Effect of type 2 diabetic mellitus in the prognosis of acute-onchronic liver failure patients in China

Prevalence of Drug Resistant Uropathogenic Escherichia coli from Immunocompromised diabetic Patients Attending Selected Health Facilities in Benue State

Escherichia coli is the commonest bacterial uropathogen of UTIs, the commonest infections in immunocompromised diabetic patients. Better understanding of their main resistance mechanisms to commonly used antibacterial agents will help to reduce the burden of this infection. The prevalence of drug resistant uropathogenic Escherichia coli isolates from immunocompromised diabetic patients attending selected health facilities in Benue State was investigated. Two hundred and ninety-six midstream urine samples were collected for both study and control diabetic patients. Bacterial isolation was done using semi-quantitative method. Drug resistant Escherichia coli were identified as multidrug resistant (MDR), extensive drug resistant (XDR) and pan-drug resistant organisms (PDR). Statistical significance was considered at p E. coli isolates from the study and control subjects with overall prevalence of 20.9% and 8.4% respectively. The isolates were highly resistant to penicillin (ampicillin), monobactam (aztreonam), older quinolone (nalidixic acid) whereas the majority of them showed high susceptibility to aminoglycoside (streptomycin), cephalosporin (cefotaxime) and carbapenem (imipenem). None showed complete susceptibility to all the tested antibiotics. Twenty-five E. coli were identified in this MDR, eight, XDR while 5 were PDR. High numbers of drug resistant E. coli isolates were identified in the study group of which 25 were MDR, 8 XDR while 5 were PDR isolates. High prevalence of UTI and drug resistant isolates occur in diabetic patients with hyperglycemic condition.

Aim:To explore the effects and mechanisms of caffeine and coffee on bladderdysfunction in streptozotocin-induced diabetic ***:Sprague-Dawleymale rats were divided randomly into 4 groups:control,diabetes mellitus(DM),DM with coffee treatment,and DM with caffeine *** diabetic rat wasinduced by intraperitoneal injection of streptozotocin(60 mg/kg).After 7 weeks oftreatment with coffee and caffeine,cystometrogram,contractile responses to elec-trical field stimulation(EFS)and acetylcholine(ACh),and cyclic AMP(cAMP)concentration of the bladder body and base were ***:The bladderweight,volume threshold for micturition and post-void residual volume(PVR)inthe diabetic rats were significantly higher compared to those in the control *** or caffeine treatment significantly reduced the bladder weight,bladdercapacity and PVR in the diabetic *** caused significant decreases in cAMPconcentration of the bladder and coffee and caffeine caused upregulation of cAMPcontent in the diabetic *** addition,coffee and caffeine tended to normal-ize the altered detrusor contractile responses to EFS and ACh in the diabetic ***:These results indicate that caffeine and coffee may have beneficialeffects on bladder dysfunction in the early stage of diabetes by increasing cAMPcontent in the lower urinary tract,recovering the micturition reflex and improvingthe detrusor contractility.

Luteolin attenuates neuronal apoptosis in the hippocampi of diabetic encephalopathy rats

Luteolin （3＇,4＇,5,7-tetrahydroxyflavone） has powerful anti-apoptotic and antioxidant properties. This study aimed to investigate the effects of luteolin on hyperglycemia-mediated apoptosis in the hippocampi of rats with streptozotocin-induced diabetic encephalopathy after injection into the tail veins, and the molecular mechanisms involved. Biochemistry and terminal deoxynucleotidyl transferase mediated dUTP nick end labelling detection results showed that luteolin treatment （given twice daily for 15 days） significantly inhibited hyperglycemia-mediated apoptosis, decreased malondialdehyde levels and increased glutathione levels in the hippocampi of streptozotocin- induced diabetic rats. Western blot analysis revealed that luteolin also inhibited the expression of apoptosis-related factors and cytochrome c release from mitochondria. Luteolin also improved the learning and memory abiJities of rats with diabetic encephalopathy in a water maze test. Further western blot analysis revealed that luteolin treatment facilitated neuronal cell survival through activation of the phosphatidylinositol 3-kinase/Akt signaling pathway, an extracellular signal pathway involved in the suppression of cell apoptosis and promotion of cell survival. These experimental findings indicate that luteolin can inhibit apoptosis of hippocampal nerve cells in rats with diabetic encephalopathy, and that this effect is mediated by an indirect antioxidative effect, the inhibition of activation of apoptosis-related factors and the activation of phosphatidylinositol 3-kinase/Akt signal pathway.

BACKGROUND： Protein nonenzymatic glycosylation is supposed to be one of mechanisms for chronic complications development in diabetes mellitus, and therefore, might play an important role in the neuronal degeneration. OBJECTIVE： To study the protein nonenzymatic glycosylation in brain neurons of diabetic rats, and to analyze the pathway of neuronal degeneration at the early stage of hyperglymecia. DESIGN： Randomized controlled animal experiment. SETTING： Department of Endocrinology, First hospital Affiliated to General Hospital of Chinese PLA and Beijing Laboratory for Brain Aging, Xuanwu Hospital Affiliated to Capital Medical University. MATERIALS： Thirty-five male Wistar rats （grade Ⅱ ）, aged 3 months old, and 11 male purebred Kunming mice （grade Ⅲ） without special pathogen, aged 3 months old, were provided by the Animal Room of Capital Medical University. METHODS： This experiment was carried out in the Beijing Laboratory for Brain Aging, Xuanwu Hospital Affiliated to Capital Medical University in 1998. The rats in the diabetic model group were intraperitoneally injected into 10 g/L STZ according to 60 mg/kg to establish rat models of diabetes mellitus. The blood glucose and body mass of rats in each group were determined respectively at 1, 2 and 3 months after modeling. Lipoxin A4 improves erectile dysfunction in rats with type I diabetes by inhibiting oxidative stress and corporal fibrosis

Previous studies have shown that oxidative stress and corporal fibrosis in penile tissues of rats were key pathological factors of erectile dysfunction induced by diabetic mellitus （DMED）. Lipoxin A4 （LXA4） was reported to inhibit oxidative stress and fibrosis diseases, while whether it could exert a protective role on erectile function was not clear. Type I diabetic mellitus （DM） was induced in thirty male lO-week-old Sprague-Dawley rats using streptozotocin. Ten weeks later, twenty-two rats with DMED confirmed by an apomorphine test were divided into two groups： the DMED group （n = 11） and the DMED ＋ LXA4 group （n = 11; LXA4 injection daily for 4 weeks）. In addition, another ten age-matched rats formed the Control group. We found that erectile function was significantly impaired in the DMED group compared with the Control group, but was improved in the DMED ＋ LXA4 group. Similarly, the over-activated oxidative stress and impaired endothelial function in the DMED group were both improved in the DMED ＋ LXA4 group. Moreover, the DMED group showed serious corporal fibrosis, which was also inhibited by the treatment of LXA4 in the DMED ＋ LXA4 group. Taken together, LXA4 could exert an inhibition role on oxidative stress and fibrosis to improve DMED effectively.

Hypoglycemic and Hypolipidemic Effect of Bitter Kola (<i>Garcinia kola</i>) Seed Extract on Alloxan-Induced diabetic Albino Rats

The treatment and management of diabetes mellitus has been a challenge to achieve a total cure using conventional drugs. Hypoglycemic and hypolipidemic effects of bitter kola (Garcinia kola) seed extract on alloxan-induced diabetic rats were studied as a local product. Albino rats weighing about 200 g were investigated in groups using 200 - 800 mg/kg weight of bitter kola seed extract and the sample examined for glucose and lipid profiles using enzymatic methods and statistically analyzed using statistical package for social science (SPSS) windows, version 20.0. The results were expressed as the Mean ± SD. The results for the serum glucose and low density lipoprotein cholesterols (LDL-C) concentrations of untreated diabetic rats, and treated rats showed a significant (P diabetic rats, and treated rats showed increase in concentration which was not significant (P > 0.05) from 200 - 800 mg/kg weight when compared with the controls. High density lipoproteins (HDL-C) concentrations for untreated diabetic rats, and treated rats showed increase in concentration which was significant (P Garcinia kola) seed extract.
