Peritumoral edema is often associated with glioma and leads to surgical complications and prolonged hospitalization. Inflammation significantly influences the formation of peritumoral edema. cysteinyl leukotrienes, metabolites of arachidonic acid, are potent inflammatory mediators that induce inflammation by activating cysteinyl receptor 2(Cys LT2). The aim of this study was to investigate Cys LT2 expression in human gliomas and evaluate its correlation with peritumoral edema. We retrospectivelystudied 76 patients with glioma that were surgically treated in our institute over a 2-year period. Immunouorescence staining was used to detect Cys LT2. The expression level of Cys LT2 was determined with western blot techniques. Peritumoral edema was classified into 3 degrees based on preoperative MRI imaging. Cys LT2 expression was located in the astrocytes of gliomas. A significant upregulation of Cys LT2 expression was observed in higher peritumoral edema grade gliomas. There was a statistically significant positive correlation between Cys LT2 expression and the presence of peritumoral edema. In summary, This study suggests Cys LT2 plays an important role in the cerebral water homeostasis of human gliomas.

Distribution of cysteinyl leukotriene receptor 2 in human traumatic brain injury and brain tumors

Aim: To determine the distribution of cysteinyl leukotriene receptor 2 （CysLT2）, one of the cysteinyl leukotriene receptors, in human brains with traumatic injury and tumors. Methods: Brain specimens were obtained from patients who underwent brain surgery. CysLT2in brain tissues was examined using immunohis-tochemical analysis. Results: CysLT2was expressed in the smooth muscle cells （not in the endothelial cells） of arteries and veins. CysLT2was also expressed in the granulocytes in both vessels and in the brain parenchyma. In addition, CysLT2was detected in neuron- and glial-appearing cells in either the late stages of traumatic injury or in the area surrounding the tumors. Microvessels regenerated 8 d after trauma and CysLT2expression was recorded in their endothelial cells. Conclusion: CysLT2is distributed in vascular smooth muscle cells and granulocytes, and brain trauma and tumor can induce its expression in vascular endothelial cells and in a number of other cells.

Objective cysteinyl leukotrienes(CysLTs) induce inflammatory responses mediated by activating CysLT1 and CysLT2 *** have recently reported CysLT2 receptor is involved in neuron injury,astrocytosis and microgliosis after focal cerebral ischemia in ***3379 is a selective CysLT2 receptor antagonist,whether HAMI3379 can exert protective effect on focal cerebral ischemia in rats remains *** chloral hydrate anesthesia,transient focal cerebral ischemia was induced in rats by 30min of middle cerebral artery occlusion(MCAO),followed by 24h of *** as vehicle,pranlukast(0.1mg/kg,i.p.) as a positive control or HAMI3379(i.c.v.)at the doses of 1,10,100 ng was administered at 30 min before induction of ***,24h after MCAO behavior assessment,brain edema, infarct volume,neuronal damage and IgG exudation were ***3379 at the doses of 10,100 ng could decrease lesion volume,brain edema,IgG exudation,neuron degeneration,and neuron *** protective effect was similar to CysLTl receptor antagonist ***3379 at the dose of lng had no obvious effect on the cerebral ischemia injury. Thus,HAMI3339 has a dose-dependent neuroprotective effects on transient focal cerebral ischemia in rats,with an effective dose range of 10-100 *** findings further support CysLT2 receptor plays regulatory roles in acute neuron injury and the therapeutic potential of CysLT2 receptor antagonists in the treatment of cerebral ischemia at earlier *** studies need to be conducted in order to determine the effects of CysLT2 receptor on the late phase of ischemic injury and the potential mechanism.
