Aphis glycines Matsumura is an important pest of soybean in Asia and North America. Hymenoptera parasitoids play a key role in the control of the soybean aphid. Thecorrect identification of parasitoids is a critical step that precedes the assessment of their potential biological control agents. Accurate identification of the majority of the speciesattacking the soybean aphid often requires elaborate specimen preparation and expert taxonomic knowledge. In this study, we facilitated the identification of soybean aphidparasitoids by applying a DNA barcoding approach following a preliminary morphological identification. We generated DNA sequence data from the mitochondrial COI gene andthe D2 region of 28S rDNA to assess the genetic variation within and between parasitoid species emerging from the soybean aphid in China. Fifteen Hymenoptera parasitoid speciesbelonging to 10 genera of five families were identified with little intra-specific variation （0.09% ± 0.06% for 28S and 0.36% ± 0. 18% for COI） and large inter-specific divergence（30.46% ± 3.42% for 28S and 20.4% ± 1.20% for COI）.

Based on morphological and genetic evidence we evaluated the taxonomic status of a newly discovered forest-dwelling population of skink （genus Scincella） from the Keo Selma Wildlife Sanctuary, Mondulkiri Province, Cambodia. From phylogenetic analysis of a 668-bp fragment of the mtDNA col and diagnostic morphological characters we allocate the newly discovered population to the Scincella reevesii-S. rufocaudata species complex and describe it as Scincella nigrofasciata sp. nov. The new skink species can be distinguished from all other Southeast Asian congeners by the following combination of morphological characters： snout-vent length （SVL） 40.0-52.6 mm; relative tail length （TaL/SVL ratio） 1.25-1.94; prefrontals in broad contact; infralabials 6; primary temporals 2; relative forelimb length （FIL/SVL ratio） 0.20-0.22; relative hindlimb length （HIL/SVL ratio） 0.30-0.33; relative forearm length （FoL/SVL ratio） 0.14-0.16; adpressed forelimbs and hind limbs either overlapping （0.4-2.2 mm） or separated （1.9-2.3 mm）; midbody scale rows 32-33, paravertebral scales 69-74, vertebral scales 65-69; dorsal scales between dorsolateral stripes 8; comparatively slender fingers and toes, subdigital lamellae under fourth toe 15-17; dark discontinuous regular dorsal stripes 5-7; distinct black dorsolateral stripes, narrowing to lateral sides and extending to 52%-86% of total tail length. We provide additional information on the holotype of Scincella rufocaudata （Darevsky ＆ Nguyen, 1983）, and provide evidence for the species status of Scincella rupicola. Our discovery brings the number of Scincellaspecies in Cambodia to five and emphasizes the incompleteness of knowledge on the herpetofaunal diversity of this country.

Objective: To investigate the potential mechanisms that curcumin reverses 5-fluorouracil(5-FU) multidrug resistance(MDR). Methods: Cell growth and the inhibitory rate of curcumin(2–25 μg/mL) and/or5-FU(0.05–1000 μg/mL) on human colon cancer HCT-8 and HCT-8/5-FU(5-FU-resistant cel line) were determined using cel counting kit-8(CCK-8) assay. Apoptosis and cel cycle after 5-FU and/or curcumin treatment were detected by ?ow cytometry(FCM) and transmission electron microscopy(TEM). The expression of the multidrug resistance related factors p-glycoprotein(P-gp) and heat shock protein 27(HSP-27) genes and proteins were analyzed by reverse transcription polymerase chain reaction(RT-PCR) and Western blotting(WB), respectively. Results: The inhibitory rate of curcumin or 5-FU on HCT-8 and HCT-8/5-FU cells proliferation at exponential phase were in a dosedependent manner, HCT-8 cell line was more sensitive to curcumin or 5-FU when compared the inhibitory rate of HCT-8/5-FU. The 50% inhibitory concentration(IC50) of combination 5-FU and curcumin(4.0 μg/mL)in HCT-8/5-FU was calculated as 179.26 μg/mL, with reversal fold of 1.85. Another IC50 of combination 5-FU and curcumin(5.5 μg/mL) in HCT-8/5-FU was calculated as 89.25 μg/mL, with reversal fold of 3.71. Synergistic effect of 5-FU and curcumin on HCT-8 and HCT-8/5-FU cells were found. The cell cycle analysis performed by FCM showed that HCT-8 and HCT-8/5-FU cel s mostly accumulated at G0/G1 phase, which suggested a synergistic effect of curcumin and 5-FU to induce apoptosis. FCM analysis found that the percentage of apoptosis of cel s treated with curcumin, 5-FU and their combination were signi?cantly increased compared to the control group(P<0.05), and the percentage of apoptosis of the combination groups were slightly higher than other groups(P<0.05). The m RNA levels of P-gp(0.28±0.02) and HSP-27(0.28±0.09) in HCT-8/5-FU cel s treated with combination drugs were lower than cel s treated with 5-FU alone(P-gp, 0.48±0.07, P=0.009;HSP-27, 0.57±0.10, P=0.007). In the Memorandum of Understanding on Health Technology Cooperation issued in Beijing in 1978,the Ministry of Health(MOH)of China and the World Health Organization(WHO)agreed to create a series of WHO collaborating Centres(WHOCC).The Institute of Parasitic Diseases at the Chinese Academy of Medical Sciences(the predecessor of the National Institute of Parasitic Diseases[NIPD]at China CDC)was officially designated as a“WHO collaborating Centre for Malaria,Schistosomiasis,and Filariasis”in 1980 based in Shanghai(1).Since then,the WHOCC for Malaria,Schistosomiasis,and Filariasis has played an important role in promoting the development of medical science in China.

Let G=(V,E)be a graph andφbe a total coloring of G by using the color set{1,2,...,k}.Let f(v)denote the sum of the color of the vertex v and the colors of all incident edges of v.We say thatφis neighbor sum distinguishing if for each edge uv∈E(G),f(u)=f(v).The smallest number k is called the neighbor sum distinguishing total chromatic number,denoted byχ′′nsd(G).Pil′sniak and Wo′zniak conjectured that for any graph G with at least two vertices,χ′′nsd(G)(G)+3.In this paper,by using the famous Combinatorial Nullstellensatz,we show thatχ′′nsd(G)2(G)+col(G)-1,where col(G)is the coloring number of G.Moreover,we prove this assertion in its list version.

目的对乳猪窦房结所在部位进行准确定位,建立一套可靠的窦房结细胞取材、分离、纯化培养技术;观察窦房结细胞与colⅠ纤维支架的相容性。方法24h内新生纯种长白山乳猪5只,雌雄不拘,体重0.45～0.55kg。实验动物采用多导电生理记录仪标记房波最早出现的部位,在该位置取材行原代窦房结细胞培养,分别采用常规方法培养和差速贴壁分离技术、5-BrdU处理纯化培养;于左心房部位取材行心房肌细胞纯化培养作为对照。倒置显微镜观察细胞形态、细胞贴壁时间、单细胞出现搏动时间、搏动频率等。将纯化培养的窦房结细胞以5×105个/mL密度接种至经预湿处理的colⅠ纤维支架复合培养5d,行HE染色观察及扫描电镜观察。结果房波最早出现的位置即为窦房结所在部位。纯化培养的窦房结细胞有3种形态,即梭形、三角形与不规则形,其中梭形细胞最多;心房肌细胞主要为三角形及不规则形,而无梭形细胞。窦房结细胞纯化培养与常规培养比较,梭形细胞所占比例分别为73.0%±2.9%、44.7%±2.3%,二者差异有统计学意义(P0.05)。窦房结细胞复合colⅠ纤维支架培养5d,HE染色观察示colⅠ纤维支架材料中有大量细胞;扫描电镜见细胞成团吸附于支架表面及孔隙侧壁,并可见细胞间通过突起相互连接,或伸出伪足贴附于支架孔隙壁上。结论通过准确窦房结定位,采用差速贴壁结合5-BrdU处理的纯化培养可明显提高乳猪窦房结梭形细胞比例,是一种可靠的窦房结细胞纯化培养技术。窦房结细胞和colⅠ纤维支架有较好的细胞相容性。

目的观察TGF-β1中和抗体对TGF-β诱导的肌腱细胞胶原产生及术后粘连形成的影响,探讨生物学调节在肌腱粘连防治中的作用。方法取6只成年新西兰大白兔12条屈趾肌腱分离肌腱成纤维细胞、腱外膜细胞和腱内膜细胞,将细胞随机分成2组,实验组加入1ng/mL TGF-β后,再加入不同浓度TGF-β1中和抗体,对照组不添加任何试剂,培养3d后ELISA测定colⅠ的产生。取84只兔行中趾屈趾肌腱切断吻合术,随机分成3组,腱鞘内分别注入生理盐水(NS组,n=36)、1.0μg/mL TGF-β1中和抗体(1.0μg/mL TGF-β1组,n=36)和2.0μg/mL TGF-β1中和抗体(2.0μg/mL TGF-β1组,n=12)。术后4、8周取出肌腱行肌腱粘连检测、生物力学测定、组织学观察和扫描电镜观察。1、2、4、8周后取出NS组及1.0μg/mL TGF-β1组肌腱,原位杂交方法测定TGF-β1和colⅠmRNA的表达。结果ELISA检测显示,TGF-β1能明显提高肌腱细胞colⅠ的产生;TGF-β1抗体能降低肌腱成纤维细胞、腱外膜细胞和腱内膜细胞colⅠ的产生,且呈剂量依赖关系。术后4、8周,NS组屈趾肌腱滑动距离较短,模拟主动屈曲度明显受限,与1.0μg/mL TGF-β1组和2.0μg/mL TGF-β1组比较差异均有统计学意义(P0.05)。扫描电镜和组织学观察结果显示,术后4、8周NS组胶原纤维排列紊乱,1.0μg/mL TGF-β1组和2.0μg/mL TGF-β1组胶原纤维排列整齐。原位杂交结果显示,术后各时间点1.0μg/mL TGF-β1组TGF-β1 mRNA和colⅠmRNA表达水平均低于NS组,差异均有统计学意义(P<0.05)。结论TGF-β1中和抗体能有效抑制TGF-β1在肌腱损伤修复中的作用,减少粘连形成。

先天性肌无力综合征(CMS)是一种神经肌肉传导障碍性疾病,好发于新生儿和儿童期。神经肌肉活检和基因检测是诊断CMS的重要检查方法。该文回顾性分析1例CMS患儿的临床表现及肌肉活检表现,并复习相关文献。患儿为12岁男性,四肢无力3年入院。镜下见肌纤维明显肥大和萎缩,萎缩肌纤维以圆形、椭圆形为主,ATP酶染色示萎缩和肥大累及两型肌纤维。糖原和脂肪染色均为阴性。基因检测显示患儿colQ基因突变。CMS的治疗因不同的基因突变类型,对药物有不同的疗效。
