Detection of CD2 expression in chicken hematogenic embryo yolk sac lymphoid cells prior to thymus genesis

Lymphoid mononuclear cells from chick embryos at stage 16 were collected prior to fetal liver and thymus genesis to study the dif- ferentiation and function of the hematogenic yolk sac and to detect whether CD2 occurs on the surface of lymphoid mononuclear cells. The phenotype and functional activity of the cell surface protein E receptor and the ultrastructure of embryonic E~+ cells were compared with those of mature T cells.Our results indicate 99.36% homology between the E receptors of embryonic lymphocytes and mature T cells.Other similarities,including molecular distribution,motivation,the ability to form an erythrocyte rosette,the structure of the receptor-ligand complex,and the conformation of the signal channel,were detected between embryonic lymphocytes and mature CD2-expressing T cells.These results indicate that CD2 is already expressed prior to fetal liver and thymus genesis and that its expression is not dependent on the thymic microenvironment.

The paper was to study the inhibitory effect of recombinant freeze-drying chicken interferon against Newcastle disease virus （NDV） F48E10. Nine-day-old Chicken embryos were inoculated with recombinant freeze-drying chicken interferon via allaotoic sack, while 10-day-old Chicken embryos were inoculated with NDV F48E10, and in vivo protective efficacy of interferon on chichen embryos was studied. The results showed that the recombinant freeze-drying chicken interferon at the dose of 1.28 mg/embryo reached the protection ratio of 90% on Chicken embryo infected by F48E10.

Medium-chain fatty acids and their derivatives are natural ingredients that support immunological functions in animals.The effects of glycerol monolaurate(GML)on intestinal innate immunity and associated molecular mechanisms were investigated using a Chicken embryo model.Sixty-four Arbor Acres broiler embryos were randomly allocated into four groups.On embryonic day 17.5,the broiler embryos were administered with 9 mg of GML,which was followed by a 12-h incubation period and a12-h challenge with 32μg of lipopolysaccharide(LPS).On embryonic day 18.5,the jejunum and ileum were harvested.Results indicated that GML reversed the LPS-induced decline in villus height and upregulated the expression of mucin 2(Pembryos.

Lipid transport to avian oocytes and to the developing embryo

Studies of receptor-mediated lipoprotein metabolic pathways in avian species have revealed that physiological intricacies of specific cell types are highly analogous to those in mammals. A prime example for the power of com- parative studies across different animal kingdoms, elucidated in the chicken, is that the expression of different lipo- protein receptors in somatic cells and oocytes are the key to oocyte growth. In avian species, yolk precursor transport from the hen＇s liver to rapidly growing oocytes and the subsequent transfer of yolk nutrients via the yolk sac to the developing embryo are highly efficient processes. Oocytes grow from a diameter of 5 mm to 2.5-3 cm in only 7 days, and the yolk sac transfers nutrients from the yolk stored in the mature oocyte to the embryo within just 2 weeks. The underlying key transport mechanism is receptor-mediated endocytosis of macromolecules, i.e., of hepatically synthesized yolk precursors for oocyte growth, and of mature yolk components for embryo nutrition, respectively. Recently, the receptors involved, as well as the role of lipoprotein synthesis in the yolk sac have been identified. As outlined here, lipoprotein degradation/resynthesis cycles and the expression of lipoprotein receptors are not only coordinated with the establishment of the tbllicular architecture embedding the oocyte, but also with the generation of the yolk sac vasculature essential for nutrient transfer to the embryo.

Background:Studies of molecular mechanism of lipid metabolism of broiler liver tissue during embryonic period are important for broiler production and breeding.The results of the studies both on mammals and on the chicken showed that the lipid metabolism activities of the liver tissue are complex,and Liver plays vital roles on lipid metabolism. Methods:In the current study,Fluorscence difference gel electrophoresis(2D-DIGE) method combined with matrixassisted laser desorption-ionization time-of-flight tandem mass spectrometry(MALDI-TOF-TOF-MS) were performed to analyses protein expression profiles of liver tissue of the Chicken embryos including 7-old-day、12-old-day、14-old-day、17-oldday and 1-old-day after hatching of fourteenth generation of Northeast Agricultural University broiler lines divergently selected for abdominal fat content(NEAUHLF). Results:A total of 22 differentially expressed protein spots were found in liver adipose tissue between fat and lean broilers.Among them,14 protein spots were downregulated and 8 protein spots were upregulated in fat birds compared with those in lean birds.These 22 protein spots were then identified by MALDI-TOF-TOF-MS and matched to 17 proteins by searching against the NCBInr and UniProt databases,including apolipoprotein A-I(APO A-I),intestine fatty acid binding protein(I-FABP),lysyl-tRNA synthetase,Tyrosyl-tRNA synthetase,cytoplasmic and Nucleoside diphosphate kinase(NDK),Alpha-fetoprotein(AFP), Succinate dehydrogenase[ubiquinone]flavoprotein subunit,mitochondrial(SDHA), Ovalbumin,sulfotransferase,heat shock proteinβ1(HSPβi),thioredoxin domain-containing protein 5 and 3 hypothetical proteins.These proteins are mainly related to lipid metabolism, energy metabolism,chaperone,transport.KEGG pathway analyses revealed that these 17 differentially expressed proteins were involved in the tricarboxylic acid cycle pathway and oxidative phosphorylation pathway. A one-step real-time RT-PCR assay(rRT-PCR) was developed for efficient detection of Duck hepatitis virus typel(DHV-1).A pair of specific primers was designed against the conserved region in the 3D gene that encodes the RNA dependent RNA polymerase with a single conserved TaqMan? probe.The detection limit of this assay was 10 viral genomic copies per reaction and it was highly specific to DHV-1.The rRT-PCR assay was used to determine the distribution and concentration of DHV-1 virulent strain in duck embryos as well as the DHV-1 attenuated vaccine strain in Chicken embryos.The results revealed that the copy numbers of DHV-1 reached a peak in duck embryos and Chicken embryos at 28-40h,44-56h post inoculation respectively. The comparative tests for ducklings infected artificially and clinical samples between neutralization test and rRT-PCR showed that the positive results of infected samples were the same,while the rRT-PCR method was more sensitive than neutralization test for detection of clinical samples.The rapid,sensitive and specific rRTPCR assay will be a powerful tool for detection of suspected cases of DHV-1,distribution pattern of DHV-1 in vivo and molecular epidemiological screening.

A one-step real-time reverse-transcription polymerase chain reaction assay (rRT-PCR) was developed for efficient detection of Duck hepatitis virus type 1 (DHV-1).A pair of specific primers which showed 87bp fragment was designed against the conserved region in the 3D gene with a single conserved TaqManTM probe.It could obtain excellent linear when the DHV-1 RNA concentration between 1×10 and 1×10 copies.The detection limit of this assay was 10 viral genomic copies per reaction and it was highly specific to DHV-1 genera.Then,the rRT-PCR assay was used to determine the distribution and concentration of DHV-1 virulent strain in duck embryos as well as the DHV-1 less-virulent strain in Chicken embryos.The results revealed that the copy number of DHV-1 reached a peak in duck embryos and Chicken embryos at 28-40h,44-56h post inoculation respectively,maintaining 10 copies/g level in embryoid bodies and 10-10 copies/ml level in allantoic fluid.The RNA copy number in chorioallantoic membrance and muscle was higher than that in allantoic fluid,heart,liver,spleen,brain,intestine from dead duck and Chicken embryos inoculated with DHV-I.Using this method and neutralization test to detect 10 liver samples taking from ducklings after artificially infected and 40 clinical liver samples taking from suspected ducklings showed that the positive results of artificially infected samples were the same,while the rRT-PCR method was more sensitive than neutralization test for clinical samples detection,and the positive rate was significant difference (P<0.01).In conclusion,the rRT-PCR assay is rapid,sensitive, specific,reliable and quantitive.It will be a powerful tool for DHV-1 suspected case detection,distribution pattern of DHV-1 in vivo and molecular epidemiological screening.
