Objective To investigate the effect of (-)-Stepholidine (SPd) on enhancing d receptor mediated contraction of cardiac muscle in isolated rat heart and to examine whether SPd has a direct effect on the heart dopamine d *** an active ingredient of the Chinese herb Stephania intermedia,binds to dopamine d and d like ***,electrophysiological and behavioural experiments have pro- vided strong evidence that SPd is both a d(1/5) agonist and a d(2/4) antagonist,which could indicate u- nique antipsychotic *** Normal adult rat working hearts were isolated by Langendorff tech- *** SPd significantly increased the cardiac muscle contraction in a dose-dependent *** selective d dopamine receptor antagonist SCH23390 (1μM) blocked the SPd induced heart contraction, however,neither theβ-receptor antagonist propranolol (1μM) nor theα-receptor antagonist prazosin (1μM) had any effect on blocking SPd induced heart ***,the L-type Ca channel in- hibitor nimodipine (1μM) completely blocked the effect of SPd on cardiac muscle *** SPd show the effect on enhancing contraction of isolated rat heart through activating L-type Ca channel mediated by heart d receptors.

distinct Roles of dopamine receptors in the Lateral Thalamus in a Rat Model of decisional Impulsivity

The thalamus and central dopamine signaling have been shown to play important roles in high-level cognitive processes including impulsivity. However, little is known about the role of dopamine receptors in the thalamus in decisional impulsivity. In the present study,rats were tested using a delay discounting task and divided into three groups： high impulsivity（HI）, medium impulsivity（MI）, and low impulsivity（LI）. Subsequent in vivo voxel-based magnetic resonance imaging revealed that the HI rats displayed a markedly reduced density of gray matter in the lateral thalamus compared with the LI rats. In the MI rats, the dopamine d1 receptor antagonist SCH23390 or the d2 receptor antagonist eticlopride was microinjected into the lateral thalamus. SCH23390 significantly decreased their choice of a large, delayed reward and increased their omission of lever presses. In contrast,eticlopride increased the choice of a large, delayed reward but had no effect on the omissions. Together, our results indicate that the lateral thalamus is involved in decisional impulsivity, and dopamine d1 and d2 receptors in the lateral thalamus have distinct effects on decisional impulsive behaviors in rats. These results provide a new insightinto the dopamine signaling in the lateral thalamus in decisional impulsivity.

Objective Risk management can guide individual’s behavior by accessing the degree of risk. It is well known that the prefrontal cortex is involved in decision making, and dopaminergic system plays an important role in decision making. Our work aims at constructing risk management behavior on rats and exploring the function of dopamine in prefrontal cortex when the animal is executing risk decision making task. Methods We apply animal ethology method on establishing behavior model of risk management in rat. Chemical reagents can specifically effects on prefrontal by fixed point micro injection. dopamine can temporally flexible attend or not in the behavioral tasks by applying the agonist and antagonist of the main dopamine receptor d1 and d2. Results We found that most of the rats are more inclined to choose risk option, rather than safe option. When prefrontal cortex is blocked, rats prefer risk option. Risk management behavior can be modulated by the agonist and antagonist of dopamine receptor. Conclusion Prefrontal cortex is necessary in risk management behavior, and PFC dopamine modulates risk decision making behavior.

dopamine plays an critical role in many disease,such as Parkinson's *** is widely reported to decrease insulin secretion in cultured pancreatic β cells,while some reported the effects of dA on insulin secretion displayed dose ***'s declared dA inhibits insulin secretion directly via d which has been declared express on β cells,but others have found the inhibition effects can't be blocked by d *** has been reported that d antagonist decreased the occurrence of diabetes mellitus which occurs naturally in NOd *** have demonstrated that d is presented in β cells while d is presented in 8 cells in rat ***,we found that d antagonist can not only block the inhibition of dA on insulin secretion,but also promote insulin *** hypothesis that dA can decrease insulin secretion via activating d in δ cells and increase insulin secretion via activating d in β cells,which depends on its *** results demonstrate that 1) high concentration of dA and d agonists promoted somatostatin secretion combine with inhibited insulin ***,the inhibition effects can be blocked by SST receptor 2 antagonists.2) Low concentration of dA and d agonist promoted insulin secretion in rats and isolated islets.3) d siRNA mice manifested increased insulin secretion and decreased blood glucose.4) In human islets,d located in β cells and d presented in 5 cells;in β cells destroyed rat model,d significantly decreased while d manifested constantly normally *** research demonstrates that dA can both inhibit insulin secretion indirectly by activating d and increase insulin secretion via directly activating by d,and the final effects depend on its concentration.
