Construction and Identification of cDNA Library from chaetomium cupreum

chaetomium cupreum is one kind of biocontrol fungus. To promote the application of rare earths on agriculture, La3+ and Ce3+ were utilized to observe the effect on C. cupreum antagonism. The results show that low concentration of Ce(NO3)3 and La(NO3)3 can increase the biocontrol ability of C. cupreum (cupreum. With mycelia of C. cupreum, a cDNA library was constructed and quality identification was carried out. The titer of library was 0.93×106 pfu·ml-1, the recombinant efficiency was 94.2%, and the average insert size was over 1.3 kb with 94% cDNA fragments over 700 bp. Partial clones of the cDNA library were randomly selected to be sequenced from 5′end and 110 high quality ESTs were obtained; Those ESTs were assembled to 91 unigenes with 10 Contigs and 81 Singlets. 36 (39.6%) of those clones showed significant homology to known genes representing known genes. 55 (60.4%) cDNA clones represented novel genes show no significant homology to nr-protein database of NCBI. The cDNA library built a base for deeper research towards C. cupreum biocontrol mechamism.

chaetomium cupreum is a saprophytic fungus widely used as an efficient biological *** investigate its molecular mechanism further and build a foundation for the genome analysis,we constructed cDNA library. The titer of the library was 0.9×10 pfu/ml,the recombinant rate was 94.2%,the insert size was above 700bp,most of the cDNA fragments were more than 1kb. The high quality cDNA library built a profound base for further research towards chaetomium cupreum genetics and genomics.

＜正＞Endophytic chaetomium cupreum from Macleava cordata（Willd.） *** herb was identified by analysis of morphological characteristics and the sequence of the internal transcribed spacers（28S rDNA） of *** crystal,isolated from culture broth of chaetomium cupreum,was identified by X-ray single crystal diffraction as 3,3’,6,6’- tetrahydroxy-4,4’-dimethyl-1,1’-bi（cyclohexa-3,6-diene）-2,2’,5,5’-tetraone,oosporein. We demonstrated the oosporein had antifungal activity against rhizoctonia solani, botrytis cinerea,pytium ultimum,antitumor activities against HL-60 and A549 and toxicity through artemia salina.

＜正＞Research and development on biological control of plant diseases has been being undertaken in the past several decades by both government and private company sectors for taking place the chemical *** problem worldwide which associated to the use of hazardous chemicals for disease control that has been interesting attention due to the pathogen become resistant to chemical fungicide and it causes the environmental pollution and ecological *** implementation of practical integrated biological control technology of plant diseases in the fields has been successfully introduced to the growers in Thailand,Philippines,People Republic of China,Russia and Vietnam by using a new broad spectrum biological fungicides namely Ketomium （Thailand Patent No.6266,International Code:AO1N 25/12）.The biological product has been gradually developed since 1989 from 22-strains of chaetomium cupreum CC01-CC10 and chaetomium globosum CG01-CG12 in the form of pellet and powder *** has been successful applied in the infested field-soils which integrated with cultural control measures and organic amendment for long-term protection of Phytophthora rot of Durian（Durio zibethinus L）,and Tangerine（Citrus’reticulate Blanco）,Black Pepper（Piper nigram L.）and Strawberry（Fragaria spp.）,Fusarium Wilt of Tomato （Lycopersicon esculentum L.）,and Basal rot of Corn（Zea mays L.）*** concluded that the research findings on bio-control technology of plant disease in practice has successfully demonstrated either completely or partially when integrating to other control measures for appropriate disease management.
