Evidence for the paternal mitochondrial DNA in the crucian carp-like fish lineage with hybrid origin

In terms of taxonomic status,common carp(Cyprinus carpio,Cyprininae)and crucian carp(Carassius auratus,Cyprininae)are different species;however,in this study,a newborn homodiploid crucian carp-like fish(2n=100)(2nNCRC)lineage(F1–F3)was established from the interspecific hybridization of female common carp(2n=100)×male blunt snout bream(Megalobrama amblycephala,Cultrinae,2n=48).The phenotypes and genotypes of 2 n NCRC differed from those of its parents but were closely related to those of the existing diploid crucian *** further sequenced the whole mitochondrial(mt)genomes of the 2n NCRC lineage from F1 to *** paternal mt DNA fragments were stably embedded in the mt-genomes of F1–F3 generations of 2n NCRC to form chimeric DNA *** with this chimeric process,numerous base sites of F1–F3 generations of 2 n NCRC underwent *** of these mutation sites were consistent with the existing diploid crucian ***,the mt DNA organization and nucleotide composition of 2n NCRC were more similar to those of the existing diploid crucian carp than those of the *** inheritable chimeric DNA fragments and mutant loci in the mt-genomes of different generations of 2nNCRC provided important evidence of the mt DNA change process in the newborn lineage derived from hybridization of different *** findings demonstrated for the first time that the paternal mt DNA were transmitted into the mt-genomes of homodiploid lineage,which provided new insights into the existence of paternal mt DNA in the mt DNA inheritance.

crucian carp(Carassius auratus gibelio),an extensively cultivated freshwater fish,was one of the model species for the study of fish *** antibodies were advantageous molecular tools for studying teleost immune ***,polyclonal antibodies reacting with immunoglobulins(Ig)were used successfully in studies of the teleost *** the present study,we produced polyclonal antibody against CH2 domains of crucian carp IgM,and measured the in vivo dynamics of IgM mRNA caused by CyHV-2 *** recombinant protein IgM with relative molecular weight about 53 KD was correctly expressed in prokaryotic *** specificity of the polyclonal antibody was evaluated by Western blotting and results revealed that the antibody not only specifically recognized crucian carp serum but also cross-reacted with grass carp *** RT-PCR analysis demonstrated the expression of IgM mRNA changed significantly after CyHV-2 *** expression of IgM in the kidney increased and reached a maximum at 6 h post-infection(hpi),while dropped to a low level at 5 days post-infection(dpi).In conclusion,the expression of IgM was significantly upregulated in the kidney of crucian carp infected with CyHV-2,indicating that IgM played a potential role in systemic immunity against viral *** antibody against crucian carp IgM had certain clinical relevance,which might provide insight into the early stage of virus infection and prevention of the disease.

Neomycin belongs to the family of 2-deoxystreptamine-containing aminoglycoside *** is widely used for bacterial infections,targeting most gram-negative and some gram-positive *** has also been reported to show antiviral ***,we evaluated the toxicity of neomycin sulfate,and investigated its effect on non-specific immunity and viral infection in crucian carp(Carassius auratus gibelio).The safe concentration of neomycin sulfate for crucian carp was determined to be 102.9 mg/kg in *** oxidative damage assays,neomycin sulfate increased superoxide dismutase and catalase activity and decreased malondialdehyde in the liver of crucian *** non-specific blood immune assays,the white blood cell count and complement 3 content significantly increased after neomycin sulfate treatment,while no significant changes were observed in antibacterial or lysozyme *** a challenge test,neomycin sulfate protected the crucian carp from cyprinid herpesvirus 2(CyHV-2)infection and inhibited CyHV-2 *** cytotoxicity assays,low concentrations of neomycin sulfate had no cytotoxicity on cells from the fins of crucian *** results of the present study indicate that oral administration of neomycin sulfate reduced oxidative damage,enhanced immunity and provided protection against CyHV-2 in crucian carp.

A myofibril-bound serine proteinase(MBSP)was highly purified from the skeletal muscle of crucian carp(carasius auratus)by acidic treatment of myofibril solution and chromatographies on Q-Sepharose and benzamidine-Sepharose *** revealed a main protein band of approximately 28 kDa on SDS-PAGE and was particularly inhibited by serine proteinase *** specificity analysis revealed that the enzyme specifically cleaved at the carboxyl side of arginine and lysine residues,suggesting the characteristic of a trypsin-type serine *** gene was cloned based on the N-terminal sequence and the conserved active site peptide of serine proteinases together with 5’-RACE and 3’-*** coding region gave an amino-acid sequence of 242 residues including the initiation methionine and a signal peptide of 20 *** acid residues of His,Aspand Serconsisting the catalytic triad of serine proteinases were conserved in the *** carp MBSP shared relatively high identities with other serine proteinases,especially in well-conserved regions.

The effect of soybean trypsin inhibitor(STI)on the degradation of crucian carp myofibrillar proteins caused by an endogenous myofibril-bound serine proteinase (MBSP)was *** trypsin inhibitor was purified to high homogeneity and mixed with myofibrils and its inhibitory effect on myofibrillar protein degradation was *** the absence of STI,proteolysis of myofibrillar proteins including myosin heavy chain,α-actinin,actin and tropomyosin could be identified after incubation at 55℃ for 5 to 20 min depending on the kind of the *** contrast,in the presence of STI,with final concentration of 0.75μg/ml,proteolysis of these proteins was greatly suppressed even after incubation for 1 h,suggesting STI is an effective inhibitor in preventing myofibrillar protein degradation caused by a serine proteinase that is quite possibly *** STI reveals disadvantage for food digestion,as a native food grade inhibitor,it has the safety as food additive,especially at low *** in surimi production,the decrease of elasticity is always accompanied with the degradation of myofibrillar proteins,thus,the present result suggested the possibility that STI be applicable in surimi production in order to enhance the elasticity that is the quality of the final products.

Polybrominated diphenyl ethers(PBDEs) are a class of flame retardants that known to have bioaccumulation and potential toxicity to *** PBDEs in animals have been studied in many previous researches,little is known about the single low brominated diphenylethers of their fate in the aquatic *** the present study,we investigated the tissue distribution,excretion, depuration,and potential biotransformed products of 4,4’-dibromodiphenyl ether(BDE 15) after water exposure using freshwater fish crucian carp(Carassius auratus).Fish were exposed to BDE 15 at different concentrations for 50 days,followed by a 14-day depuration period,to examine the levels of BDE 15 in the muscle,gill,liver and gut content *** analyses,bioaccumulation parameters, such as rate constants of uptake(k) and depuration(k),and corresponding half-life(t) were also *** showed that obviously rapid uptake and elimination of BDE 15 by carps was observed in both treated groups,and BDE 15 was most concentrated in the gill and liver to reach the peak concentrations on day *** corresponding estimated bioconcentration factor(BCF) values obtained for liver and gill in the low-dose treatment(1.28×10 and 1.69×10 dry weight,respectively) were higher than for ***,five unknown compounds were detected as BDE 15 metabolites in the exposed fish,a novel finding which indicated that debromination of BDEs was not the only metabolic pathway observed in *** 4-bromodiphenyl ether and diphenyl ether, another three products were proposed tentatively as mono-OH-BDE 15,diOH-BDE 15 and bromophenol using GC/EI-MS/*** the best of our knowledge,it is the first indications for the oxidation metabolism products of PBDEs in vivo exposed freshwater *** results will be useful for determination of the origin of OH-PBDEs present in wild aquatic organisms.

Myofibril-bound serine proteinase(MBSP)in the skeletal muscle of crucian carp was *** of myofibrillar proteins by the endogenous MBSP was *** heavy chain(MHC)degraded markedly when crucian carp myofibril was incubated at around 55℃ as shown by *** incubation of myofibril at 55℃ also caused the obvious degradation of tropomyosin,while the decomposition of other myofibrillar proteins such as α-actinin and actin was slight as detected by Western *** results suggest the existence of an endogenous myofibril associated proteinase in crucian carp myofibril,which efficiently cleaves MHC and *** proteinase inhibitors(Lima bean trypsin inhibitor,PMSF and benzamidine)greatly suppressed the degradation of MHC caused by the enzyme,while inhibitors for cysteine,metallo,and asparatic proteinases only partially or completely did not show any inhibitory effect,indicating that the endogenous proteinase is a serine *** specificity analysis using partial purified crucian carp MBSP suggested that the enzyme is a trypsin-like serine proteinase.
