Metal-organic frameworks(MOFs)have a high specific surface area,adjustable pores and can be used to obtain functional porous materials with diverse and well-ordered structures through coordination and self-assembly,which has intrigued wide interest in a broad range of disciplines.In the arena of biomedical engineering,the functionalized modification of MOFs has produced drug carriers with excellent dispersion and functionalities such as target delivery and response release,with promising applications in bio-detection,disease therapy,tissue healing,and other areas.This review summa-rizes the present state of research on the functionalization of MOFs by physical binding or chemical cross-linking of small molecules.***.bioma romolecules.and hvdrogels and evaluates the role and approach of MOFs functionalization in boosting the reactivity of materials.On this basis,research on the application of functionalized MOFs composites in biomedical engineering fields such as drug delivery,tissue repair,disease treatment,bio-detection and imaging is surveyed,and the development trend and application prospects of functionalized MOFs as an important new class of biomedical materials in the biomedical field are anticipated,which may provide some inspiration and reference for further development of MOF for bio-medical applications.

Clustered regularly interspaced short palindromic repeat(CRISPR)has been gaining much attention in the modern medical field and has been widely used for the diagnosis and treatment of diseases in recent years.In this review,we will introduce the application of CRISPR in disease diagnosis and treatment,including its use in detecting pathogens,gene mutations,and genetic diseases,as well as its application in gene therapy for single-gene diseases,cancer,viral infectious diseases,and cardiovascular diseases.Additionally,we will discuss the potential future directions and challenges of CRISPR in the diagnosis and treatment of diseases,and provide a thorough overview of the ways in which CRISPR is used for diagnosing and treating diseases.

Recent advances in zwitterionic nanoscale drug delivery systems to overcome biological barriers

Nanoscale drug delivery systems(nDDS)have been employed widely in enhancing the therapeutic efficacy of drugs against diseases with reduced side effects.Although several nDDS have been successfully approved for clinical use up to now,biological barriers between the administration site and the target site hinder the wider clinical adoption of nDDS in disease treatment.Polyethylene glycol(PEG)-modification(or PEGylation)has been regarded as the gold standard for stabilising nDDS in complex biological environment.However,the accelerated blood clearance(ABC)of PEGylated nDDS after repeated injections becomes great challenges for their clinical applications.Zwitterionic polymer,a novel family of antifouling materials,have evolved as an alternative to PEG due to their super-hydrophilicity and biocompatibility.Zwitterionic nDDS could avoid the generation of ABC phenomenon and exhibit longer blood circulation time than the PEGylated analogues.More impressively,zwitterionic nDDS have recently been shown to overcome multiple biological barriers such as nonspecific organ distribution,pressure gradients,impermeable cell membranes and lysosomal degradation without the need of any complex chemical modifications.The realization of overcoming multiple biological barriers by zwitterionic nDDS may simplify the current overly complex design of nDDS,which could facilitate their better clinical translation.Herein,we summarise the recent progress of zwitterionic nDDS at overcoming various biological barriers and analyse their underlyingmechanisms.Finally,prospects and challenges are introduced to guide the rational design of zwitterionic nDDS for disease treatment.

Decades have passed since the first nanoparticles-base medicine was approved for human cancer treatment, and the research and development of nanoparticles for drug delivery are always undergoing.Nowadays, the significant advances complicate nanoparticles’ branches, including liposomes, solid lipid nanoparticles, inorganic nanoparticles, micelles, nanovaccines and nano-antibodies, etc. These nanoparticles show numerous capabilities in treatment and diagnosis of stubborn diseases like cancer and neurodegenerative diseases, emerging as novel drug carriers or therapeutic agents in future. In this review, the complicated branches of nanoparticles are classified and summarized, with their property and functions concluded. Besides, there are also some delivery strategies that make nanoparticles smarter and more efficient in drug delivery, and frontiers in these strategies are also summarized in this review. Except these excellent works in newly-produced drug delivery nanoparticles, some points of view and future expectations are made in the end.

Exosomes are membrane-bound nanoscale extracellular vesicles,which produced by almost all organisms.Due to the excellent biocompatibility,long circulation time as well as low immunogenicity,exosomes as naturally-derived drug delivery carriers have experienced explosive growth over the past decades.However,issues such as insufficient loading efficiency,heterogeneous delivery efficiency,uncontrollable targeting ability,and low production limit their wide application.Recently,the emerging exosome-liposome fusion strategy has become a potential approach to solve such issues.Thus,this review mainly focuses on the currently developed exosome-liposome fusion strategy and their application in drug delivery as well as disease treatment.This review aims to shed light on the advantages of fusion strategy in drug delivery and provides a better understanding for more rational design.The current challenge and future perspective regarding their clinical translation and application will also be discussed.

Flexible bioelectronics for physiological signals sensing and disease treatment

Flexible bioelectronics,including wearable and implantable electronics,have revolutionized the way of human-machine interaction due to the fact that they can provide natural and seamless interactions with humans and keep stable and durable at strained states.As sensor elements or biomimetic actuators,flexible bioelectronics can dynamically sense and monitor physiological signals,reveal real-time physical health information and provide timely precise stimulations or treatments.Thus,the flexible bioelectronics are playing increasingly important roles in human-health monitoring and disease treatment,which will significantly change the future of healthcare as well as our relationships with electronics.This review summarizes recent major progress in the development of flexible substrates or encapsulation materials,sensors,circuits and energy-autonomous powers toward digital healthcare monitoring,emphasizing its role in biomedical applications in vivo and problems in practical applications.A future perspective into the challenges and opportunities in emerging flexible bioelectronics designs for the next-generation healthcare monitoring systems is also presented.

Recent advances in the application of biomimetic nanomedicines in disease treatment

In recent years,nanomedicine plays an increasingly important role in improving the effectiveness,accuracy and safety of disease diagnosis and treatment.Happily,some of these nanoparticles are in clinical trials or have been approved by the Food and Drug Administration.With the deepening of research,people’s understanding of the interaction between nanomedicine and body system is increasing,and a new generation of biomimetic nanomedicine platforms has been developed to further improve the treatment effect of diseases.In this review,we will discuss recent advances in biomimetic nanosystems for the treatment of diseases,including cardio-cerebrovascular diseases,diabetes,and cancer.We hope that there will be more advanced biomimetic nanomedicines that can be widely used in the treatment of diseases in the future.

Metal nanozymes with multiple catalytic activities: regulating strategies and biological applications

Nanozymes with high enzyme-like catalytic activity have been recognized as promising alternatives for nature enzymes and exhibit great potentials in biodetection,disease diagnosis and treatment. Metal nanozyme is an important member of the nanozyme family. When metal nanozymes are controllably regulated by compositions,sizes, morphologies, crystal defects, surface states or stimulated by diversified external fields, they will acquire unique geometric and electronic structures and exhibit various physical/chemical properties in catalytic behaviors.Although significant progress has been made in metal nanozymes, there is still large space for further improving the catalytic efficiency, developing the synthetic process for large-scale production, facilitating the clinical transformation and exploring the untapped applications.Directing at these issues, rational design of novel nanozymes for better applications and deeper understanding of catalytic mechanisms are urgently required. Accordingly,this review focuses on the recent advances on regulating strategies, catalytic mechanisms and relevant biological applications of metal nanozymes with multiple enzymelike performance. Furthermore, future perspectives are outlooked to inspire more attention and efforts on this interesting multi-disciplinary field.

As a kind of nature-derived bioactive materials, polyphenol-based hydrogels possess many unique and outstanding properties such as adhesion, toughness, and self-healing due to their specific crosslinking structures, which have been widely used in biomedical fields including wound healing, antitumor, treatment of motor system injury, digestive system disease, oculopathy, and bioelectronics. In this review, starting with the classi-fication of common polyphenol-based hydrogels, the pyramid evolution process of polyphenol-based hydrogels from crosslinking structures to derived properties and then to biomedical applications is elaborated, as well as the efficient reverse design considerations of polyphenol-based hydrogel systems are proposed. Finally, the existing problems and development prospects of these hydrogel materials are discussed. It is hoped that the unique perspective of the review can promote further innovation and breakthroughs of polyphenol-based hydrogels in the future.
