Striped snakehead(channa striata)has enormous fisheries and aquaculture importance due to its high market demand and food ***,Vietnam strain of the species has been introduced to Bangladesh for ***,it is essential to develop a reliable molecular technique that can distinguish the Bangladesh strain from the Vietnam ones as biological invasion by feral exotic fish to the wild may cause enduring damage to ecosystem and species *** clips from snakehead samples were collected from six locations and total DNA was *** cytochrome oxidase c 1(COI)gene fragment was amplified by PCR using the Fish F1/R1 primer set and sequenced for analysis of intra-and inter-strain genetic diversity and detection of single nucleotide polymorphism to distinguish the Vietnam strain from Bangladesh ones.A total of 15 haplotypes were identified with an overall haplotype diversity of *** median-joining network created based on COI gene sequences clearly differentiated the Bangladesh strain from the strain of Vietnamese origin by a total of 18 single nucleotide *** In silico restriction analysis revealed AluI was the most suitable restriction enzyme that produced diagnostic restriction profiles of the two strains of ***-RFLP analysis by the restriction enzyme AluI created distinguishing restriction profile similar to that in the In silico *** work illustrates a simple,reliable and inexpensive molecular tool for the identification of the Bangladesh and Vietnam strains of *** by one-step PCR and restriction of the COI gene fragment.

Potential effect of striatin(DLBS0333),a bioactive protein fraction isolated from channa striata for wound treatment

Objective:To characterize proteins and other nutrients in striatin(DLBS0333),a bioactive protein fraction isolated from snakehead fish(channa striata) and to investigate its wound healing ***:Proteins and other constituents in striatin were characterized by sodium dodecyl sulfate-polyacrylamide gel electrophoresis,two dimension electrophoresis,immunoblotting assay,spectroscopy and high performance liquid *** wound healing activity of striatin was studied in vitro using 3T3 fibroblast cells and in vivo using wound-induced animal *** parameters related to wound healing process were ***:Striatin contained four major bioactive proteins with approximate molecular weight of 8.3,10.9,15.4 and 16.7 *** addition to proteins,striatin also contained amino acids(10 essential and 7 non essential amino acids),fatty acids(palmitic acid,oleic acid,stearic acid,linoleic acid,arachidonic acid),vitamins(vitamin A,vitamin B_6)and other nutrients(carbohydrate,dietary fiber,biotin,choline,inositol,L-carnitine,selenium) which are potential for wound healing and increasing serum albumin *** with striatin both in vitro and in vivo indicated that striatin enhanced cell ***-induced animal model treated with striatin showed significantly faster wound healing process,as confirmed by wound size and faster serum albumin level *** striatin does not contain albumin,this bioactive protein fraction may lead to enhanced albumin synthesis in the liver thereby maintaining the blood albumin ***,consuming striatin is a better option to improve albumin levels in diseased condition and the condition of being ***:Striatin(DLBS0333) is a potential natural compound for accelerating wound healing in conditions such as post surgery and post partum,and increasing albumin level.

The variations in the capacity of fish species to biosynthesize the n-3 highly polyunsaturated fatty acids （HUFA）, eicosapentaenoic （EPA） and docosahexaenoic （DHA） acids which are crucial to the health of higher vertebrates has been well documented. HUFA biosynthesis has been reported to involve enzyme-mediated fatty acyl desaturation and elongation processes. In previous studies, various authors have cloned, characterized and identified different genes for fatty acyl desaturase and elongase （FAD and FAE, respectively） enzymes from fish species such as Atlantic salmon and zebra fish, and have also shown the up and down regulation of the activity of these enzymes in response to fluctuations in dietary HUFA. In this paper, we report on the tissue distribution in the expression of genes （mRNA） for FAD and FAE enzymes in channa striata （Bloch, 1793） fingerlings, to evaluate the tissues of the fish which show the highest activity of these enzymes. For this purpose, mRNA from 8 different tissues of the fish （maintained on a commercial diet） were isolated and analyzed, using conventional PCR technique. The tissue distribution of the genes was estimated to be significantly （P＜0.05） higher for both FAD and FAE enzymes in the liver and brain of C. striata than in the other 6 tissues measured （muscle, ovary, testis, gill and skin） respectively. The implication of this observation with respect to the use of vegetable oils to substitute fish oil in diets for freshwater fish species is discussed accordingly.
