Many diseases and health conditions are closely related to various microbes,which participate in complex interactions with diverse Drugs;nonetheless,the detailed targets of such Drugs remain to be elucidated.Many existing studies have reported causal associations among Drugs,gut microbes,or diseases,calling for a workflow to reveal their intricate interactions.In this study,we developed a systematic workflow comprising three modules to construct a Quorum Sensing-based Drug-Microbe-Disease(QSDMD)database(http://***/),which includes diverse interactions for more than 8,000 Drugs,163 microbes,and 42 common diseases.Potential interactions between microbes and more than 8,000 Drugs have been systematically studied by targeting microbial QS receptors combined with a docking-based virtual screening technique and in vitro experimental validations.Furthermore,we have constructed a QS-based drug-receptor interaction network,proposed a systematic framework including various drug-receptor-microbe-disease connections,and mapped a paradigmatic circular interaction network based on the QS-DMD,which can provide the underlying QS-based mechanisms for the reported causal associations.The QS-DMD will promote an understanding of personalized medicine and the development of potential therapies for diverse diseases.This work contributes to a paradigm for the construction of a molecule-receptor-microbe-disease interaction network for human health that may form one of the key knowledge maps of precision medicine in the future.

Type 2 diabetes mellitus(T2DM),more commonly known as chronic metabolic disease,is characterized by insufficient insulin secretion or insulin resistance.Due to the rise of T2DM globally,it is of great importance to decrease the number of complications,consequential morbidity,and mortality stemming from hyperglycemia.[1]The normal steady-state blood glucose level has become an important metric for T2DM prognosis and is recognized as one of the major results of drug therapies for diabetes.[2]This review outlines nine major types of antidiabetic Drugs,relevance to functional mechanisms,which could provide a reference for the research and development of T2DM treatment Drugs.

Capillary electrophoresis(CE)is widely used for the impurity profiling of Drugs that contain stereochemical centers in their structures,analysis of biomolecules,and characterization of biopharmaceuticals.Currently,CE is the method of choice for the analysis of foodstuffs and the determination of adulterants.This article discusses the general theory and instrumentation of CE as well as the classification of various CE techniques.It also presents an overview of research on the applications of different CE techniques in the impurity profiling of Drugs in the past decade.The review briefly presents a comparison between CE and liquid chromatography methods and highlights the strengths of CE using drug compounds as examples.This review will help scientists,fellow researchers,and students to understand the applications of CE techniques in the impurity profiling of Drugs.

阿司匹林激发试验是诊断非甾体抗炎药加重呼吸道疾病(non-steroidal anti-inflammatory Drugs-exacerbated respiratory disease,NERD)最常用的方法。根据给药途径不同,可分为鼻腔激发试验、支气管激发试验、口服激发试验及静脉激发试验,其中前3种激发试验在临床中常规应用。我国仅有极少数相关研究,尚未常规临床应用。本文就阿司匹林激发试验适应证及禁忌证、检查注意事项、操作步骤、评估方法、诊断标准、安全性等进行综述,以提高医务工作者对NERD的诊断水平。

Dear Editor,The Coronavirus disease 2019(COVID-19)pandemic is caused by severe acute respiratory syndrome coronavirus 2(SARS-CoV-2).The genome of this virus encodes two overlapping polyproteins,pp1a and pp1ab(Wu et al.,2020;Zhou et al.,2020).The functional polypeptides are released from those two polyproteins by extensive proteolytic events.

OBJECTIVES To investigate the prevalence of potentially inappropriate prescribing(PIP)for cardiovascular system(CVS)and antiplatelet/anticoagulant(AP/AC)Drugs among Korean elderly patients,using the Screening Tool of Older Persons’Prescriptions(STOPP)criteria version 2 and to identify the risk factors related to PIP.METHODS The 2016 National Aged Patient Sample data,comprising National Health Insurance claim records for a random sample of 20%of patients aged≥65 years,were used to calculate PIP prevalence of outpatient prescriptions.For criteria including drugdisease interactions,PIP prevalence per indication was estimated.RESULTS Among 1,274,148 elderly patients and 27,062,307 outpatient prescription claims,100,085 patients(7.85%)and 341,664 claims(1.27%)had one or more PIP.The most frequent PIP was“nonsteroidal antiinflammatory drug with concurrent antiplatelet agent(s)without protonpump inhibitor prophylaxis”in the claimlevel(0.97%)and patientlevel(6.33%)analyses.“Betablocker with bradycardia”(16.47%of claims)and“angiotensin receptor blockers in patients with hyperkalaemia”(23.89%of claims)showed the highest PIP prevalence per indication.Logistic regression analysis revealed that,among the patient and health care provider characteristics,female,older age,more severe comorbidities,polypharmacy,higher level of healthcare organization,and specialty of prescriber were significantly associated with a higher risk of PIP.CONCLUSIONS Our findings of a high prevalence of PIP for CVS and AP/AC Drugs among the elderly suggest that an effective strategy is urgently needed to improve the prescription practices of these Drugs.

Effective treatment for COVID-19 remains elusive,though urgently needed in the current pandemic.Repurposing marketed therapies may be an effective strategy for finding treatments quickly and recently,in vitro and clinical testing of such therapies against severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)has skyrocketed.However,not all marketed Drugs showing in vitro efficacy could achieve therapeutic concentrations in humans and discernment of Drugs that have favorable pharmacokinetic properties can save time and resources for future studies.
