Comparison of trigeminal lateralization with differing stimulants

Trigeminal-specific stimulants have been shown to activate different receptors preferentially and this likely accounts for variation in sensory *** is unclear whether trigeminal sensitivity is similar across different transient receptor potential(TRP)receptors or if dysfunction of different receptors results in differing patient ***,a prospective cohort study was conducted,consisting of trigeminal lateralization testing with three different stimulants(eucalyptol,isothiocyanate,acetic acid),olfaction testing with Sniffin'Sticks,and measurement of various patient‐reported outcome measures(PROMs).A total of 50 participants were enrolled across the olfactory *** TDI score was 27.1±8.3(range 7.0–39.5)with 38%normosmic and 62%*** trigeminal lateralization scores out of 20 in the overall cohort were 16.18(2.78)for eucalyptol,14.94(3.49)for mustard oil,and 15.28(3.68)for *** showed a significant correlation with threshold scores of Sniffin'Sticks.A significant correlation was found between acetic acid and various *** of the lateralization scores of the trigeminal stimulants correlated to each other significantly and there was no correlation to *** lack of correlation suggests that the measured sensitivity of one type of TRP receptor may not translate to similar sensitivity of the other *** investigations with TRPV1 and TRPA1 agonists are needed to corroborate our findings.

A small number of male-biased candidate pheromone receptors are expressed in large subsets of the olfactory sensory neurons in the antennae of drones from the European honey bee Apis mellifera

In the European honey bee(Apis mellifera),the olfactory system is essential for foraging and intraspecific communication via *** bees are equipped with a large repertoire of olfactory receptors belonging to the insect odorant receptor(OR)*** studies have indicated that the transcription level of a few OR types including OR11,a receptor activated by the queen-released pheromone compound(2£)-9-oxodecenoic acid(9-ODA),is significantly higher in the antenna of males(drones)than in female ***,the number and distribution of antennal cells expressing male-biased ORs is ***,we analyzed antennal sections from bees by in situ hybridization for the expression of the male-biased receptors OR11,OR18,and *** results demonstrate that these receptors are expressed in only moderate numbers of cells in the antennae of females(workers and queens),whereas substantially higher cell numbers express these ORs in ***,the reported male-biased transcript levels are due to sex-specific differences in the number of antennal cells expressing these *** analyses for OR11 and OR18 in drone antennae revealed expression in two distinct subsets of olfactory sensory neurons(OSNs)that in total account for approximately 69%of the OR-positive *** high percentages of OSNs expressing given receptors are reminiscent of male-biased ORs in moths that mediate the detection of female-released sex pheromone ***,our findings indicate remarkable similarities between male antennae of bees and moths and support the concept that male-biased ORs in bee drones serve the detection of female-emitted sex pheromones.

Influence of medications on taste and smell

Medications frequently have chemosensory side effects that can adversely affect compliance with medical treatment regimens. Hundreds of drugs have been reported to induce unpleasant tastes and/or odors as well as altered chemosensations when administered alone or in combination with other medications. Some chemosensory complaints are due to the sensory properties of the drug itself such as aversive bitter and metallic tastes. However, most chemo-sensory side effects of drugs are due to alterations in the transduction pathways, biochemical targets, enzymes, and transporters by the offending medications. Studies of chemosensory perception in medicated older individuals have found that taste and smell loss is greatest for those consuming the largest number of prescription drugs. There are no standard treat-ments for drug-induced chemosensory disorders because each drug has unique biological ef-fects. However, there are a few treatment options to ameliorate chemosensory alterations including addition of simulated flavors to food to compensate for losses and to override offend-ing tastes and smells.

Caenorhabditis elegans live in the soil, a complex environment. They has developed astructure-simple but fully functional system of neurons,which make them become amodel organism for studying the development of neurons system and the related behaviors.C. elegans can employ chemosensory neurons to detect a wide range of environmentalfactors that are commonly associated with the food or dangerous clues, distinguish thesurroundings, and finally change their behaviors, such as chemotaxis, aversion or someother stress behaviors, to response the *** is one of the most versatile and sensitive strategies to seek food and avoiddanger by sensing volatile chemicals. The recent researches have suggested that somepathogenic microorganisms, including the pathogenic bacteria Pseudomonas aeruginosaand Serratia marcescens, can attract nematodes via producing the attractive volatile chemicalsas the signals. But how the worms mistake the perception of compounds as well asthe underlying molecular mechanism remains unknown. Therefore, our current studyshowed that the pathogenic bacteria *** B16 can at least secrete three volatilechemicals, benzaldehyde, 2-heptanone and 2-ethyl alcohol, which have the remarkableabilities to lure C. elegans. Among the attractive signals, benzaldehyde and 2-heptanonewere detected by AWC olfactory neurons, and 2-ethyl alcohol was detected by AWA olfactoryneurons. Furthermore, chemotaxis assay was used to identify the response ofmutants to 2-heptanone. It was found that the worms with the str-2 gene mutation weredefective in their response to 2-heptanone, but exhibited wild-type behavior in their responsesto benzadelhyde and 2-ethyl alcohol. After str-2 was expressed in mammaliancells HEK293 T, assay of the Ca2+ revealed that the receptor STR-2 mediated the elevationof Ca2+ in response to 2-heptanone, implying STR-2 is the candidate receptor of 2-heptanone. Differential expression of pancreatic protein and chemosensing receptor m RNAs in NKCC1-null intestine

AIM: To investigate the intestinal functions of the NKCC1 Na^+-K^+-2Cl cotransporter(SLC12a2 gene), differential m RNA expression changes in NKCC1-null intestine were ***: Microarray analysis of m RNA from intestines of adult wild-type mice and gene-targeted NKCC1-null mice(n = 6 of each genotype) was performed to identify patterns of differential gene expression changes. Differential expression patterns were further examined by Gene Ontology analysis using the online Gorilla program, and expression changes of selected genes were verified using northern blot analysis and quantitative real time-polymerase chain reaction. Histological staining and immunofluorescence were performed to identify cell types in which upregulated pancreatic digestive enzymes were ***: Genes typically associated with pancreatic function were upregulated. These included lipase, amylase, elastase, and serine proteases indicative of pancreatic exocrine function, as well as insulin and regenerating islet genes, representative of endocrine function. Northern blot analysis and immunohistochemistry showed that differential expression of exocrine pancreas m RNAs was specific to the duodenum and localized to a subset of goblet cells. In addition, a major pattern of changes involving differential expression of olfactory receptors that function in chemical sensing, as well as other chemosensing G-protein coupled receptors, was observed. These changes in chemosensory receptor expression may be related to the failure of intestinal function and dependency on parenteral nutrition observed in humans with SLC12a2 mutations. CONCLUSION: The results suggest that loss of NKCC1 affects not only secretion, but also goblet cell function and chemosensing of intestinal contents via G-protein coupled chemosensory receptors.
