Stem cells(SCs)with their self-renewal and pluripotent differentiation potential,show great promise for therapeutic applications to some refractory diseases such as stroke,Parkinsonism,myocardial infarction,and diabetes.Furthermore,as seed cells in tissue engineering,SCs have been applied widely to tissue and organ regeneration.However,previous studies have shown that SCs are heterogeneous and consist of many cell subpopulations.Owing to this heterogeneity of cell states,gene expression is highly diverse between cells even within a single tissue,making precise identification and analysis of biological properties difficult,which hinders their further research and applications.Therefore,a defined understanding of the heterogeneity is a key to research of SCs.Traditional ensemble-based sequencing approaches,such as microarrays,reflect an average of expression levels across a large population,which overlook unique biological behaviors of individual cells,conceal cell-to-cell variations,and cannot understand the heterogeneity of SCs radically.The development of high throughput single cell RNA sequencing(scRNA-seq)has provided a new research tool in biology,ranging from identification of novel cell types and exploration of cell markers to the analysis of gene expression and predicating developmental trajectories.scRNA-seq has profoundly changed our understanding of a series of biological phenomena.Currently,it has been used in research of SCs in many fields,particularly for the research of heterogeneity and cell subpopulations in early embryonic development.In this review,we focus on the scRNA-seq technique and its applications to research of SCs.

Chronic lung diseases have been recognized as one of the world's leading causes of death in recent decades. Lacking effective treatments brings the patients not only bad quality of life but also higher risk for lung cancer development. By increasing the understanding of deeper mechanism of how lung develops and regenerates, researchers now focus on studying lung regenerative medicine, aiming to apply different and more efficient therapies to treat chronic lung diseases. This review will provide a wide picture of both basic lung developmental, regeneration mechanism and different designed strategies for treating chronic lung diseases in the future decades.

The c/s-regulatory elements facilitate the binding of sequencespecific transcription factors for activating or repressing the expression of target genes.The expression level of each gene may vary in different cell types due to differential regulatory ele-ments within the accessible chromatin regions(ACRs),which can dictate cell-fate decisions and developmental trajectories.Therefore,it has become essential to identify genome-wide ACRs across cell types and developmental stages to understand the impact of chromatin accessibility on gene expression regula-tion and,consequently,trait variation.

Objective:To reveal the effects of 9-cis retinoic acid (RA) on cortical granules (CGs) migration andin vitro embryo production (IVP) rate in meiosisⅡvitrified bovine oocyte. Methods:Followingin vitro maturation (IVM) in a medium containing 5 nM of 9-cis-RA, 60 oocytes were vitrified and then thawed. Then, half of them were evaluated for CGs migration and the other half were used for in vitro fertilization and IVP (on day 3). In addition, the other 60 oocytes were considered as the control group, which did not receive RA in IVM medium. The data were presented to one way analysis of variance (ANOVA) and Duncan's test.Results:Results showed that IVP rate (4-8 cells embryos) was significantly (Pdevelopmental competence and CGs distribution of meiosisⅡvitrified bovine oocyte. Therefore, adding RA in IVM medium can decrease the ultrastructural changes during vitrification and can improve the efficiency of bovine oocyte vitrification.

Acrylamide has been brought to the world attention in 2002 with the information that it was found contaminated in many heat treated carbohydrate rich foods. This creates a lot of concern worldwide since the appearance of Aflatoxins many decades ago. The dilemma occurs as the heat treated carbohydrate rich foods are being consumed by population around the world while the high in complex carbohydrate diet is being promoted by international organizations as FAO and WHO in the effort for the prevention of some chronic diseases.
