Drying Drop Technology as a Possible Tool for Detection Leukemia and Tuberculosis in Cattle

Usual response of organism to viral or bacterial invasion represents antibodies production, qualitative and quantitative changes in composition of biological fluids. These changes influence conformation and surface characteristics of macromolecules (proteins), which become apparent in sessile drying drops, when they form aggregates due to salting-out effect and sediment. The bottom adsorption layers change their adhesive and viscoelastic properties in time depending on fluid composition and structure. The aim of this study was verification the idea of using this phenomenon in rapid vet diagnostics. Milk, blood and serum samples of 183 cows were tested using Drop Drying Technology (DDT). A drop of tested fluid dried on a polished quartz plate, oscillated with constant frequency—60 kHz. Mechanical properties of the drop changed during drying, influenced the electrical conductivity of the quartz plate. This signal was converted to the Acoustical- Mechanical Impedance (AMI) and displayed as a curve in coordinates AMI vs. Time. Shape of the AMI curve reflected this dynamics, and was used as a target for quantitative comparison between control and infected animals. Frequency analysis of the estimated parameters of the curves was performed using features of the Excel program. Powerful method of artificial neural network processing of the experimental data was also tested in this work as a possible tool for future development. Significant differences between control, Bovine leucosis virus positive (BLV+) and Bovine tuberculin positive (BTub+) cattle groups were obtained using all biological fluids—blood, serum and milk. We fixed also a season shift of the data, but distinction between groups still remained. In serum and milk some features of the AMI curves were more stable, and retained diagnostic properties when combined winter and spring databases. Further development of DDT is proposed.

Hepatic hydrothorax:An update and review of the literature

This review considers the modern concepts of pathogenesis,diagnostic methods,and treatment principles of hepatic hydrothorax(HH).HH is the excessive(>500 mL)accumulation of transudate in the pleural cavity in patients with decompensated liver cirrhosis but without cardiopulmonary and pleural *** causes respiratory failure which aggravates the clinical course of liver cirrhosis,and the emergence of spontaneous bacterial pleural empyema may be the cause of *** information was collected from the PubM ed database,the Google Scholar retrieval system,the Cochrane reviews,and the reference lists from relevant publications for 1994-2016 using the keywords:"liver cirrhosis","portal hypertension","hepatic hydrothorax","pathogenesis","diagnostics",and"treatment".To limit the scope of this review,only articles dealing with uncomplicated hydrothorax in patients with liver cirrhosis were *** analysis of the data showed that despite the progress of modern hepatology,the presence of HH is associated with poor prognosis and high *** patients suffering from it are candidates for orthotopic liver *** routine clinical practice,stratification of the risk for an adverse outcome and the subsequent determination of individual therapeutic strategies may be the keys to the successful management of the patient’s *** development of pathogenetic pharmacotherapy and optimization of minimally invasive treatment will improve the quality of life and increase the survival rate among patients with HH.

Simultaneous Detection of Colletotrichum acutatumand C. gloeosporioidesfrom Quiescently Infected Strawberry Foliage by Real-Time PCR Based on High Resolution Melt Curve Analysis

Anthracnose of strawberry, caused primarily by the fungal pathogens belonging to Colletotrichum acutatum species complex (CASC) and C. gloeosporioides species complex (CGSC) is an economically important disease in the Southeast United States. Quiescently infected (QI) planting stock is one of the most important sources of inoculum in the fruiting field that can only be reliably detected by highly sensitive real time quantitative PCR (q-PCR) assay. In this study, a q-PCR assay was developed and optimized that can discriminate anthracnose fruit rot (AFR) and anthracnose crown rot (ACR) causing species based on the difference in post PCR melting temperatures of amplicons. Controlled environment grown plants artificially inoculated with different levels of CASC and CGSC showed a significant (P 0.001) correlation with levels of quantification expressed by Ct values in q-PCR from petioles and leaf blades. The leaf blade was a significantly larger reservoir of QI than that of the petiole. Both TaqMan and SYBR Green assay showed similar sensitivity and specificity. Detection of QI on leaves at young middle and older stages from inoculation with same number of conidia indicated that middle aged leaves were the best for assessing QI. Quantification of QI from middle aged leaf samples from a strawberry fruiting field that has been planted with pre-inoculated plants at both ends of rows and let inoculum spread showed higher sensitivity and precision by q-PCR compared to that of a traditional paraquat assay. The assay developed and validated in this study offers a new tool for evaluating planting stocks for QI to make decision on preventative control for strawberry anthracnose.

Possibility of Use of Noise Analysis for Identification of Reactor Conditions during Accidents

Knowledge and control of the thermo-hydraulic conditions in the reactor is required for the effective accident management. Dedicated and qualified for harsh environment instrumentation has to be in place for this purpose. Experience of the Fukushima Dai-ichi plant and the lessons learned from the European stress tests demonstrated that alternative and divers tools and methods are needed for the identification of reactor condition in extreme situations. In the paper the feasibility of development of an alternative accident monitoring via well-known noise diagnostics methods is proposed and demonstrated. The possibility of identification of reactor accident conditions using temperature and pressure fluctuations, noise of the neutron and gamma field is considered on the basis of research achievements in reactor noise. As an example the use of pressure fluctuations for accident monitoring is presented.

A Self-Learning Diagnosis Algorithm Based on Data Clustering

The article describes an approach to building a self-learning diagnostic algorithm. The self-learning algorithm creates models of the object under consideration. The models are formed periodically through a certain time period. The model includes a set of functions that can describe whole object, or a part of the object, or a specified functionality of the object. Thus, information about fault location can be obtained. During operation of the object the algorithm collects data received from sensors. Then the algorithm creates samples related to steady state operation. Clustering of those samples is used for the functions definition. Values of the functions in the centers of clusters are stored in the computer’s memory. To illustrate the considered approach, its application to the diagnosis of turbomachines is described.

Pediatric radiological diagnostic procedures in cases of suspected child abuse

Advanced and specialized radiological diagnostic procedures are essential in cases of clinically diagnosed injuries to the head, thorax, abdomen or extremities of a child, especially if there is no case history or if the reporting of an inadequate trauma suggests battered child syndrome. In particular, these diagnostic procedures should aim at detecting lesions of the central nervous system (CNS), so that the treatment can be immediately initiated. If the diagnostic imaging reveals findings typically associated with child abuse, accurate documentation constituting evidence, which will stand up in court, is required to prevent any further endangerment of the child’s welfare.

Different Patterns of Fluorescence in the Quantisation of anti-Leishmania Infantum Antibodies by Indirect Immuno-Fluorescent Antibody Test (IFAT) in the Serological Diagnosis of Canine Leishmaniasis

Although Indirect Immuno-Fluorescent Antibody Test (IFAT), performed employing “in house” prepared antigen, is considered by several authors as the golden standard for the quantisation of anti-leishmania antibodies in dogs, there is a lack of papers reporting a description of the different patterns of fluorescence that can be observed. An incorrect identification of patterns of fluorescence may be an important source of bias in the interpretation of results. Previous papers report different criteria to define as “positive” a specific pattern of fluorescence, namely: membrane fluorescence, homogeneous fluorescence of the body, or homogeneous fluorescence of the body plus flagellum. In this paper, we report a detailed description of preparation of slides and of the patterns of fluorescence that can be obtained employing “in house” prepared antigen. At least six main patterns of fluorescence may be observed: 1): homogeneous cytoplasmatic green fluorescence;2): membrane pattern, in which the fluorescence is mainly localized along the entire perimeter of the parasites;3): coarse-speckled cytoplasmatic fluorescence;4): flagellar pattern, in which the fluorescence is localized exclusively onto the flagellum;5): punctiform pattern, in which the fluorescence is localized exclusively at the basis of the flagellum;6): nuclear pattern, in which only the nucleus of the parasite shows a homogeneous green fluorescent. The significance of each pattern is discussed.

X-Ray and VUV Spectra from the Laser Plasma Produced with “Kanal-2” Facility

The paper presents experimental results obtained on “Kanal-2” facility. Laser radiation focusing on the surface of plane magnesium targets created the high temperature plasma, which emitted X-ray and vacuum ultraviolet (VUV) radiation. This radiation spectrum was investigated with two spectrographs: the mica crystal spectrograph (working range 8.2 ? - 9.6 ?) and the grazing incidence VUV spectrograph (working rage 30 ? - 130 ?). A set of beryllium stepwise attenuators appended the diagnostic complex and allowed us to get an approximated picture of a continuous spectrum within the range of 2.2 ? - 6.2 ?. The estimation of the plasma electron temperature Te from the ratio between the intensity of the dielectronic satellites and the resonance line gives Te ~ 180 eV. The ratio between the intensity of the resonance and intercombination lines gives the electron density of the emitting zone ne ~ 2 × 1019 cm-3. Some lines observed within the spectral range of 8.5 ? - 9.1 ? belong to none of the transitions of Mg ions. Perhaps the observed spectrum is determined by the transitions in so-called hollow ions of Mg, i.e. in the ions with unfilled inner shells. The spectra obtained with the grazing incidence spectrograph and with the minimum-directioned discrepancy iteration method of spectrum reconstruction from the attenuation curve in the beryllium stepwise attenuators are also presented.

Flow Cytometric Detection of Circulating Tumor Cells in Breast Cancer Patients: A Blinded Study

Circulating tumor cells are cells that detach from the primary tumor site and migrate to the bone marrow or other tissues where they can initiate a metastatic site. Liquid biopsies are an emerging tool in the past decades that enables us to detect Circulating Tumor Cells in patients’ blood. Flow cytometry is a powerful tool used in liquid biopsy diagnostics. This aims to prove the sensitivity and specificity of a flow cytometric panel for the detection of CTCs in breast cancer patients using healthy individuals’ samples as controls. The study was blinded to the data analyzing researcher. Statistical analysis followed and results show 86.9% area under the curve which indicates that the particular method can be very promising for diagnosing breast cancer.

Fast and Accurate Identification of M. tuberculosisComplex Using an Immunochromatographic MPT64 Antigen Detection Test

Background: A new rapid Immunochromatographic test (ICT) kit (MPT64 TB Ag Kit) for detection of MPT64 Antigen in M. tuberculosis (MTB) isolates used for rapid identification of MTB isolates developed by SD (Standard diagnostics) Bio line, South Korea was evaluated. The ICT is a rapid, reliable and cheaper method that can be used instead of conventional biochemical tests for confirming MTB in culture isolates in resource limited laboratories. The study also evaluated the ability of ICT to detect MPT64-Antigen before the micro MGIT could signal positive. Material/Methods: A total of 450 sputum samples of individual patients were used for the study. 152 isolates of Mycobacteria were recovered from solid and liquid media. These strains were tested for the detection of MPT64-antigen. H37Rv strain was served as the positive reference control and also used for early detection of Antigen experiment. Findings: The development of bands on both test and sample region when H37Rv strain was tested were seen (MPT64 antigen positive). When 138 MTB isolates were tested, it showed a similar banding pattern indicating 100% sensitivity. MPT64 band formation was not detected in any of the 14 isolates indicating 100% specificity. Both PPV & NPV were 100%. All the isolates negative for MPT64 Ag were confirmed as MOTT by conventional bio-chemical PNBA. The H37Rv strain showed a faint band from the 2nd day onwards from inoculation till 3rd day in the earlier Antigen detection experiment. Conclusion: Rapid identification of MTB culture isolate is a pressing need for diagnosis and proceeding to perform drug susceptibility testing. MPT64 TB Ag detection ICT kit is a rapid, reliable method, good substitute for molecular identification methods, and conventional biochemical test which is time-consuming and technically demanding. The early detection of Antigen can be used as an effective tool in diagnosis.
