Active tracking of rejected dried blood samples in a large program in Nigeria

AIM: To study the impact of rejection at different levels of health care by retrospectively reviewing records of dried blood spot samples received at the molecular laboratory for human immunodeficiency virus(HIV) early infant diagnosis(EID) between January 2008 and December ***: The specimen rejection rate, reasons for rejection and the impact of rejection at different levels of health care was examined. The extracted data were cleaned and checked for consistency and then deduplicated using the unique patient and clinic identifiers. The cleaned data were ciphered and exported to SPSS version 19(SPSS 2010 IBM Corp, New York, United States) for statistical ***: Sample rejection rate of 2.4%(n = 786/32552) and repeat rate of 8.8%(n = 69/786) were established. The mean age of infants presenting for first HIV molecular test among accepted valid samples was 17.83 wk(95%CI: 17.65-18.01) vs 20.30 wk(95%CI: 16.53-24.06) for repeated samples. HIV infection rate was 9.8% vs 15.9% for accepted and repeated samples. Compared to tertiary healthcare clinics, secondary and primary clinics had twofold and three-fold higher likelihood of sample rejection, respectively(P < 0.05). We observed a significant increase in sample rejection rate with increasing number of EID clinics(r = 0.893, P = 0.041). The major reasons for rejection were improper sample collection(26.3%), improper labeling(16.4%) and insufficient blood(14.8%). CONCLUSION: Programs should monitor pre-analytical variables and incorporate continuous quality improvement interventions to reduce errors associated with sample rejection and improve patient retention.

The Use of dried Blood Spot Samples in Screening Drugs of Abuse

The present article will provide an overview of use of dried blood sampling method for analysis of drug of abuse. Relatively short half life and instability in blood, calls for alternative sampling method for determination of drugs of abuse. dried blood spot (DBS) method has many advantages over the conventional sampling methods. The available method for DBS sample collection, storage and transport are described here. The techniques involved in and the factors that may influence the accuracy and reproducibility of the DBS methods for determination of drugs of abuse are presented. The DBS sampling has the potential to be a useful technique to detect drugs of abuse. The use of DBS for any drug should be judged against the potential error involved with the method.

Ochratoxin A in dried figs, raisings, apricots, dates on Iranian retail market

Ochratoxin A (OTA) is among the most important mycotoxins, and the International Agency for Research on Cancer (IARC) classifies it as possibly carcinogenic to humans (group 2B). A total of 121 samples of dried fruits from the central zone of Iran were analyzed for OTA by enzyme linked immunosorbent assay (ELISA) technique. The recovery percentages of OTA in spiked dried fruit samples at concentrations of 5, 10 and 20 ng/g were found to be 84.9%, 89.3% and 90.4% as mean, respectively. OTA was found in 20.7% of the analyzed samples by average concentration of 6.7 ± 3.9 ng/g. The incidence rates of OTA contamination in dried fig, raising, apricots, and date samples were 10.4%, 44.7%, 6.7% and 10.0%, respectively. The concentrations of OTA in 7.9% of contaminated dried raising samples and 2.1% of dried fig samples were higher than maximum tolerance limit accepted by European Union (10 ng/g). This value reflects that the analyzed samples have a minimal contribution to toxicological risk. To our best knowledge, the present study is the first report of the presence of OTA by ELISA in dried fruit samples in Iran.

Study on Sedation of dried Ginger

[Objective] The experiment aimed to explore sedation of dried ginger.[Method] By compared the experimental results before and after intragastric administration,the influences of decoction of dried ginger at different concentrations(1,2,3 mg/ml)on locomotor activities of white mice.[Result]When the decoction of dried ginger was 1 g/ml,the time of free movements and the number of raising forelimbs of mice were obviously detained(2.5 h after intragastric administration).When the decoction of dried ginger was 2 g/ml,the time of free movements and the number of raising forelimbs of mice were obviously influenced however the inhibition time is short than that of 1 g/*** the decoction of dried ginger was 3 g/ml,the inhibition is not obvious.[Conclusion] dried ginger had some sedation effect which was related to its dosage.

The most economically important disease of cultivated grapevines worldwide is powdery mildew(PM)caused by the ascomycete fungus Erysiphe *** majority of grapevine cultivars used for wine,table grape,and dried fruit production are derived from the Eurasian grape species Vitis vinifera because of its superior aroma and flavor ***,this species has little genetic resistance against *** meaning that grape production is highly dependent on the frequent use of *** integration of effective genetic resistance into cultivated grapevines would lead to significant financial and environmental benefits and represents a major challenge for viticultural industries and researchers *** review will outline the strategies being used to increase our understanding of the molecular basis of *** susceptibility to this fungal *** will summarize our current knowledge of different resistance loci/genes that have evolved in wild grapevine species to restrict PM infection and assess the potential application of these defense genes in the generation of PM-resistant grapevine ***,it addresses future research priorities which will be important in the rapid identification,evaluation,and deployment of new PM resistance genes which are capable of conferring effective and durable resistance in the vineyard.
