Nucleolus-localized def-CAPN3 protein degradation pathway and its role in cell cycle control and ribosome biogenesis

The nucleolus,as the‘nucleus of the nucleus’,is a prominent subcellular organelle in a eukaryocyte.The nucleolus serves as the centre for ribosome biogenesis,as well as an important site for cell-cycle regulation,cellular senescence,and stress response.The protein composition of the nucleolus changes dynamically through protein turnover to meet the needs of cellular activities or stress responses.Recent studies have identified a nucleolus-localized protein degradation pathway in zebrafish and humans,namely the def-CAPN3 pathway,which is essential to ribosome production and cell-cycle progression,by controlling the turnover of multiple substrates(e.g.,ribosomal small-subunit[SSU]processome component Mpp10,transcription factor p53,check-point proteins Chk1 and Wee1).This pathway relies on the Ca2þ-dependent cysteine proteinase CAPN3 and is independent of the ubiquitin-mediated proteasome pathway.CAPN3 is recruited by nucleolar protein def from cytoplasm to nucleolus,where it proteolyzes its substrates which harbor a CAPN3 recognition-motif.def depletion leads to the exclusion of CAPN3 and accumulation of p53,Wee1,Chk1,and Mpp10 in the nucleolus that result in cell-cycle arrest and rRNA processing abnormality.Here,we summarize the discovery of the def-CAPN3 pathway and propose its biological role in cell-cycle control and ribosome biogenesis.

Knowledge on pancreaticobiliary reflux in normal pancreaticobiliary junction and its pathologic implications has experienced tremendous progress during the last few years.This editorial reviews the current knowledge on this condition and its pathological implications on gallbladder diseases.The following aspects were def ined appropriate for discussion:(1) Evidence of carcinogenesis associated with pancreaticobiliary reflux;(2) Evidence of pancreaticobiliary reflux in normal pancreaticobiliary junction;and(3) Evidence of sphincter of Oddi(SO) dysfunction as a cause of pancreaticobiliary reflux in normal pancreaticobiliary junction.The articles reviewed were selected and classif ied according to f ive levels of evidence:Level-this condition.Although an important body of research has been published regarding pancreaticobiliary reflux in normal pancreaticobiliary junction and its clinical significance,the current evidence does not fully support what has been suggested.Studies with evidence level Ι have not been undertaken.This is a fascinating subject of study,and if finally supported by evidence level Ι,the importance of this condition will constitute a major breakthrough in biliary pathology.

AC operation with multiple full cycles has been successfully performed on the superconducting tokamak HT-7. In the experiment, it was discovered that the saturation of the transformer magnetic flux with def, a signal name, was one of key aspects that affected the AC operation. The conditions of def were examined through the def feedback control system. By controlling the working patterns of the subsystems, namely the poloidal field control system and density control system, it was guaranteed that def would remain in the non-saturated status.

The initiation mechanism of the copolymerization of 2-vinylnaphthalene with maleic anhydride was studied under irradiation of 365 nm. The excited complex was formed from (1) the local excitation of 2-vinylnaphthalene followed by the charge-transfer interaction with maleic anhydride and (2) the excitation of the ground state charge-transfer complex, and then it collapsed to 1,4-tetramethylene biradical for initiation. A1: 1 alternating copolymer was formed in different monomer feeds. Addition of benzophenone could greatly enhance the rate of copolymerization through energy-transfer mechanism.

Recovery of liver mass to a healthy liver donor by compensatory regeneration after partial hepatectomy(PH)is a prerequisite for liver transplantation.Synchronized cell cycle reentry of the existing hepatocytes after PH is seemingly a hallmark of liver compensatory regeneration.Although the molecular control of the PH-triggered cell cycle reentry has been extensively studied,little is known about how the synchronization is achieved after PH.The nucleolus-localized protein cleavage complex formed by the nucleolar protein Digestive-organ expansion factor(def)and cysteine proteinase Calpain 3(Capn3)has been implicated to control wounding healing during liver regeneration through selectively cleaving the tumor suppressor p53 in the nucleolus.However,whether the def-Capn3 complex participates in regulating the synchronization of cell cycle reentry after PH is unknown.In this report,we generated a zebrafish capn3b null mutant(capn3b^(Δ19Δ14)).The homozygous mutant was viable and fertile,but suffered from a delayed liver regeneration after PH.Delayed liver regeneration in capn3b^(Δ19Δ14)was due to disruption of synchronized cell proliferation after PH.Mass spectrometry(MS)analysis of nuclear proteins revealed that a number of negative regulators of cell cycle are accumulated in the capn3b^(Δ19Δ14)liver after PH.Moreover,we demonstrated that Check-point kinase 1(Chk1)and Wee1,two key negative regulators of G2 to M transition,are substrates of Capn3.We also demonstrated that Chk1 and Wee1 were abnormally accumulated in the nucleoli of amputated capn3bΔ19Δ14 liver.In conclusion,our findings suggest that the nucleolar-localized def-Capn3 complex acts as a novel regulatory pathway for the synchronization of cell cycle reentry,at least partially,through inactivating Chk1 and Wee1 during liver regeneration after PH.

在分析def的产生和破坏作用机制的基础上,根据铁路高地热隧道施工过程中温度的变化条件,分别对隧道初支和二衬混凝土或砂浆试件进行def加速模拟试验。试验结果表明,采用符合现行铁路混凝土技术标准要求的原材料,并对配制参数进行合理控制的前提下,混凝土或砂浆试件的膨胀率均低于0.04%的安全限值。说明铁路高地热隧道衬砌混凝土产生def破坏的风险很低,高地热环境对混凝土耐久性的影响较小。

In this paper,experimental findings concerning the kinetics of hematopoietic reconstitution are compared to corresponding clinical data.Although not clearly apparent,the transplantation practice seems to confirm the basic proposals of experimental hematology concerning hematopoietic reconstitution resulting from successive waves of repopulation stemming from different subpopulations of progenitor and stem cells.One of the "f irst rate" parameters in clinical transplantations in hematology;i.***.the CD34+ positive cell dose,has been discussed with respect to the functional heterogeneity and variability of cell populations endowed by expression of CD34.This parameter is useful only if the relative proportion of stem and progenitor cells in the CD34+ cell population is more or less maintained in a series of patients or donors.This proportion could vary with respect to the source,pathology,treatment,processing procedure,the graft ex vivo treatment and so on.Therefore,a universal dose of CD34+ cells cannot be def ined.In addition,to avoid further confusion,the CD34+ cells should not be named "stem cells" or "progenitor cells" since these denominations only concern functionally characterized cell entities.
