＜正＞ A series of novel curcumin analogs were synthesized and screened for anti - cancer activities both at Emory University and National Cancer Institute （NCI）. A majority of the analogs demonstrate a significant degree of anti - tumor activity. ef24 is the most active compound within the series. Follow - up studies showed that ef24 induces cell cycle arrest and apoptosis by means of a redox - dependent mechanism in MDA - MB - 231 human breast cancer cells and DU - 145 human prostate cancer cells. ef24 can serve as a Michael acceptor and react with nucle-ophiles such as glutathione（ gsh）. ef24 was treated with gsh/H2O to produce a colorless solution with cytotoxic properties comparable with ef24 alone. This suggested the in situ formation of an ef24 - gsh conjugate.which releases ef24 in a rapidly established equilibrium. Treatment of breast cancer cells separately with ef24 and ef24 - gsh revealed that the two compounds are almost equally efficacious in their cell - kill capacity. The ef24 - gsh and its congeners appear to represent a promising new series of stable and water - soluble anti - tumor pro - drugs.
