A novel approach based on metabolomics coupled with network pharmacology to explain the effect mechanisms of danggui Buxue Tang in anaemia

danggui Buxue Tang(DBT) is a famous Chinese medicinal decoction. Mechanism of DBT action is wide ranging and unclear. Exploring new ways of treatment with DBT is useful. Sprague–Dawley(SD) rats were randomly divided into 3 groups including control(NC, Saline), the DBT(at a dose of 8.10 g?kg–1), and blood deficiency(BD)(Cyclophosphamide(APH)-and Cyclophosphamide(CTX)-induced anaemia). A metabolomics approach using Liquid Chromatography-Quadrupole-Time-of-Flight/Mass Spectrometry(LC/Q-TOFMS) was developed to perform the plasma metabolic profiling analysis and differential metabolites were screened according to the multivariate statistical analysis comparing the NC and BD groups, and the hub metabolites were outliers with high scores of the centrality indices. Anaemia disease-related protein target and compound of DBT databases were constructed. The TCMSP, Chem Mapper and STITCH databases were used to predict the protein targets of DBT. Using the Cytoscape 3.2.1 to establish a phytochemical component–target protein interaction network and establish a component, protein and hub metabolite protein–protein interaction(PPI) network and merging the three PPI networks basing on BisoGenet. The gene enrichment analysis was used to analyse the relationship between proteins based on the relevant genetic similarity by ClueGO. The results shown DBT effectively treated anaemia in vivo. 11 metabolic pathways are involved in the therapeutic effect of DBT in vivo; S-adenosyl-L-methionine, glycine, L-cysteine, arachidonic acid(AA) and phosphatidylcholine(PC) were screened as hub metabolites in APH-and CTX-induced anaemia. A total of 288 targets were identified as major candidates for anaemia progression. The gene-set enrichment analysis revealed that the targets are involved in iron ion binding, haemopoiesis, reactive oxygen species production, inflammation and apoptosis. Effect of the herbal medicine danggui Sini plus Wuzhuyu Shengjiang Tang on erythrocyte deformability in normal subjects: a cross-over trial

OBJECTIVE: To show whether danggui Sini plus Wuzhuyu Shengjiang Tang(DSWST) has any transient effect on erythrocyte deformability in normal ***: A total of 25 subjects [mean age(27.8± 1.8) years] was enrolled in this study. The study was designed as a cross-over trial in which the subjects took part for 2 d. On the first day, blood samples were collected at baseline and 1-2 h after administration of water, whereas, on the second day, instead of water, the subjects were administered DSWST after the baseline blood sampling. The blood samples collected at baseline and after the administration water or DSWST, were examined for erythrocyte ***: The elongation index increased significantly after 2 h(P = 0.009) compared to the baseline after DSWST intake. However, after water intake, there was no significant differenceobserved. When comparing the percent change of erythrocyte deformability between DSWST and water, we found that after 2 h of administration,DSWST improved erythrocyte deformability significantly compared to water(P < 0.001).CONCLUSION: DSWST has a transient effect on erythrocyte deformability in normal subjects.

Uncovering Chemical Interactions between Danshen and danggui Using Liquid Chromatography–Mass Spectrometry and Network Pharmacology-Based Research on Stroke

Objective: The objective of this study was to decipher chemical interactions between Danshen and danggui using liquid chromatography–mass spectrometry(LC-MS) and explore the mechanisms of Danshen–danggui against stroke using network pharmacology and molecular docking. Materials and Methods: First, the chemical compounds of Danshen–danggui were profiled using ultra-high-performance liquid chromatography(HPLC)-quadrupole time-of-flight MS. Accurately characterized compounds in various proportions of Danshen–danggui were quantified using HPLC combined with triple quadrupole electrospray tandem MS. Network pharmacology was used to uncover the essential mechanisms of action of Danshen–danggui against stroke. Discovery Studio Software was used for the molecular docking verification of key active chemicals and stroke-related targets. Results: A total of 53 compounds were characterized, and 22 accurately identified constituents(10 phenolic acids, 8 phthalides, and 4 tanshinones) were quantified in 15 proportions of Danshen–danggui. The quantification results showed that danggui significantly increased the dissolution of most phenolic acids(compounds from Danshen), whereas Danshen promoted the dissolution of most phthalides(compounds from danggui). Overall, the combination of Danshen and danggui at a 1:1 ratio resulted in the maximum total dissolution rate. Further network pharmacology and molecular docking results indicated that Danshen–danggui exerted anti-stroke effects mainly by regulating inflammation-related(tumor necrosis factor, hypoxia-inducible factor, and toll-like receptor) signaling pathways, which ranked among the top three pathways based on Kyoto Encyclopedia of Genes and Genomes(KEGG) enrichment analysis. Conclusion: The chemical compounds in Danshen–danggui could interact with each other to increase the dissolution of the most active compounds, which could provide a solid basis for uncovering the compatibility mechanisms of Danshen–danggui and Danshen–Danggu

Enhancing Schwann cell proliferation may be beneficial for peripheral nerve repair and nerve regeneration. A traditional herbal formula composed of Fuling （poria cocos）, Baizhu （Atractylodes macrocephala）, and danggui （Angelica sinensis）（FBD） improves neuronal survival and growth, and FBD may promote the secretion of brain-derived neurotrophic factor. However, the mechanism underlying Schwann cell proliferation remains unclear. We tested whether FBD enhanced the proliferation of human Schwann cells. FBD （20 ug/mL） increased Schwann cell viability and survival and increased the number of cells at G2/M and S phases. FBD also increased nerve growth factor and brain-derived neurotrophic factor expression in Schwann cells, with maximum efficacy at 20 ug/mL.

Advanced glycation end products (AGEs) have been regarded as a pivotal inducer in diabetes and kinds of diabetic nephropathy. The present studies explored the effects of danggui Buxue Tang (DBT) that is a Chinese medicinal de- coction on negative charge to Human Umbilical Vein Endothelial Cell (HUVEC) and the related mechanism. Alcian blue staining was established to evaluate the intensity of negative charge on HUVEC. Proteoglycan expressions of AGP and avidin were determined by SDS-PAGE. We observed that DBT can significantly increase negative charge on HU-VEC and up-regulated AGP and avidin expressions and ameliorate AGEs-induced HUVEC apoptosis. Therefore, all results showed DBT had prevention effects against the progression of AGEs-induced damage, and this decoction might be promising agent against proteinuria in diabetic nephropathy.

danggui is a widely used herbal drug in Traditional Chinese Medicine（TCM）,and adulteration with European danggui is frequently encountered in the market *** study aims at the chemical and biological comparison of danggui and European danggui.1H-NMR spectroscopy coupled with multivariate analysis was used to investigate the different metabolites between danggui and European danggui,and the results showed that they differed not only in the secondary metabolites,but also regarding the primary *** danggui contained higher contents of valine,proline,fumaric acid,phenylalanine,nicotinamide derivative,Z-butylidenephthalide,coniferyl ferulate,ferulic acid,Z-ligustilide and Z,Z-6,6’7,3a-diligustilide,while danggui showed higher levels of alanine,γ-aminobutyrate,adenosine,arginine,sucrose,α-glucose,β-glucose,tryptophan,and cis-Z,Z’-3a.7a’,7a.3a’-*** blood deficiency model was used to compare their biological effect,and danggui extracts exerted better effects than that of European *** European danggui contained higher level of Z-ligustilide and ferulic acid,danggui showed better blood enriching effects than European ***,the bioactive compounds responsible for the blood enriching effect in danggui,as well as their possible synergistic effects,should be further studied.
