Malignant tumors have always threatened human health.Among them,platinum metal complexes have been more mature clinical applications,but because they still have some shortcomings,it is the main direction to explore high-efficiency and low-toxic anti-tumor drugs.Three new iridium(Ⅲ) complexes [Ir(ppy)(cdppz)](PF)(Ir-1),[Ir(bzq)2(cdppz)](PF6)(Ir-2) and [Ir(ppy)2(cdppz)](PF6)(Ir-3,Scheme 1) were synthesized and characterized.These three complexes were then evaluated in vitro cytotoxicity by MTT assay.IC values of the complexes toward cancer cells are listed in Table 1.The results showed that SGC-7901 was highly inhibited by the complexes.Flow cytometry was used to detect ROS,mitochondrial membrane potential and cell cycle.The results showed that the level of ROS in the cells increased significantly and the mitochondrial membrane potential decreased,which further proved that the complexes could block the cell cycle at G0/G1 phase to inhibit the cancer cell growth.In addition,cytotoxicity in vivo showed that complex Ir-2 exhibits high antitumor activity(Figure 2).Taken together,the complex induces apoptosis in SGC-7901 cells via ROS-mediated mitochondrial dysfunction pathway.
