Brain-derived neurotrophic factor is a key factor in stress adaptation and avoidance of a social stress behavioral *** studies have shown that brain-derived neurotrophic factor expression in stressed mice is brain region–specific,particularly involving the corticolimbic system,including the ventral tegmental area,nucleus accumbens,prefrontal cortex,amygdala,and *** how brain-derived neurotrophic factor participates in stress processing in different brain regions will deepen our understanding of social stress *** this review,we discuss the expression and regulation of brain-derived neurotrophic factor in stress-sensitive brain regions closely related to the pathophysiology of *** focused on associated molecular pathways and neural circuits,with special attention to the brain-derived neurotrophic factor–tropomyosin receptor kinase B signaling pathway and the ventral tegmental area–nucleus accumbens dopamine *** determined that stress-induced alterations in brain-derived neurotrophic factor levels are likely related to the nature,severity,and duration of stress,especially in the above-mentioned brain regions of the corticolimbic ***,BDNF might be a biological indicator regulating stress-related processes in various brain regions.

ETS-1 is a transcription factor that is a member of the E26 transformation-specific(ETS) family. Galanin receptor 2(Gal R2), a subtype of receptors of the neuropeptide galanin, has been shown to have an antidepressant-like effect after activation in rodents. Our previous study has shown that overexpression of ETS-1 increases the expression of Gal R2 in PC12 phaeochromocytoma cells. However, whether ETS-1 has an antidepressant-like effect is still unclear. In this study, we found that chronic mild stress(CMS) decreased the expression of both ETS-1 and Gal R2 in the ventral hippocampus of rats. Meanwhile,we demonstrated that overexpression of ETS-1 increased the expression of Gal R2 in primary hippocampal ***, we showed that overexpression of ETS-1 in the ventral hippocampus counteracted the depression-like behaviors of CMS rats. Furthermore, we found that overexpression of ETS-1 increased the level of downstream phosphorylated extracellular signal-regulated protein kinases 1 and 2(p-ERK1/2) of Gal R2 in the ventral hippocampus of CMS rats. Taken together, our findings suggest that ETS-1 has an antidepressant-like effect in rats,which might be mediated by increasing the level of Gal R2 and its downstream p-ERK1/2 in the ventral hippocampus.

AIM:To evaluate the effect of chronic mild stress(CMS) on the emergence of gastric ulcers and possible modulation by octreotide,a synthetic somatostatin analogue. METHODS:Adult male Wistar rats were subjected to nine different unpredictable random stress procedures for 21 d,a multifactorial interactional animal model for *** was administered daily for 21 d at two dose levels(50 and 90μg/kg)before exposure to stress ***-and microscopical assessments were made,in addition to quantification of plasma corticosterone and gastric mucosal inflammatory,oxidative stress, and apoptotic biomarkers. RESULTS:Exposure to CMS elevated plasma corticosterone(28.3±0.6μg/dL,P=0.002),an event that was accompanied by gastric lesions(6.4±0.16 mm,P=0.01) and confirmed ***,the insult elevated gastric mucosal lipid peroxides(13±0.5 nmol/g tissue,P=0.001),tumor necrosis factor-α(3008.6±78.18 pg/g tissue,P<0.001),prostaglandin E2(117.1 ±4.31 pg/g tissue,P=0.002),and caspase-3 activity (2.4±0.14 OD/mg protein,P=0.002).Conversely,CMS mitigated interleukin-10(627.9±12.82 pg/g tissue,P= 0.001).Furthermore,in animals exposed to CMS,octreotide restored plasma corticosterone(61%and 71%from CMS,P=0.002)at both dose *** beneficial effects were associated with a remarkable suppression of gastric lesions(38%and 9%from CMS,P=0.01)and reversal of derangements in gastric mucosa. CONCLUSION:The current investigation provides evidence that exposure to CMS induces gastric ulceration, which was alleviated by administration of octreotide possibly possessing antioxidant,anti-inflammatory,and anti-apoptotic actions.

Depression is a chronic,recurring and potentially life-threatening illness that affects up to 20%of the population across the *** its prevalence and considerable impact on human,little is known about its *** of the major reasons is the restricted availability of validated animal models due to the absence of consensus on the pathology and etiology of ***,some core symptoms such as depressed mood,feeling of worthlessness,and recurring thoughts of death or suicide,are impossible to be modeled on laboratory ***,the criteria for identifying animal models of depression rely on either of the 2 principles：actions of known antidepressants and responses to *** review mainly focuses on the most widely used animal models of depression,including learned helplessness,chronic mild stress,and social defeat ***,the behavioral tests for screening antidepressants,such as forced swimming test and tail suspension test,are also *** advantages and major drawbacks of each model are *** prospective,new techniques that will be beneficial for developing novel animal models or detecting depression are discussed.

＜正＞Objective Paeoniflorin（PF） is the major bioactivity of peony root which has been used to treat depressive patients in Traditional Chinese ***,whether PF has antidepressant effect and if so,what may be the underlying mechanism remain *** Adult male,C57BL/6J mice and wistar rats were administered PF（0.25- 1mg/kg,i.p.） for 7-14 days,and then tested in the forced swimming test（FST）,novelty-suppressed feeding test（NSF） and chronic mild stress paradigm（CMS） to determine the antidepressant effects of *** immunofluorecence studies were used to investigate the effects of PF on neurogenesis in adult hippocampus and ELISA was used to quantify BDNF and VEGF *** PF produced antidepressant actions in various models,including the FST,the NSF and the CMS. Interestingly,there were more new neurons in the hippocampus in PF-treated *** was associated with an increase in BDNF in the hippocampus induced by *** PF demonstrates antidepressant-like effects in several models of *** effect was probably mediated by increased neurogenesis in a BDNF-related mechanism.

Background Arctigenin（ARG）, the main active ingredient extracted from Great Burdock Achene, has been reported to exert anti-decrepitude and antiinflammatory neuropharmacological activities. It’s potential antidepressant-like effects are still poorly understood. Objective To investigate the potential antidepressant-and anxiolytic-like effects of ARG in acute and chronic mild stress（CMS） mice model. Methods Both acute stress and CMS depression mice model were used in the present study. One group of mice were divided into three subgroups and were intraperitoneal injected with vehicle（VEH）, 10 and 30 mg/kg dose of ARG for 14 continuous days. Open field test（OFT）, novelty suppressed feeding（NSF）, sucrose preference test（SPT）, forced-swimming test（FST） and tail suspension test（TST） were conducted 24 hours after the last ARG treatment. Another group of mice were divided into na?ve group and CMS group. Mice in CMS groups were randomly divided into 4 subgroups and were treated with VEH, ARG（10 and 30 mg/kg） and fluoxetine hydrochloride（10 mg/kg） daily during the last 14 days of CMS. OFT, NSF, SPT and FST were conducted 24 hours after the last ARG treatment. Results The behavioral results showed that repeated ARG administration significantly antidepressant-and anxiolytic-like activities, including decreased immobility time of mice in the TST（10, 30 mg/kg, P ＜ 0.05）, decreased floating time in FST（10 mg/kg, P ＜ 0.05）, increased sucrose preference in the SPT（10 mg/kg, P ＜ 0.05）, increased time spent in the central zone in the OFT（10, 30 mg/kg, P ＜ 0.05）, and decreased latency time to feeding in the NSF（10, 30 mg/kg, P ＜ 0.05 respectively）, compared with VEH-treatment mice. For mice exposed to CMS, a 28-d CMS procedure could induce depressive-and anxiety-like behaviors, which could be significant blocked by repeated ARG treatment.
