The intestinal health of chick embryos is vital for their life-long growth,and exogenous nutrition intervention may provide sufficient nutrition for embryonic development.In the present study,we investigated the effect of in ovo injection of L-methionine(L-Met)on the intestinal structure and barrier function of chick embryos.There were 4 groups of treatments:the control(CON)group injected with phosphate-buffered saline(PBS)and the other 3 groups injected with 5,10,and 20 mg L-Met/egg,respectively.The injection was performed on embryonic day 9(E9),and intestinal samples were collected on the day of hatching for analysis.The results showed that,compared with the CON group,the groups administered an in ovo injection of L-Met increased relative weights of the duodenum,jejunum,and ileum(Pchick embryos due to the activation of the JAK2/STAT3 signaling pathway.Our results may provide new insights for regulating the intestinal development of embryonic chicks and the rapid growth of chicks after hatching.

Background: D-Limonene, a monoterpene from citrus fruit has been found to have chemopreventive and chemotherapeutic activities in various types of cancers. In this study, we evaluated the in vivo effect of D-Limonene on a K562-induced model of chronic myeloid leukemia(CML) in C57 BL/6 mice.Method: The tail vein injection model of K562 cells in immunocompromised C57 BL/6 mice was developed and evaluated for characteristics of the disease. The mice were treated with D-Limonene and evaluated for haematological parameters. We also evaluated the effect of D-Limonene on angiogenesis using the chick chorioallantoic membrane(CAM) assay.Results: In a complete blood count, a significant dose-dependent reduction in white blood cell, neutrophil and lymphocyte counts, but an elevation in red blood cell count and haemoglobin content was observed with D-Limonene treatment compared to the disease control or untreated group. In the CAM assay, D-Limonene produced a significant dose-dependent reduction in number of blood vessels in treatment groups compared to the vehicle-treated group.Conclusion: These studies suggest promising anti-leukemic and anti-angiogenic effects of D-Limonene in the treatment of CML.

Nutritional stimulation of the developing small intestine of chick embryos can be conducted by in-ovo feeding(IOF).We hypothesized that IOF of glutamine and leucine can enhance small intestinal development by promoting proliferation and differentiation of multipotent small intestinal epithelial cells.Broiler embryos(n=128)were subject to IOF of glutamine(IOF-Gln),leucine(IOF-Leu),NaCl(IOF-NaCl)or no injection(control)at embryonic d 17(E 17).Multipotent,progenitor and differentiated cells were located and quantified in the small intestinal epithelium between E 17 and d 7 after hatch(D 7)in all treatment groups by immunofluorescence of SRY-box transcription factor 9(Sox9)and proliferating cell nuclear antigen(PCNA),in-situ hybridization of leucine-rich repeat containing G-protein coupled receptor 5(Lgr5)and peptide transporter 1(PepT1)and histochemical goblet cell staining.The effects of IOF treatments at E 19(48 h post-IOF),in comparison to control embryos,were as follows:total cell counts increased by 40%,33%and 19%,and multipotent cell counts increased by 52%,50%and 38%,in IOF-Gln,IOF-Leu and IOF-NaCl embryos,respectively.Only IOF-Gln embryos exhibited a significance,36%increase in progenitor cell counts.All IOF treatments shifted Lgr5+ stem cell localizations to villus bottoms.The differentiated,PepT1+region of the villi was 1.9 and 1.3-fold longer in IOF-Gln and IOF-Leu embryos,respectively,while goblet cell densities decreased by 20%in IOF-Gln embryos.Postehatch,crypt and villi epithelial cell counts were significantly higher IOF-Gln chicks,compared to control chicks(P<0.05).We conclude IOF of glutamine stimulates small intestinal maturation and functionality during the peri-hatch period by promoting multipotent cell proliferation and differentiation,resulting in enhanced compartmentalization of multipotent and differentiated cell niches and expansions of the absorptive surface area.

The current research aimed to optimize the preparation technology of tributyrin nano-emulsion(TBNE)to improve its digestion properties, which were investigated through digestion models in vitro and in vivo. The content of each component of TBNE was optimized by response surface methodology(RSM) to improve the z-average particle size and stability of TBNE. The optimized TBNE was evaluated for its digestion properties by in vitro and in vivo models. The preparation conditions of TBNE optimized by RSM were as follows: 61.3% of sorbitol, 32.7% of tributyrin, and 6.0% of modified phospholipid(MP). The predicted z-average particle size of TBNE was(246.02 ± 18.10) nm. The results of the verification test showed that the z-average particle size, zeta potential, conductivity, emulsification activity index, and emulsification stability index of TBNE were(250.02 ± 7.18) nm,(-40.23 ± 0.76) m V,(31.80 ± 2.09) μS/cm,(848.00 ± 84.53) min and(1.14 ± 0.02) m^(2)/g, respectively. The in vitro digestion experiment results showed that the TBNE remained stable in the stomach and was released in the intestine, while the size of TBNE in the gastrointestinal tact was significantly smaller than that of tributyrin(P chicks(P < 0.05), and the butyric acid content in the ileal chyme from TBNE and tributyrin treatment were172.18 mg/m L and 100.85 mg/m L(P < 0.01). Therefore, the established TBNE technology in this study could be supposed to improve the digestion properties of tributyrin.

Lens fibre differentiation is a life-long process related with lens transparency, and is particularly intense during development, being related with an FGF-2 antero-posterior gradient at the equator level as the main growth factor involved which has been related with the basal membrane of the lens anlagen known as “Lens capsule”. However the lens fibre differentiation induced by FGF2 depends, as in other biological systems, on the local bioavailability of FGF-2 regulated by their relationship with extracellular matrix molecules as Heparan Sulphate Proteoglycans. Here, we try to clarify how Perlecan (a heparan sulphate proteoglycan specific from basement membranes) is involved in lens fibre differentiation at earliest stages of eye development. Our results show that Perlecan, is a major component in the lens capsule during the earliest stages of lens development in chick embryos being present during lens plate induction, lens vesicle stage and the onset of lens fibre differentiation. In order to demonstrate a direct involvement of HSPG-Perlecan in lens fibre differentiation, we generate depleted lenses by HSPG-Perlecan synthesis disruption and specific enzymatic digestion. The HSPG-Perlecan depleted lens show a significant delay or abolition in the lens fibre differentiation which remains in an immature cells displaying DNA synthesis in the posterior epithelium and a decrease in FGF2 lens expression. These data support the hypothesis that lens capsule HSPG-Perlecan is a key molecule involved in lens fibre differentiation during development, probably by involvement in FGF-2 biodisponibility.

Performance of chick pea under the influence of gibberellic acid and oxygenated peptone during germination

The experiments were carried out at the Post Graduate Research Center, to study the influence of Gibberellic Acid (50 ppm) and Oxygenated Peptone (1% aqueous solution) on chick pea (Cicer arietinum L. cv. Vijay) during germination by giving pre-sowing soaking treatment for 6 hours using petriplate method. Both the treatments enhanced the germination process. GA treatment was useful to increase shoot length, mobilization efficiency, emergence index, speed of germination and co-efficient of germination while oxygenated peptone showed an upper hand in root length, shoot/root ratio, biomass and vigour index. GA led to comparatively more synthesis of nucleic acids while oxygenated peptone showed more increase in total carbohydrates and soluble protein content. However, the activity of enzymes like amylase, catalase and protease showed upper hand with oxygenated peptone as compared to GA. In fact GA is costlier and can not be used in organic farming as it enters metabolic pathways of plant and alters them. Hence the use of oxygenated peptone is recommended being less expensive and usable under organic farming condition as it does not enter the plant metabolic pathways and yet brings about significant positive effect.

Spontaneous and light-induced photon emission from intact brains of chick embryos

Photon emission (PE) and light-induced photon emission(LPE) of intact brains isolated from chick embryos have been measured by using the single photon counting device. Experimental results showed that the intensi-ty level of photon emission was detected to be higher from intact brain than from the medium in which the brain was immerged during measuring, and the emission intensity was related to the developmental stages, the healthy situation of the measured embryos, and the freshness of isolated brains as well. After white light illumination, a short-life de-layed emission from intact brains was observed, and its relaxation behavior followed a hyperbolic rather than an expo-nential law. According to the hypothesis of biophoton emission originating from a delocalized coherent electromagnetic field and Frohlich’s idea of coherent long-range interactions in biological systems, discussions were made on the signifi-cance of photon emission in studying cell communication, biological regulation, living system’
