32 compounds of P. cuspidatum and 23 compounds of Streptomyces sp. A0916 were isolated and identified by GC/MS. Antimicrobial activities of the extracts were evaluated using eight microbial strains(3 Gram-positive bacteria, 3 Gram-negative bacteria, and 2 fungi). The Streptomyces sp. A0916 extracts showed a wide range of antimicrobial activities and presented greater antimicrobial effectiveness than the P. cuspidatum extracts. The minimum inhibitory concentration(MIC) of Streptomyces sp. A0916 extracts against the ampicillin-resistant strain Enterococcus faecium SIIA843 was 32 μg·m L-1. Furthermore, the extracts had greater antimicrobial effect against Gram-positive bacteria than Gram-negative bacteria. Finally, the antioxidant activity of the Streptomyces sp. A0916 extracts was equal to that of the P. cuspidatum extracts. In conclusion, our results suggest that the endophytic actinomycetes of the medicinal plants are an important source of bioactive substances.

Identification of a DNase activated in Xenopus egg extracts undergoing apoptosis

A cell-free apoptosis system was established by adding dATP and cytochrome c to Xenopus laevis egg extracts S-150. Accompanied by an incubation process, an apoptosis-specific DNase was activated in egg extracts which depended on Mg 2+ and inhibited by Zn 2+. Two nucleases existing in egg extracts were revealed by in-gel nuclease assay. Further experiments showed that 27 ku nuclease which was different from other Ca 2+/Mg 2+-dependent nucleases was a possible candidate involved in apoptosis.

Chemical Composition and Use of Momordica charantia L. and Hyptis spicigera Lam. extracts as Mosquito Larvicides and Insect Growth Regulators against Malarial Vector, Anopheles gambiae Giles

Background: Mosquitoes are responsible of numerous vector-borne diseases and among these diseases is malaria which takes away lives of thousands of people, especially children of under five, particularly in Africa. To reduce the mortality and economic burdens of this deadly disease, synthetic insecticide has been of use to control its main vector, Anopheles gambiae. Due to adverse effects caused by these conventional products on non-target organisms and the environment, plants have been of first choice as they proved their effectiveness against mosquitoes and are said to be eco-friendly and relatively safer. Therefore, the aim of this study was to screen the phytochemicals and determine the larvicidal and adult emergence exhibitory activities of Momordica charantia and Hyptis spicigera leaves extracts against Anopheles gambiae larvae. Methods: The WHO standard protocol for larvicidal activity and IGRs was followed up and concentrations from 500 - 2000 ppm for larvicidal and 500 - 1500 ppm for IGRs were set up. Results: There was the presence of alkaloids, flavonoids, tannins, phenols, steroids, terpenoids and oils in Momordica charantia against alkaloids, taponins, tannins, phenols, steroids, terpenoids and oils in Hyptis spicigera. Methanol crude extract registered the lowest LC50 value of 270.6 ppm followed by ethyl acetate and hexane fractions recorded the LC50 of 742.1 and 756 ppm, respectively for larvicidal activity of Momordica charantia against LC50 of 760 and 867.5 ppm for hexane fraction and methanol crude extract, respectively for Hyptis spicigera. Methanol crude extract and hexane fraction of both plants as well as the ethyl acetate fraction of M. charantia registered greater emergence inhibition with IE50 values of 590.2, 842.3 and 982 ppm for methanol crude extract, hexane and ethyl acetate fractions of M. charantia, respectively against 901.7 and 873.2 for methanol crude extract and hexane fraction of H. spi

Our study focused on phytochemical tests and evaluation of the anti-free radical activity of seed extracts of two oleaginous plants from Gabon used in traditional medicine or as condiments: Panda oleosa and Isolona hexaloba. The extraction was carried out by maceration with solvents of increasing polarity: cyclohexane, trichloroethylene, acetone, ethanol and finally distilled water. The total yields of the extracts are about 69.50% for Panda oleosa and 34.28% for Isolona hexaloba. The phytochemical tests carried out on the extracts of the seeds of Panda and Isolona highlight in both seeds the presence of alkaloids, polyphenols, triterpenes, carotenoids, reducing compounds, flavonoids, total sugars, coumarins, anthraquinones, free quinones, free anthracene derivatives, and terpenoids. Isolona seeds also contain leucoanthocyanins, sterols, cardiac glycosides and saponins. Phytochemical tests revealed the absence of tannins and mucilage in both seeds. The free radical scavenging activity was measured by scavenging the free radical cation of 2,2’-azino-bis[3-ethylbenzothiazoline-6-sulfonic acid] (ABTS-+) with gallic acid as the reference antioxidant. The results of the free radical scavenging activity of the aqueous and ethanolic extracts of both seeds showed that the aqueous extracts were more active than the ethanolic extracts. The IC50s of the aqueous and ethanolic extracts of Panda seeds are 40 and 60 μg·mL-1 respectively, and those of the aqueous and ethanolic extracts of Isolona are 37.5 and 95 μg·mL-1 respectively. Gallic acid, the reference antioxidant (IC50 = 0.37 μg·mL-1) is about 10 times more active than the aqueous extracts of both seeds, 16 times more active than the ethanolic extract of Panda and 25 times more active than the ethanolic extract of Isolena.

Antioxidant Properties and Antimicrobial Activity in the extracts of Two Edible Mushroom, Pleurotus sajor caju and Schizophyllum commune

extracts of two edible mushrooms, Pleurotus sajor-caju (commercial) and Schizophyllum commune (wild) were used to compare the antioxidant and antimicrobial properties. Aqueous and three types of organic solvents, like 50% of ethanol, methanol and acetone extracts were used in trial. DPPH scavenging activity in P. sajor-caju extract was determined in the range of 53.13% to 85.08%, whereas extracts of S. commune were observed in the range of 54.11% to 97.19% at a concentration of 5 mg/ml. The highest DPPH scavenging activity of 97.19% was observed in ethanol extract of S. commune (97.19%), higher than butyl hydroxytoluene (BTH). Half effective concentration (EC50) in extracts of P. sajor-caju was found in the range of 1.47 to 4.23 mg/ml and that of S. commune in the range of 1.52 to 4.52 mg/ml. The reducing power of P. sajor caju aqueous concentration extract was found to be the closest of 3.353 (700 nm) that of antioxidant activity to BHT (3.445) at 2 mg/ml concentration. The best reducing power EC50 was obtained in P. sajor caju aqueous extract (0.09 mg/ml), but in S. commune with acetone extract (0.22 mg/ml). Minimum inhibition concentration (MIC) was compared in extracts of mushrooms in various Vibrio species. All extracts were able to inhibit V. harveyi growth with MIC of lower than 1.25 mg/ml. In aqueous and methanol extracts of current study showed that bacteria inhibition activity occurred at the concentration of P. sajor-caju was able to act as reducing agent as functional as the commercial antioxidant agent, BHT. Crude extracts of P. sajor-caju and S. commune were observed to contain antibacterial potential against these mentioned Vibrio bacteria.

Divergent Immunomodulatory Effects of extracts and Phenolic Compounds from the Fern Osmunda Japonica Thunb

Objective： To study possible immunobiological potential of Osmunda japonica Thunb. Methods： Immunomodulatory effects of ethanol extracts prepared from rhizomes of O. japonica and phenolic compounds isolated from the extracts were investigated under the in vitro conditions using the rat peritoneal cells （2 × 10_6/mL; 24 h culture）. Biosynthesis of nitric oxide （NO） was assayed by Griess reagent, production of prostaglandin E2 （PGE2） and secretion of cytokines were determined by enzyme-linked immunoabsorbent assay. Resalts： The extracts activated dose dependently, with the onset at 2.5-5 μ mol/L concentrations, the high output NO production, and secretion of interferon-r, （IFN-r）, tumor necrosis factor-oL （TNF-α） and interleukin-1 （IL-1 13 ）. Mild enhancement of NO was produced by the aldehyde-type phenolics 4-hydroxybenzaldehyde and 3,4-hydroxybenzaldehyde. In contrasts, the acetone-type phenolics 4-hydroxybenzalacetone and 3,4-hydroxybenzalacetone inhibited production of immune mediators including cytokines （TNF-α, IL-β, IL-6）, NO, and PGE2. The 3,4-hydroxybenzalacetone was more effective than 4-hydroxybenzaldehyde. The ICsoS estimates ranged within the interval of 5-10 μ mol/L. No signs of cytotoxicity were observed up to the 50 μ-mol/L concentration of the compounds. Coaclasiea： Phenolic compounds contained in medicinal herb Osmunda japonica possess distinct immunomodulatory activity.

Effects of Angelica sinensis extracts on lipid oxidation in fish feeds and growth performance of juvenile Jian carp(Cyprinus carpio var. Jian)

The study was to explore the effect of the extracts of Angelica sinensis(EAs) on lipid oxidation in fish feeds compared with ethoxyquin(EQ) and the effect of dietary EAs on growth performance of carp(Cyprinus carpio var. Jian). Firstly, fish feeds were respectively added with EQ, and ethyl ether extract ethyl acetate extract(EAE), acetone extract, ethanol extract(EE) and aqueous extract(AQE) of Angelica sinensis, except for the control. The results showed that EAs and EQ inhibited lipid oxidation in fish feed(P < 0.05). Of all of the examined EAs, EAE showed the strongest protective effects against lipid oxidation(P < 0.05). Moreover, EAE at high concentrations showed a stronger effect on lipid oxidation compared with EQ(P < 0.05). Then, 7 experimental diets respectively supplemented with 0.0, 1.0, 2.0, 3.0, 4.0, 5.0 and 6.0 g/kg of EAE were fed to the respective treatment groups for 30 d. Four replicates were performed for each treatment group; 20 carp(mean weight: 12.10 ± 0.13 g) were in each replicate. The result indicated that dietary EAE improved the growth performance in carp(P < 0.05). The appropriate con centration of EAE for carp growth was estimated to be 3.643 g/kg diet. Thus, EAE could be used as a natural antioxidant in feeds for Jian carp.

The control of Spodoptera frugiperda is based on synthetic insecticides,so some alternatives are the use of entomopathogenic fungi(EF)and neem *** objective of the study was to evaluate in vitro effectiveness of native EF and neem extracts on *** *** EF were identified by DNA sequencing of ITS regions from three EF(Fusarium solani,Metarrhizium robertsii,Nigrospora spherica and Penicillium citrinum).They were evaluated in concentrations of 1×10⁸spores/*** addition,a second bioassay was carried out evaluating only ***,*** and *** and the addition of vegetable *** the other hand,extraction of secondary metabolites from neem seed(Azadirachta indica)was carried out by performing,mass(g)and solvent volume(mL ethanol and water)combinations,which were subjected to microwaves and ***,these extracts were evaluated in concentrations of 3%,4%and 5%.A survival analysis was performed for each of the *** respect to the results of the first bioassay,*** obtained a probability of survival of 0.476 on the seventh day,while in the second bioassay,*** obtained 0.488 survival *** suggests that the expected percentage of larvae that stay alive on the sixth day is 48.8%.However,in the evaluation of the neem extract the combination 1:12/70%to 4%caused 84% mortality of *** use of native HE and neem extracts has potential for the control of ***.

Adult emergence inhibition and adulticidal activities of medicinal plant extracts against Anopheles stephensi Liston

Objective:To determine the adult emergence inhibition（EI） and adulticidal activities of hexane,chloroform,ethyl acetate,and acetone leaves extracts of Anisomeles malabarica（A. malabarica）,Euphorbia hirta（E,hirta）,Ocimum basilicum（***）,Ricinus communis（R. communis）,Solanum trilobatum（***）,Tridax procumbens（***） and seeds of Gloriosa superba（***） against Anopheles stephensi（An,stephensi）.Methods:The EI and adulticidal trials were carried out according to World Health Organization（WHO） procedures with slight *** extracts were diluted in dimethyl sulphoxide in order to prepare a serial dilution of test dosages（15.625,31.25,62.5,125,250,500 and 1 000μg/mL）.Five duplicate trials were carried out for every sample concentration,and for each trial a negative control was included and the mortality was determined after 24 h of ***:The highest EI activity was found in ethyl acetate extracts of A malabarica,chloroform extracts of ***, ***,acetone of extract of ***,***,and seed extract of *** with EI50 values 143.12,119.***.157.87,139.39,111.19,and 134.85μg/mL,and the effective adulticidal activity was observed in chloroform,acetone extracts of ***,***,R. communis,*** and ethyl acetate extract of *** with LD50 values 120.17,108.77, 127.22,163.11,118.27,and 93.02μg/mL,***-square value was significant at P<0.05 ***:These results should encourage further efforts to investigate the compounds that might possess good EI and adulticidal properties when isolated in pure form.
