Phylogeny and Biogeography of Thuja L. （cupressaceae）, an Eastern Asian and North American Disjunct Genus

In order to develop better insights into biogeographic patterns of eastern Asian and North American disjunct plant genera, sequences of nuclear ribosomal DNA internal transcribed spacer (nr DNA ITS) region were used to estimate interspecific relationships of Thuja L. (cupressaceae) and infer its biogeography based on the phylogeny. According to the phylogenetic analysis, two clades were recognized. The first clade included Thuja plicata D. Don (western North America) and T. koraiensis Nakai (northeastern Asia), and the second one contained T. occidentalis(Gord.) Carr. (Japan). The ancestral area of Thuja was inferred to be eastern Asia, and two dispersal events were responsible for the modern distribution of Thuja in North America. Both the North Atlantic land bridge and Bering land bridge were possible routes for the migration of ancestral populations to North America.

This paper examed the cupressaceae taxa in China. It is identifled that 30 species and 16 varieties in 7 genera in thefamily are indigenous, and estimated that about 25 species and 4 varieties have been introduced into this country since the beginning of this century. It is urgent that more furthur studies need to be carried out in the country on the taxonomy especially of junipers and cypresses, on the re-classification ofexisting cultivars, and on the development of new cultivars for future needs.

After a merger of the former Taxodiaceae and cupressaceae(sensu stricto), currently the conifer family cupressaceae(sensu lato) comprises seven subfamilies and 32 genera, most of which are important components of temperate and mountainous forests. Previous phylogenetic studies of cupressaceae were based mainly on morphological characters or a few molecular markers, and did not completely resolve the intergeneric relationships. In this study, we reconstructed a robust and well-resolved phylogeny of cupressaceae represented by all 32 genera, using 2070 genes(Orthogroups) generated from transcriptome sequencing. The phylogenetic placements of some genera that remained controversial, such as Athrotaxis, Papuacedrus and Calocedrus, have been anchored. Further analyses suggest an ancient hybridization event that possibly occurred between two subclades of Cupressoideae and gave rise to a monophyletic group comprising five genera(Juniperus, Cupressus, Callitropsis, Hesperocyparis and Xanthocyparis). Based on the robust phylogeny, we also conducted divergence time estimation and ancestral area reconstruction. The results indicate that cupressaceae very likely originated in Asia in the Triassic.

Epicuticular waxes have vital roles in the growth and development of plants and in defense. Conifers have a considerable amount of waxes on their cones and leaves.Here we characterized the n-alkane composition of Iranian conifers, including Juniperus oblonga, J. foetidissima, J.sabina, J. communis subsp. hemisphaerica, J. excelsa,Cupressus sempervirens, Platycladus orientalis from cupressaceae and Taxus baccata from Taxaceae for the first time using GC-FID analyses. In the waxes, 25 n-alkane homologs with chain lengths ranging from C7 to C32 were identified. Short-chain n-alkanes were dominant in almost all samples with some exceptions. Complementary studies to elucidate complete wax constituents of Iranian conifers and n-alkane distribution pattern as a function of geographical and bioclimatic variables are recommended.
