A me.cury biosensor was constructe. by inte.rating biosensor ge.e.ic e.e.e.ts into e. coli JM109 chromosome.in a single.copy numbe., using the.attP/attB re.ombination me.hanism of λ phage. The.ge.e.ic e.e.e.ts use. include.a re.ulatory prote.n ge.e.（me.R） along with ope.ator/promote. （O/P） de.ive. from the.me.cury re.istance.ope.on from pDU1358 plasmid of Se.ratia marce.ce.s. The.e.pre.sion of re.orte. ge.e.gfp is also controlle. by me.R/O/P. Inte.ration of the.construct into the.chromosome.was done.to incre.se.the.stability and pre.ision of the.biosensor. This biosensor could de.e.t Hg（Ⅱ） ions in the.conce.tration range.of 100–1700 nmol/L, and manife.t the.re.ult as the.e.pre.sion of GFP. The.GFP e.pre.sion was significantly diffe.e.t （P 0.05） for e.ch conce.tration of inducing Hg（Ⅱ） ions in the.de.e.tion range. which re.uce. the.chance. of misinte.pre.ation of re.ults. A mode. using re.re.sion me.hod was also de.ive. for the.quantification of the.conce.tration of Hg（Ⅱ） in wate. sample..
