Updated roles of the gut microbiota in exploring shrimp etiology, polymicrobial pathogens, and disease incidence

Litopenaeus vannamei is the most extensively cultured shrimp species globally,recognized for its scale,production,and economic ***,its aquaculture is plagued by frequent disease outbreaks,resulting in rapid and massive *** research often lags behind the emergence of new diseases,leaving the causal agents of some shrimp diseases unidentified and leading to nomenclature based on symptomatic presentations,especially in cases involving co-and polymicrobial *** data on shrimp disease statuses remain *** this review,we summarize current knowledge on shrimp diseases and their effects on the gut ***,we also propose a workflow integrating primary colonizers,“driver”taxa in gut networks from healthy to diseased states,disease-discriminatory taxa,and virulence genes to identify potential polymicrobial *** examine both abiotic and biotic factors(e.g.,external and internal sources and specific-disease effects)that influence shrimp gut microbiota,with an emphasis on the“holobiome”concept and common features of gut microbiota response to diverse *** excluding the effects of confounding factors,we provide a diagnosis model for quantitatively predicting shrimp disease incidence using disease common-discriminatory taxa,irrespective of the causal *** to the conservation of functional genes used in designing specific primers,we propose a practical strategy applying qPCR-assayed abundances of disease common-discriminatory functional *** review updates the roles of the gut microbiota in exploring shrimp etiology,polymicrobial pathogens,and disease incidence,offering a refined perspective for advancing shrimp aquaculture health management.

Integrated analysis of human influenza A(H1N1)virus infectionrelated genes to construct a suitable diagnostic model

The genome characteristics and structural functions of coding proteins correlate with the genetic diversity of the H1N1 virus,which aids in the understanding of its underlying pathogenic *** this study,analyses of the characteristic of the H1N1 virus infection-related genes,their biological functions,and infection-related reversal drugs were ***,we used multi-dimensional bioinformatics analysis to identify the key genes and then used these to construct a diagnostic model for the H1N1 virus *** was a total of 169 differently expressed genes in the samples between 21 h before infection and 77 h after *** were used during the protein-protein interaction(PPI)analysis,and we obtained a total of 1725 interacting ***,we performed a weighted gene co-expression network analysis(WGCNA)on these genes,and we identified three modules that showed significant potential for the diagnosis of the H1N1 virus *** modules contained 60 genes,and they were used to construct this diagnostic model,which showed an effective prediction ***,these 60 genes were involved in the biological functions of this infectious virus,like the cellular response to type I interferon and in the negative regulation of the viral life ***,20 genes showed an upregulated expression as the infection *** 36 upregulated genes were used to examine the relationship between genes,human influenza A virus,and infection-related reversal *** study revealed numerous important reversal drug molecules on the H1N1 *** included rimantadine,interferons,and shikimic *** study provided a novel method to analyze the characteristic of different genes and explore their corresponding biological function during the infection caused by the H1N1 *** diagnostic model,which comprises 60 genes,shows that a significant predictive value can be the potential biomarker for the diagnosis of the H1N1 virus infection.

Aim: Esophageal cancer(EC)is one of the common malignant tumors in the world, and its incidence and mortality are relatively high. Global cancer statistics released in 2020 showed that EC has a higher incidence in East Asia, especially in China, and males are significantly higher than females. The number of new cases of EC in China in 2020 was about 324,422, accounting for about 50% of the world;the number of deaths was 301,135, accounting for about 55% of the world. According to the clinical histopathological type, EC was mainly divided into esophageal adenocarcinoma(EAC) and esophageal squamous cell carcinoma(ESCC). Importantly, the incidence of ESCC in China accounts for more than 70% of the world. The onset of ESCC was insidious and there were no obvious clinical symptoms in the early stage. At present, the pathogenesis of ESCC was not clear, and there was lack of effective treatment measures for advanced ESCC patients. Therefore, finding a noninvasive diagnostic method for ESCC of early stage is crucial to improve the 5-year survival rate of *** years, many studies have found that autoantibodies against tumor-associated antigen(TAAbs)can be used as biomarkers in the diagnosis of esophageal squamous cell carcinoma(ESCC). The aim of this study is to screen novel TAAbs through proteomic chip and constructing a diagnostic model to discriminate ESCC cases from benign esophageal diseases(BED) cases and healthy controls(HCs).Methods: HuProt human proteomic chip was purchased from BCBIO Biotechnology(Guangzhou,China). The chip is currently the world’s largest collection of full-length human proteins available. It contains more than 21,000 recombinant proteins and covers all human recombinant proteins that can be purified, covering 81% of the human proteome. The human proteomic chip was used to screen TAAbs. Enzyme-linked immunosorbent assay(ELISA) was used to verify and validate the candidate TAAbs screened by the chip in cohort 1(90 ESCC cases and 90 HCs)

Aim:This study was designed to construct a salivary protein fingerprint model used for early diagnosis of breast cancer by detecting salivary proteins of breast cancer patients and screening specific protein ***:Surface-enhanced laser desorption ionization time-of-flight mass spectrometry（SELDI-TOF-MS） technique was applied to determine the protein mass spectrums of 91 cases of saliva samples（among which 47 cases were breast cancer patients and 44 cases were healthy subjects） to establish a diagnosis model of breast ***:311 protein peaks were detected in the samples of breast cancer patients and healthy control,and 32 protein peaks had statistical significance difference between the two groups.A template consisting of 4 proteins with molecular weights of 4849.31,5224.96,3439.02 and 3559.89 Da respectively was *** could be correctly grouped to breast cancer patients and healthy *** 47 cases of breast cancer,37 cases were correctly diagnosed;34 cases of healthy subjects were correctly *** sensitivity was 78.7%（37/47）;the specificity was 77.2%（34/44）.Conclusions:the salivary protein fingerprint model of breast cancer constructed by SELDI-TOF-MS technique provided a new method with strong specificity and high sensitivity for early screening and diagnosis of breast cancer;and it was worth further research and application.
