Colorectal cancer is the most prevalent among digestive system *** relies on disrupted control of cellular processes,such as metabolism,proliferation,DNA damage recognition and repair,and ***,tissue,organ and body physiology is characterized by periodic fluctuations driven by biological clocks operating through the clock gene *** of molecular clockworks and cellular oscillators is involved in tumorigenesis,and altered expression of clock genes has been found in cancer *** studies have shown that circadian disruption,that is,alteration of bodily temporal organization,is a cancer risk factor,and an increased incidence of colorectal neoplastic disease is reported in shift *** this review we describe the involvement of the circadian clock circuitry in colorectal carcinogenesis and the therapeutic strategies addressing temporal deregulation in colorectal cancer.

circadian clocks time developmental stages of fruit flies Drosophila melanogaster, while light/dark （LD） cycles delimit emergence of adults, conceding only during the ＂allowed gate.＂ Previous studies have revealed that time-to-emergence can be altered by mutations in the core clock gene period （per）, or by altering the length of LD cycles. Since this evidence came from studies on genetically manipulated flies, or on flies maintained under LD cycles with limited range of periods, inferences that can be drawn are limited. Moreover, the extent of shortening or lengthening of time-to-emergence re- mains yet unknown. In order to pursue this further, we assayed time-to-emergence of D. rnelanogaster under 12 different LD cycles as well as in constant light （LL） and constant dark conditions （DD）. Time-to-emergence in flies occurred earlier under LL than in LD cycles and DD. Among the LD cycles, time-to-emergence occurred earlier under T4-T8, followed by T36-T48, and then T12-T32, suggesting that egg-to-emergence duration in flies becomes shorter when the length of LD cycles deviates from 24 h, bearing a strong positive and a marginally negative correlation with day length, for values shorter and longer than 24 h, respectively. These results suggest that the extent of mismatch between the period of circadian clocks and environmental cycles determines the time-to-emergence in Drosophila.

The adipose tissue homeostasis is profoundly affected by circadian rhythms of corticosteroid secretion and chronic loss of hormonal oscillations is associated with obesity. How adipose tissue differentially responds to pulsatile vs continuous presence of glucocorticoids is poorly defined. To address this question, Bahrami-Nejad et al studied differentiation of pre-adipocytes, containing endogenously tagged CCAAT/enhancer binding protein and peroxisome proliferator-activated receptor(PPAR) γ(key regulators of adipocyte differentiation), in response to corticosteroids that were delivered either in an oscillatory fashion or continuously. The authors show that the bi-stable state of differentiation of pre-adipocytes and adipocytes was regulated by a combination of fast and slow positive feedback networks, that determined unique threshold of PPARγ in these cells. Evidently, pre-adipocytes used the fast feedback loop to reject differentiation cues of oscillating pulses of glucocorticoids and failed to differentiate into fat cells. In contrast, when glucocorticoids were delivered continuously, precursor cells exploited the slow feedback loop to embark on a path of maximal differentiation. This differential differentiation response of pre-adipocytes to pulsatile vs continuous exposure to glucocorticoids was corroborated in vivo. Thus, mice receiving non-oscillating doses of exogenous glucocorticoids, for 21 d, elicited excessive accumulation of visceral and subcutaneous fat. These data shed new light on the mechanisms of obesity caused by putative misalignment of circadian secretion of glucocorticoids or their persistently high levels due to chronic stress or Cushing's disease.

circadian clock genes in peripheral tissues usually play an important role in regulating the circadian rhythms. Light is the most important environmental signal for synchronizing endogenous rhythms with the daily light-dark cycle, and compound eyes are known as the principal circadian photoreceptor for photic entrainment in most moths. However, there is little evidence for circadian timing in compound eyes. In the current study, we isolated the timeless gene, designated Ha-tim (GenBank accession number: KM233162), from the cotton bo 11 worm Helicoverpa armigera. Ha-tim and period (Haper) showed low messenger RNA levels in the compound eyes compared to the other tested adult organs. Ha-tim and Ha-per transcript levels were dependent on an endogenous rhythm that fluctuated over a daily cycle in the compound eyes and heads. The cycles of Hatim and Ha-per transcript levels followed similar time courses, and identical expression patterns of the two genes were observed in the compound eyes and heads. Ha-tim and Ha-per were dowregulated in the compound eyes after light exposure, copulation and starvation. These results indicated that Ha-tim and Ha-per transcript levels were regulated by endogenous and exogenous factors. Our study helped to improve our understanding of the circadian clock machinery in compound eyes and other peripheral tissues.

Human beings live in a world defined by daily cycles of light and darkness caused by the Earth’s rotation around its ***,like most living organisms,have evolved internal circadian clocks that time biological processes in anticipation of these daily environmental changes(Young and Kay,2001).The plant clock relies on core genes encoding transcription factors(TFs),which regulate each other’s expression through intricate networks of interlocking transcriptional-translational feedback loops(Nohales and Kay,2016).These loops ultimately control the expression of thousands of genes,allowing plants to adapt to daily fluctuations in light,temperature,and humidity(Covington et al.,2008).In addition to daily rhythms,the Earth experiences yearly seasonal cycles marked by longer,warmer days in spring and summer,followed by shorter,cooler days in autumn and *** changes in day length and temperature intensify with distance from the ***,the ability of plants to adjust their growth and development in anticipation of seasonal changes determines their latitudinal distribution(McMillan,1960).
