DIABETIC INTERSTITIAL GLUCOSE IN THE SKIN TISSUE BY ATR-FTIR SPECTROSCOPY VERSUS CAPILLARY BLOOD GLUCOSE

Recently introduced horizontal attenuated total reflectance(HATR)Fourier transform infrared(FTIR)spectroscopy for real-time assessment and continuous monitoring of glucose biomolecules in the skin tissue directly on the patients might appear a promising alternative to interpret the activity of interstitial glucose metabolism in vivo by means of evaluating the dynamics of changes of glucose concentrations in interstitial fluid(IF).In the present study,in vivo spectra by ATR-FTIR spectroscopy were obtained post-prandially during a 120–180-minute continuous monitoring in three patients with type 2 diabetes.and compared to pre-prandial *** all patients with diabetes.interstitial glucose levels at 1030 and 1041cm^(−1) reflected the best relationship with blood *** lag time(LT)required for glucose to diffuse from the capillary to epidermal skin tissue was calculated between 0 and 60 minutes at all measured glucose *** showed intra-and inter-subject variations of each glucose biomolecule,pointing to similarities and differences among interstitial glucose metabolism of the ***,the findings suggest that HATR-FTIR spectroscopy might have the potential for clinical interpretation of activity of glucose metabolism for diagnosis,management,and treatment of patients with diabetes.

Casearia sylvestris Improved Cutaneous Burn Repair in Diabetic Rats

Objectives： The burn repair associated with diabetes.complications showed considerable changes in the healing process and the development of alternative therapies to favor the repair is important. This study investigated the efficacy of the Casearia sylvestris in the burn repair in diabetic and non-diabetic rats. Methods： The animals were divided into four groups （n = 20）：（C） non-diabetics treated with carbopol gel; （G） non-diabetic treated with C. sylvestris extract in carbopol gel; （DM-C） diabetics treated with carbopol gel; （DM-G） diabetics treated with C. sylvestris extract in carbopol gel. Burn was induced using a metal plate （2 cm diameter/120 °C/20 sec）. Samples were collected on the 3rd, 7th, 14th and 21st days after the injury for histomorphometric analysis （inflammatory infiltrate, fibroblasts, blood vessels and collagen）, MPO （Myeloperoxidase） and NAG （N-Acetilglicosaminidase） dosage, hydroxyproline, glycosaminoglycans, TGF-β1, VEGF, collagen I and III. Key findings： C. sylvestris decreased the inflammatory process during the experimental period in diabetic and non-diabetic animals which has been demonstrated by MPO, NAG quantification, and TGF-β1 expression. C. sylvestris also increased blood vessels and decreased VEGF expression during the studied period in non-diabetic animals. Fibroplasia and collagenase were increased in all experimental periods in the DM-G group by means of de hydroxyproline quantification, collagen I, collagen III and glycosaminoglycans. Conclusions： C. sylvestris application can modulate favorably the time of inflammation and collagenous process in the repair of burn injuries in diabetic rats.

Immune Reactive Ezrin Surface Area Increases in Glomerular Podocytes of STZ Diabetic Rats Precede Their Detachment, Is Prevented by Phlorizin But Not by Insulin

Glomerular tuft immune reactive Ezrin surface area (EzA) and fraction of EzA to total glomerular tuft area significantly increased, indicating podocyte growth, rounding and altered cytoskeletal interactions at 1 week of STZ diabetes. Podocyte number per glomerulus (WT1+ nuclei) did not change indicating no detachment, but density decreased due to tuft hypertrophy. Treatment with PLZ or Insulin for one week, prevented increase in proteinuria and hyperglycemia but not the decrease in podocyte density. PLZ but not Insulin prevented increase in ezrin positive area in glomeruli and per podocyte. In podocytes in culture neither 25 mM glucose with or without PLZ (2.5 or 25 uM) altered Ezrin expression measured in western blots. In summary, the Ezrin positive glomerular surface area increase seen after 1 week of STZ diabetes. reflects altered podocyte morphology and cytoskeletal interactions, prevented by PLZ but not by insulin. Ezrin area increase preceded podocyte detachment and in podocytes in culture is not associated with increases in podocyte Ezrin protein expression. It is a likely precursor of shape changes in podocytes and of alterd interactions with basement membrane that contribute to detachment and thickening. Glomerular capillary tuft hypertrophy and reduced podocyte density persisted despite PLZ or insulin treatments, independently of levels of glycemia and of proteinuria.

Permeation of vanadium(III,IV,V)-dipicolinate complexes across MDCK cell monolayer and comparison with Caco-2 cells

The permeation and cytotoxicity of three insu-lin-mimetic vanadium(III, IV, V)-dipicolinate complexes were studied using the MDCK cell monolayer in comparison with the Caco-2 cells. On MDCK cell monolayer, the appar-ent permeation coefficients (Papp) were estimated to be (7.5 ± 1.0)×10?6, (1.0 ± 0.2)×10?6, (1.7±0.4)×10?6 cm/s for V(V), V(IV), and V(III)-dipic complexes, respectively. The perme-ability of V(V)-dipic complexes is much better than the oth-ers, which is in agreement with its better hypoglycemic effect in animal tests. On Caco-2 cell monolayer, Papp were found to be in the range of 1-3×10?6 cm/s and not to be affected by excessive amounts of dipicolinate ligand. By contrast, the permeability in the AP→BL direction across the MDCK monolayer increased greatly in the presence of free ligands, suggesting existence of active transport mechanism of vana-dium complex anions on the MDCK cells. The cytotoxicity of the three complexes was found similar and the IC50 were measured in the range of 0.6―0.9 mmol/L for MDCK cells and 1.6―2 mmol/L for Caco-2 cells. The cytotoxicity of three vanadium complexes was conceivably in consistence with their permeability, suggesting that the toxicity, permeation and cellular metabolism of vanadium complexes are closely related.

In the SMA the expression of Nox4 （4wks） and gp91 （8wks） sub-units of NADPH oxidase from STZ - apoE - / - were enhanced as was eNOS mRNA （P ＜ 0.05）. Oxidative stress was increased in mouse aortic endothelial cells treated with high glucose （HG） compared to normal low glucose medium;oxidative stress in HG was lowered by treatment with sepiapterin and eNOS mRNA and protein were increased significantly Conclusions: Increased oxidative stress in the vasculature of STZ - apoE - / - mice is linked to changes in eNOS and NADPH oxidase expression.
