Dynamic transcriptome analysis of Bombyx mori embryonic development

Bombyx mori has been extensively studied but the gene expression control of its embryonic development is *** this study,we performed transcriptome profiling of six stages of *** embryonic development using RNA sequencing(RNA-seq).A total of 12894 transcripts were obtained from the *** these,12456 transcripts were shared among the six stages,namely,fertilized egg,blastoderm,germ-band,organogenesis,reversal period,and youth period *** were 111,48,41,54,77,and 107 transcripts specifically expressed during the six stages,*** analyzing weighted gene correlation networks and differently expressed genes,we found that during embryonic development,many genes related to DNA replication,transcription,protein synthesis,and epigenetic modifications were upregulated in the early *** of cuticle proteins,chitin synthesis-related proteins,and neuropeptides were more abundant in the late *** pathways of juvenile hormone and the ecdysteroid 20-hydroxyecdysone,and transcription factors were expressed throughout the embryonic development stages,more regulatory pathways were highly expressed around the organogenesis stage,suggesting more gene expression for *** results of RNA-seq were confirmed by quantitative real-time polymerase chain reaction of 16 genes of different *** acid methylation and seven sites in histone H3 modifications were confirmed by dot blot and western *** study increases the understanding of the molecular mechanisms of the embryonic developmental process and information on the regulation of *** development.

Harmful algal bloom(HAB)is an ecological disaster to local *** present,its impact on macrophytes has not been well *** this study,we cultivated sexually propagated embryos of S argassum fusiformis-an edible seaweed-in Prorocentrum donghaiense suspensions at different cell densities(0,0.50×10^(5),0.75×10^(5),1.00×10^(5),and 1.50×10^(5) cells/mL)for 10 days,during which growth and photosynthetic activities of the embryos were determined,and a monocultivation was set up for *** show that the relative growth rate and photosynthetic activities of the embryos co-cultivated with *** were inhibited mostly and significantly in the cell densities of 0.75×10^(5),1.00×10^(5),and 1.50×10^(5) cells/mL,and the inhibitory effects increased in overall with increased cell *** maximum relative electron transport rates(rETR max)and apparent photosynthetic efficiency(a)of co-cultivated embryos were all significantly lower than monocultivation ones on the 10 th ***,the photosynthetic activity detected by chlorophyll-a fluorescence transient(i.e.,OJIP),the electron transport among electron transfer accepters of PSII(photosystem II)and that from PSII to PSI(photosystem I)was restricted,which is probably responsible for the decreases of rETR max andain the co-cultivated *** addition,parts of the photosynthetic reaction centers of PSII in the co-cultivated embryos were ***,*** bloom could restrain the development and photosynthetic activities of *** embryos,reduce the seedlings stock,and eventually hinder the development of *** production industry.

The landscape of chromatin accessibility in skeletal muscle during embryonic development in pigs

Background:The development of skeletal muscle in pigs during the embryonic stage is precisely regulated by transcriptional mechanisms,which depend on chromatin ***,how chromatin accessibility plays a regulatory role during embryonic skeletal muscle development in pigs has not been *** gain insight into the landscape of chromatin accessibility and the associated genome-wide transcriptome during embryonic muscle development,we performed ATAC-seq and RNA-seq analyses of skeletal muscle from pig embryos at 45,70 and 100 days post coitus(dpc).Results:In total,21,638,35,447 and 60,181 unique regions(or peaks)were found across the embryos at 45 dpc(LW45),70 dpc(LW70)and 100 dpc(LW100),*** than 91%of the peaks were annotated within−1 kb to 100 bp of transcription start sites(TSSs).First,widespread increases in specific accessible chromatin regions(ACRs)from embryos at 45 to 100 dpc suggested that the regulatory mechanisms became increasingly complicated during embryonic ***,the findings from integrated ATAC-seq and RNA-seq analyses showed that not only the numbers but also the intensities of ACRs could control the expression of associated ***,the motif screening of stage-specific ACRs revealed some transcription factors that regulate muscle developmentrelated genes,such as MyoG,Mef2c,and *** potential transcriptional repressors,including E2F6,OTX2 and CTCF,were identified among the genes that exhibited different regulation trends between the ATAC-seq and RNA-seq ***:This work indicates that chromatin accessibility plays an important regulatory role in the embryonic muscle development of pigs and regulates the temporal and spatial expression patterns of key genes in muscle development by influencing the binding of transcription *** results contribute to a better understanding of the regulatory dynamics of genes involved in pig embryonic skeletal muscle development.

ATF6 aggravates apoptosis in early porcine embryonic development by regulating organelle homeostasis under high-temperature conditions

Activating transcription factor 6(ATF6),one of the three sensor proteins in the endoplasmic reticulum(ER),is an important regulator of ER stress-induced ***6 resides in the ER and,upon activation,is translocated to the Golgi apparatus,where it is cleaved by site-1 protease(S1P)to generate an amino-terminal cytoplasmic *** recent studies have made progress in elucidating the regulatory mechanisms of ATF6,its function during early porcine embryonic development under high-temperature(HT)stress remains *** this study,zygotes were divided into four groups:control,HT,HT+ATF6 knockdown,and HT+PF(S1P inhibitor).Results showed that HT exposure induced ER stress,which increased ATF6 protein expression and led to a decrease in the blastocyst ***,ATF6 expression was knocked down in HT embryos under microinjection of ATF6 double-stranded RNA(dsRNA).Results revealed that ATF6 knockdown(ATF6-KD)attenuated the increased expression of CHOP,an ER stress marker,and Ca2+release induced by *** addition,ATF6-KD alleviated homeostasis dysregulation among organelles caused by HT-induced ER stress,and further reduced Golgi apparatus and mitochondrial dysfunction in HT ***2 is an important downstream effector of *** showed that ATF6-KD reduced the occurrence of AIFM2-mediated embryonic apoptosis at *** together,our findings suggest that ATF6 is a crucial mediator of apoptosis during early porcine embryonic development,resulting from HT-induced ER stress and disruption of organelle homeostasis.

Effects of salinity on embryonic and larval development of Chinese mitten crab Eriocheir sinensis(Decapoda: Brachyura) and salinity-induced physiological changes

To investigate the effects of salinity on early development of Chinese mitten crab ( Eriocheir sinensis ), and the salinity tolerance mechanism of embryos, different developmental stages of embryos (gastrula, eyespot and pre-hatching stage), and hatched stage I zoea and megalopa, were exposed to a range of salinities (1, 5, 10, 15 (control), 20, 25, 30, 35 and 40). Hatching, survival and molting were monitored. Effects of 24-hour hypersaline (35) and hyposaline (1) stress on egg diameter, water content, Na +/K +-ATPase (NKA) activity, and crustacean hyperglycemic hormone (CHH) gene mRNA expression in embryos and megalopa, are reported. embryos are more tolerant of low (≤ 5) than high (≥25) salinities, with optimum ranges for gastrula and pre-hatching stage embryos being 5-20, and for eyespot embryo and stage I zoea, 10-20. Most megalopa can molt to the first juvenile instar by day 5 at salinities between 1 and 40, whereas molting of megalopa stages was delayed at 40. Hypersaline conditions resulted in a loss of moisture, reduction of egg volume, and a signifi cant increase in NKA activity and CHH mRNA expression at some developmental stages. Hyposaline conditions did not affect moisture content or egg volume, but resulted in decreased NKA activity and CHH mRNA expression in embryos. For megalopa stages, NKA activity was significantly upregulated following both hypo- and hypersaline stress. Our results suggest high salinity will inhibit development and hatching of E. sinensis embryos, and low salinity will affect the survival of their stage I zoea. Increased NKA activity in embryos and megalopa stages might indicate a hyporegulation response under hypersaline conditions. These findings provide useful information for spawning ground protection of indigenous E . sinensis and enrich the knowledge of embryonic tolerance mechanisms of hyperregulating crustaceans following osmotic stress.

Observations on the morphology of embryonic and larval development in Styela canopus Savigny

The morphological characters on different developmental phases of embryo and larva and the schedule of the whole early developmental process in Styela canopus were observed and described. The types of reproduction and early development as well as the morphology, of egg and larva in different ascidian species were compared. Styela canopus is oviparous. Its egg, 230.4 similar to 336.0 mum in diameter, is equipped with extraembryonic cell layers measured 43.2 similar to 63.0 mum thick. The early development of Styela canopus is typical urodele development, including fertilization, cleavage, gastrulation, tadpole in membrane, tadpole, initiating metamorphosis and juvenile. The tadpole of Styela canopus, with a length of 0.6 similar to 0. 9 mm, consists of trunk and tail. There are obvious notochord, ocellus and adhesive papillae in the tadpole. Under the water temperature of (25 +/- 0-5) degreesC and the salinity of 27.0, the larva was hatched after 9.5 similar to 11.0 h since the fertilization.

Comparing salinity tolerance in embryonic and larval development of two species of water strider, Aquarius paludum and Gerris latiabdominis （Hemiptera： Gerridae）

Water strider Aquarius paludum （Fabricius） is a cosmopolitan species colonizes mainly freshwater but occasionally brackish habitats throughout the Palearctic and Oriental regions. Water strider Gerris latiabdominis （Miyamoto） is a common species in Japan lives in temporary habitats as freshwater paddy fields. These two species often occur syntopically. We investigated differences in the developmental response to brackish water during embryonic and larval stages between the two species. Eggs were exposed to 0-1.8% NaC1 solutions within 24 h of oviposition. Larvae of G. latiabdominis were exposed to salinities of 0, 0.5%, and 0.9% from the first instar until adult emergence. Limits of NaC1 concentration for hatching were 1.3% and 1.0% for A. paludum and G. latiabdominis, respectively. The hatching rate of G. latiabdominis was lower than that ofA. paludum at salinities 〉0.9%. The period of embryonic development of G. latiabdominis was more prolonged than that ofA. paludum at a given salinity. Although the salinity tolerance of G. latiabdominis was lower than that ofA. paludum, our results suggest G. latiabdominis has the physiological capacity to expand into brackish waters. High and low salinity tolerances of A. paludum and G. latiabdominis, respectively, reflect the relatively wide range of habitat salinities utilized by A. paludum and the relatively restricted habitats preferred by G. latiabdominis. The high salinity tolerance ofA. paludum could be an important factor contributing to their cosmopolitan distribution because high tolerance to salinity means the possibility of them to be dispersed via ocean or sea to other continents and islands.

Development of setal patterns in embryonic and adult aphids of the Hormaphidinae （Hemiptera： Aphididae）

The elementary traits and diversity of embryonic setal patterns of 31 species of 22 genera, in three tribes of Hormaphidinae are studied. The embryonic setal patterns to every species are provided. Based on the parsimony principle and the developmental principle, these patterns are analyzed in each tribe following the terminology of Richards （1965）. It is suggested that the setal patterns of abdominal tergites are the most important characters in embryonic and adult setal patterns. In conclusion, neoteny is a phylogenetic trend in the Hormaphidinae. Nipponaphidini and Hormaphidini have more typical neoteny in each genus and more diversity in different types of adult viviparous females than Cerataphidini, by showing more frequent and various protopatterns or mesopatterns.

Effects of temperature and salinity on the development of embryos and larvae of the veined rapa whelk Rapana venosa(Valenciennes, 1846)

The major population of the veined rapa whelk Rapana venosa （Valenciennes, 1846）, which is an important fishery resource, is facing a large decline in China. We studied the effects of incubation temperature （16-34℃ at salinity 30） and salinity （5-45 at 25℃） on the incubation period and subsequent larval development. In the temperature experiment, the shortest incubation period was 12 days at 34℃, the lower temperature limit was 16~C, the longest mean shell length （1 193＋17 pan） occurred at 25℃ and the highest survival rate 72.28%＋5.62% was observed at 28℃. In the salinity experiment, the shortest incubation period was 15 days at 25. The salinity tolerance range was 15-40, the longest mean shell length （855-4-9 μm） and the highest survival rate 72.93%~4.85% were both observed at 35. This study demonstrated that, during the egg-mass stage, temperature and salinity regimes influence later growth and survival of larvae. These observations deepen our understanding of the ecology and conservation of natural populations of Rapana venosa.
