Separation and Anti-Hantaan Virus Activity of Extracts from Alternanthera philoxcroides in vitro and in vivo

A water-soluble substance was extracted from the Chinese herb, Alternantheraphiloxcroides with hot water and alcohol. Aliquots of this initial extract were further fractionated by treatment with ether, ethyl acetate and alcohol respectively. The four extracts were assayed for anti-viral activity against three serum, Hantaan virus 114 （HV114）, HV435 and A9 strains. Results show that the four extracts are capable of inhibiting Hantaan virus propagation, of which extract No. 1 has the best efficiency. The three dosage of extract No. 1, which are used upon three Hantaan virus serum IC50, are 153, 157, 154 μg/mL. New-born mice were made to be infected with HV114 and then fed in vivo with extract No.l on the 3rd, 10th and 14th days after being infected by the virus. The treatment continued for 8 days with a dosage of 2.5 g/kg. Result shows that survival rates of mice were 75%, 50% and 0, respectively. The median time to death （MTDs） of the three groups were 37, 30, 23 days.

Methylene blue injection has a stronger direct inactivation on VZV in vitro. When the injection was diluted from 116 to 1128, which was obvious (Pcellculture method showed when the injection was diluted from 116 to 164, it also effectively inhibited the proliferation of VZV in WISH continuous celllines (P<001 and P<005). The MIC was 195, the IC50 was 145 and the IC90 was 121.

A newly established human tumor clonogenecity assay - the glass capillary tube culture technique (CAP assay) to described. With this method, we observed the in vitro anticancer efficiency of several antineoplastics on human solid tumors and acute non-lymphatic leukemia (ANLL) specimen. Comparing with the conventional two-layer agar culture assay (2-LAC assay). The CAP-assay had a higher culture success rate, higher plating efficiency (PE) and shorter experimental period. The preliminary data suggesting and that the CAP assay has the potential to replace the conventional 2- LAC assay for in vitro drug- sensitivity testing and may be applied clinically.

A new selenium source, Nano red elemental selenium (Nano-Se) was used to study the effect on the GPx activity of broiler chick kidney cells (BCKC) in vitro, Sodium selenite (Na_ 2 SeO_ 3 ) and seleno-1-methionine (Se-Met) were used as the controls. The results showed that the effects of three kinds of Se forms on the GPx activity of BCKC were accordant(p>0.05) compared with each other at 0.01,0.05 and 0.10 μmol/L Se concentrations treatments. In the range of 0.00-0.10 μmol/L Se concentrations, the GPx activity increased with elevation of Se concentrations in medium. For the three kinds of Se forms, the GPx activity reached the climax at 0.10 μmol/L Se concentration. At 0.20 and 0.30 μmol/L Se concentrations, the influnces of three kinds of Se forms were not accordant with one another. For Nano-Se, the GPx activity at 0.20 and 0.30 μmol/L Se concentrations remained the same as that at 0.10 μmol/L Se concentration treatment. For Se-Met, the GPx activity at 0.20 μmol/L Se concentration treatment remained the same with 0.10 μmol/L treatment; the GPx activity at 0.30 μmol/L Se concentration treatment was declined significantly(pSe-Met>Na_ 2 SeO_ 3 .

The growth of 111 cultures of human epidermal keratinocytes inexplant culture,dispersed cell culture and 3T3 feeder layer culturewas studied.The rate of monolayer formation was 80.0% in 3T3feeder layer cultures,52.5% in explant cultures and only 11.8% indispersed cell cultures.It is concluded that among the modelsstudied the 3T3 feeder layer culture is the most effective andpossesses many advantages for the cultivation of human keratino-cytes;the explant culture would be a simple and easy method if wecan effectively control the contamination by fibroblasts.The growthpattern of cultured keratinocytes in each model was described.

Objective To establish a method of inducing rabbit bone marrow mesenchymal stem cells (MSCs) to express chondrocytic phenotype. Methods MSCs were seperated from bone marrow extracted from the iliac of New Zealand rabbit. TGF β 1, IGF Ⅰ and Vitamin C were applied into culture medium to induce proliferation and transformation of MSCs. Colony forming efficiency(CFE) was measured to reflect cell proliferation rate; procollagen α1(Ⅱ)mRNA in cells was detected by RT PCR; ultrastructure of cells was observed under scanning electronic microscope(SEM);alkline phosphatase(ALP) in culture medium was also detected. Results Under the influence of TGF β 1, IGF Ⅰ and Vitamine C, CFE of MSCs rose from control level of 3/106 to 21/106, expression of articular cartilage specific procollagen α1(Ⅱ)mRNA by cells was promoted; At the same time, flocculent matrixes synthesized were observed between connecting polygonal MSCs. ALP in culture medium declined to control level with culture time. Conclusion Expression of chondrocytic phenotype by MSCs could be induced by the synergistic action of TGF β 1,IGF Ⅰ and Vitamine C. Growth factor induced MSCs could be a good cell source for tissue engineered cartilage.

Objective To investigate the effects of plasma from patients with acute on chronic liver failure on the proliferation and biotransformation function of C3A cells in vitro,and provide experimental data for C3A cells to be efficiently used in the bioartificial liver system.Methods C3A cells were incubated in 100%normal human plasma(NHP)and 100%abnormal plasma(AP)from patients with acute on chronic liver failure.Growth morphology of the two groups were observed under inverted microscope and scanning electron microscope.The method of methyl thiazolyl tetrazolium(MTT)was conducted for the proliferation activities of C3A cells.The cellular apoptosis rates were assessed by the flow cytometer.The biotransformation function of cells was evaluated through diazepam metabolic amount assay.The concentrations of epithelial growth factor(EGF),transforming growth factor-α(TGF-α)and interleukin-1(IL-1)were detected in plasma of the two groups.Results A:The proliferation activities of C3A cells incubated in 100%AP for 24,48,72,96 and 120 hours were significantly higher than that in 100%NHP(Pcells in 100%AP were growing faster than those in 100%NHP after cells attached to the plastic at24 and 48 hours.The same phenomena was observed under the scanning electronic microscope.C:The C3A cells cultured in both groups of plasma showed the same apoptosis rate at 48 hours and there was no statistical difference.D:The diazepam metabolic value of C3A cells incubated in 100%AP for 24,72 and 120 hours were lower than that in 100%NHP and were statistically different(Pcells,but also decreases partial biotransformation function of C3A cells.

Background: The EU ban on antimicrobial growth promoters (AGP) has initiated a search for non-antibiotic alternatives. It has been demonstrated that certain antibiotics and non-antibiotic alternatives enhance growth by inhibiting inflammatory cells, i.e. neutrophils and macrophages in the intestine. There is very little information on the effect of anti-inflammatory compounds on intestinal epithelial cells, which are known to play an important role in intestinal inflammatory responses. In order to establish this, a porcine intestinal epithelial cell line (IPEC J2) was incubated with an adherent enterotoxigenic E. coli (ETEC) to stimulate inflammation, using a non-pathogenic non-adherent E. coli (EC) as a control. The influence of the presence of the anti-inflammatory compounds Macleaya cordata extract (MCE) and acetylsalicylic acid (ASA) on inflammatory transcriptional responses was studied. Results: ETEC induced a strong inflammatory response as was most evident from the expression of IL-1β, IL-8 and TNF-α, whereas EC induced IL-1βonly. Co-incubation with MCE and ASA significantly reduced the responses of the pro-inflammatory cytokines, similarly for IL-1βand TNF-α, but ASA was more effective than MCE in reducing the IL8 response. Conclusions: The present results suggest that the in vivo anti-inflammatory growth promoting effects of AGP and effective alternatives to AGP such as MCE and ASA are not restricted to inflammatory cells and also involve the more abundant enterocytes. This suggests a major role for epithelial cells in growth promotion livestock, and it further supports the notion that effective alternatives to AGP should have anti-inflammatory activity.
