Clinical valuation of serum tumor supplied group of factors in diagnosis of Canine cancer

This study is to evaluate the clinical value of serum tumor supplied group of factors (TSGF) in diagnosis of Canine cancer. 145 dogs with malignant tumors,32 benign diseases,34 dogs with non-tumor diseases,and 6 healthy dogs. Biochemical colorimetric assay were used to detect the serum TSGF level. Blood samples were collected at the time of initial diagnostic evaluation,before each chemotherapy session,and at follow-up visits. The serum TSGF level in dogs with malignant tumors,benign diseases,non-tumor diseases and healthy dog were 67. 3 ± 22. 1 U/mL,57. 3 ± 7. 58 U/mL,60. 2 ± 14. 2 U/mL and 54. 6 ± 3. 21 U/mL,the first group was statistically significant higher than the latter (P < 0. 05). 56. 6% malignant patients exceeded the upper limit of the normal reference range (61 U/mL). The samples were classified to groups according clinic stage. The TSGF level from malignant tumor groups were stage I 56. 8 ± 5. 93 U/mL. stage II 66. 7 ± 25. 7 U/mL,stage III 75. 6 ± 22. 7 U/mL and stage IV 73. 9 ± 29. 6 U/mL. Excluding stage I, the TSGF level of all stages are statistically significant higher than healthy group,respectively,the stage III was also statistically significant higher than stage I. Further follow-up study indicated that in entirely insect group,the TSGF level decreased after treatment and kept in low level. In the partly insect group,although the TSGF activity decreased in a short term after surgery,the TSGF level would increase again and kept in high level. From this fact we considered that TSGF level is expected to be a newly potential biomarker in screening neoplastic diseases in veterinary and a monitoring factor for predicting relapse.

The Electrophysiologic Effects of Sotalol on the Action Potential of the Nascent. Canine Purkinje's Fibers

Utilizing standard microelectrode stimulation and recording techniques, the electrophysiologic effects of the new antiarrhythmic agent sotalol were studied in Purkinje′s fibers obtained from six nascent mongrel Canines (ages 5～15 days) and were compared to changes observed in the adult Canine Purkinje′s fibers (n=6). A dose dependent lengthening of the faction potential duration (APD) was observed in the nascent group and the overall increase in APD was significantly greater than that observed in the adult group. Sotalol also produced dose dependent changes in the effective refractory. period (ERP) in both groups though the overall increase in ERP in the nascent group was again greater. We conclude that the immature Canine heart appears to be quite sensitive to the class Ⅲactions of sotalol, with changes in APD and ERP being comparable if not greater in magnitude than that observed in the adult.

Ischemic preconditioning enhances donor lung preservation in Canine lung transplantation

Methods Ten pairs of adult Canines underwent left lung allotransplantation. Five donors were treated with ischemic preconditioning [their left hilus was clamped for 10 minutes and released for 15 minutes (Group IP)], and five donors were not treated with ischemic preconditioning (Group C). The donor lungs were flushed with 4℃ Euro Collin's solution (ECS) and stored in the same solution for two and a half hour, then transplanted to the recipient Canines, who were observed for one to two hours after transplantation. The lung venous blood of the recipient and the donor lung tissue were collected just after thoracotomy and one hour after reperfusion of the transplanted lung in both groups. Results The number of polymorphonuclear (PMN) was significantly higher in Group IP than in Group C (P<0.05). However, the number of PMN in lung interstitium under microscope was less in Group IP than in Group C. The thromboxane (TXB 2), malondialdehyde (MDA) and mean pulmonary artery pressure (MPAP) contents were significantly lower in Group IP than in Group C (P<0.05). The superoxide dismutase (SOD) and the lung venous blood oxygen tension (PvO 2) contents were significantly higher in Group IP than in Group C (P<0.05). Histological findings showed less damages in Group IP than in Group C. Conclusions The protective effect of ischemic preconditioning together with ECS flush and storage is superior to using ECS alone. The possible mechanisms may be that ischemic preconditioning inhibits the accumulation and activation of PMN in lung tissue and reduces the production of oxygen free radicals.

Endothelium-dependent relaxation of Canine pulmonary artery endothelium after prolonged preservation

Abstract Objective Experiments were designed to investigate the effect of Euro Collins (EC) solution and University of Wisconsin (UW) solution on function of pulmonary arterial endothelium. Methods Third order Canine pulmonary artery segments were preserved in cold (4℃) UW (group 1, n=8) or EC (group 2, n=9) solutions for 16 hours. The preserved (group 1 and 2) and control (group 3, n=7) pulmonary arterial segments with and without endothelium were studied in vitro in organ chambers to measure isometric tension. Results The endothelium dependent relaxation to acetylcholine and adenosine diphosphate of group 1 and 3 were significantly better than those of group 2. Conclusions We concluded that endothelium dependent relaxation of Canine pulmonary arterial endothelium to receptor dependent acetylcholine and adenosine diphosphate were impaired after preservation with Euro Collins solution. However, endothelium dependent relaxation of pulmonary segments were well maintained after preservation with University of Wisconsin solution.

Intra-articular transplantation of porcine adipose-derived stem cells for the treatment of Canine osteoarthritis: A pilot study

AIM: To test whether intra-articular injection of porcine adipose-derived stem cells(ADSCs) can treat Canine osteoarthritis(OA).METHODS: To enroll in this study dogs must have stifle joint OA that had lasted ≥ 3 mo and been treated with OA medication without significant improvement. Three dogs fulfilled these criteria and were thus subjects for ADSCs treatment. ADSCs were isolated from abdominal adipose tissue of a 2-mo-old female Yorkshire pig. Their stem cell marker expression was examined by immunofluorescence staining. For treatment, 5 million ADSCs were injected into the diseased joint of each dog. In the next 48 h, the patient was observed for signs of inflammatory and allergic reactions. Thepatient was then discharged to the owner and, at 2, 6, and 12 wk, followed up with orthopedic assessment, owner questionnaire, X-ray imaging, and force-plate gait analysis.RESULTS: Porcine ADSCs expressed mesenchymal stem cell markers CD90 and CD105. Injection of porcine ADSCs into Canine stifle joints did not cause any inflammatory or allergic reactions. Orthopedic evaluation found improvements in two dogs, particularly at the longest time point. Owners' evaluation found increased capacity and decreased pain in all three dogs' activities such as walking and running. Radiographic evaluation did not find statistically significant differences before and after treatment. Force-plate analysis found significant improvements in all three dogs after treatment.CONCLUSION: Xenotransplantation of ADSCs for the treatment of OA is feasible. Further studies are needed to validate this novel treatment modality, which can then be implemented for the routine treatment of OA in veterinary medicine.

Surgical management of facial dog bite trauma:A contemporary perspective and review

Objective:To perform a review evaluating management of and complications stemming from dog bite trauma sustained to the head and neck over the past decade.Data Sources:PubMed and Cochrane Library.Methods:The authors searched the PubMed and Cochrane Library databases for relevant published literature.A total of 12 peer-reviewed Canine-exclusive series inclusive of 1384 patient cases describing facial dog bite trauma met inclusion criteria.Wounds including fractures,lacerations,contusions,and other soft-tissue injuries were evaluated.Demographics related to clinical course and management,operating room requirements,and antibiotic usage were compiled and analyzed.Initial trauma and surgical management complications were also assessed.Results:75.5%of patients sustaining dog bites required surgical intervention.Of these patients,7.8%suffered from postsurgical complications,including hypertrophic scarring(4.3%),postoperative infection(0.8%),or nerve deficits and persistent paresthesias(0.8%).Prophylactic antibiotics were administered to 44.3%of patients treated for facial dog bites and the overall infection rate was 5.6%.Concomitant fracture was present in 1.0%of patients.Conclusion:Primary closure,often in the OR may be necessary,with few cases requiring grafts or flaps.Surgeons should be aware that the most common complication is hypertrophic scarring.Further research is needed to elucidate the role of prophylactic antibiotics.

Effects of Atmospheric Pressure Plasma in the Treatment of Experimental Periodontitis in Beagle Dogs

Objective The specific objective of this study was to evaluate the effects of atmospheric pressure plasma(APP)in the treatment of experimental periodontitis in Beagle dogs.Methods The APP jet was diagnosed using optical emission spectroscopy and laser-induced fluorescence spectroscopy.Six Beagles received stainless steel ligatures to establish experimental periodontitis model.The teeth in the control group were subjected to conventional root surface debridement(RSD)and chlorhexidine irrigation.The APP group also started with RSD and was then subjected to plasma irradiation.Clinical analyses including plaque index,modified sulcus bleeding index,pocket depth and attachment loss(AL),as well as cone-beam computed tomography(CBCT)analysis,were performed at baseline,4th week,8th week and 12th week after treatment.Results The results showed that typical reactive oxygen and nitrogen species were found in the full spectrum and the gas temperature of APP was close to room temperature.The highest concentrations of hydroxide and oxygen were obtained at 5 mm away from the nozzle.In both groups,all values in clinical examinations were significantly lower(P<0.05)at 12th week after treatment than those at baseline.At the 12th week,the AL in clinical examinations and the bone loss in CBCT images in the APP group were significantly lower than those in the control group(P<0.05).The hematoxylin-eosin staining showed more renascent alveolar bone in the APP group than in the control group.Conclusion These findings suggested that APP has profound potential for use as an adjunct approach for periodontitis treatment.

Can Canines alone be used for age estimation in Chinese individuals when applying the Kvaal method?

Due to the secondary dentin formation,the dental pulp undergoes changes in shape throughout life.Based on this phenomenon,the Kvaal method has been applied to various populations for age estimation,and its usefulness has been verified.When applying the Kvaal method to Chinese subjects,we observed a relatively strong correlation between mandibular Canines and age.This study notes the correlation between Canines and chronological age and is the first to identify which Canine is most closely related to chronological age.In addition,a new,simpler formula is determined based on Canines according to Kvaal’s methodology.The radiographs of 360 individuals from northern China were selected,from which the widths and lengths of the pulp from four Canines were measured according to the Kvaal method.Next,inter-and intra-observer reliabilities were analyzed in order to assess the repeatability of these measurements.The correlation between measurements and age was examined,and Chinese-specific age estimation formulae were derived.The results revealed that the ratios from the left maxillary Canine exhibited the strongest correlation with age compared to the other Canines,whereas the left mandibular Canine showed the weakest correlation,which may contribute to the overall poor correlation of mandibular Canines with age.What’s more,the formula derived from the left maxillary Canine in this study displayed the highest coefficients of determination,and the formula derived from all Canines showed the lowest residuals.Both of these formulae performed better than the Chinese-specific formula derived from six different types of teeth in our previous study,which had formerly possessed the highest coefficients of determination and the lowest residuals.Thus,we concluded that Canines do play an important role in age estimation in the Chinese population,and the correlation between maxillary Canines and chronological age is stronger than that of mandibular Canines,although no distinct trend as to which side is better correlated with age was e

Investigation of DNA Sequences Related to Latency-Associated Transcripts in the Genome of Canine Herpesvirus Type 1 (CHV-1) by Means of Bioinformatics Tools

A characteristic common to herpesviruses is the ability to establish a latent infection in the hosts, a transcriptionally active region has detected during latency as well as a set of RNA that are known as Latency Associated Transcripts (LATs), their functions have been clarified in recent work. The present work was carried using different bioinformatics method in order to determine if Herpesvirus Canine 1 (CHV-1) has a region associated with latency. Our result was the selection of nine sequences candidate of micro RNA (miRNA) (MIREval 2.0 software), and 26 miRNA (miRNAFold v.1.0 software), of them, were selected 14 with real precursors of miRNA, two were found between the RL2 and RS1 genes, one in the RL2 gene and 11 in the RS1 gene. The results showed that the similarities of these regions are very low among the herpesviruses analyzed, so it was not possible to deduce the presence of the LAT gene in Canine herpesvirus type 1 with bioinformatics. On the other hand, the comparison showed that the miRNA predicted: chv1-mir-mirnafold-8 has similarity with the ebv-mir-BART7-3p of Epstein-Barr Virus (EBV), in this way, the microRNAs predicted by means of bioinformatic programs met the theoretical requirements of these molecules, however at not having a degree of preservation in other herpesviruses, the expression by CHV-1 in latency cannot be confirmed and it is necessary to identify through experimental tests.

Role of Selective Cyclo-Oxygenase-2 Inhibitor Celecoxib in Canine Osteosarcoma Cell Culture

Background: Experimental studies have shown that cyclo-oxygenase-2 (Cox2) is related to the development and progression of tumors, since this enzyme is induced and expressed by cells such as macrophages, osteoblasts, “activated” endothelial cells, and tumor cells. The activity in tumors includes proliferation, cell transformation, tumor growth, invasion and metastasis and may play an important role in carcinogenesis of the Canine osteosarcoma, since it has high expression in tissue fragments. The combination of selective Cox2 inhibitors and other treatment modalities is the basis for a new anti-cancer therapy strategy. This in vitro study exposed primary cells of five different Canine osteosarcoma cultures to selective Cox2 inhibitor at increasing concentrations and times. Results: For Cox2 negative cultures, despite the absence of differences, greater sensitivity of cells to treatment was observed. For Cox2 positive cultures, a higher number of necrotic cells were observed (P ≤ 0.05), when compared with negative cultures. For exposure times with Celecoxib doses, no difference (P > 0.05) was found between the three times analyzed for living, apoptotic and apoptotic/necrotic cells. There are similarities in the values of 24 h and 48 h, with slight reduction of living cells, increasing those undergoing apoptosis and apoptosis/necrosis. There was significance for necrosis (P ≤ 0.05). In 72 hours, a significant difference was observed between the other two previous values (P ≤ 0.05). It was found for the group of 100 μML-1, that there was a numerically greater signaling for apoptosis and lower (P = 0.08) for necrosis, and this point was the onset of the pharmacodynamic phenomenon, with drop in the values for living cells and increased number of necrotic cells, with a tendency (P = 0.08) for reducing the percentage of necrotic cells for the group of 100 μML-1 when compared to that of 10 μML-1.
