Salmonella flagella confer anti-tumor immunological effect via activating Flagellin/TLR5 signalling within tumor microenvironment

Salmonella:mediated cancer therapy has achieved remarkable anti-tumor effects in experimental animal models,but the detailed mechanism remains *** this report,the active involvement of the host immune response in this process was confirmed by comparing the tumor-suppressive effects of Salmonella in immunocompetent and immunodeficient mice bearing melanoma *** flagella are key inducers of the host immune response during bacterial infection,flagella were genetically disrupted to analyse their involvement in Salmonella-mediated cancer *** results showed that flagellum-deficient strains failed to induce significant anti-tumor effects,even when more bacteria were administered to offset the difference in invasion *** mainly activate immune cells via Flagellin/Toll-like receptor 5(TLR5)signalling ***,we showed that exogenous activation of TLR5 signalling by recombinant Flagellin and exogenous expression of TLR5 both enhanced the therapeutic efficacy of flagellum-deficient Salmonella against *** study highlighted the therapeutic value of the interaction between Salmonella and the host immune response through Flagellin/TLR5 signalling pathway during Salmonella-mediated cancer therapy,thereby suggesting the potential application of TLR5 agonists in the cancer immune therapy.

Innate lymphoid cells(ILCs)are a group of innate immune cells,which constitute the first line of defense in the immune system,together with skin and mucous *** also play an important role in maintaining the homeostasis of the body,particularly in the complex and diverse environment of the *** respond to different microenvironments,maintaining homeostasis directly or indirectly through *** a result,ILCs,with complex and pleiotropic characteristics,are associated with many gastrointestinal *** ability of transition among those subgroups makes them function as both promoting and inhibiting cells,thus affecting homeostasis and disease progressing to either alleviation or *** these special characteristics,ILCs theoretically can be used in the new generation of immunotherapy as an alternative and supplement to current tumor *** review summarizes the characteristics of ILCs with respect to category,function,and the relationship with intestinal homeostasis and gastrointestinal *** addition,potential tumor immunotherapies involving ILCs are also discussed to shed light on the perspectives of immunotherapy.

Disturbance in tumour antigen presentation processing

Purpose: Antigen presentation process is critical for T cell recognition of malignant cells. Current opinions believed that the processing of neoantigens plays a very important role in tumor immunity. Using bioinformatics methods, we are looking for biomarkers that have a greater disturbing effect in the process of tumor antigen presentation and further screening and constructing risk models, so as to use it to predict the efficacy of ***: We first classify patients according to the effectiveness of immunotherapy and the level of immune cell infiltration of the public data set. DEG analysis based on the level of immune infiltration and immunotherapy response is used to intersect with the genes we have sought that may change MHC-peptide dramatically. Gene enrichment analysis was conducted to show pathways and mainly biological process of candidate genes. Univariant COX analysis were used to screen risk genes and Lasso regression is used to construct a prognostic model to evaluate the efficacy of *** then classify the patients in validation cohort using the risk score and evaluate the overall survival of patients, immune checkpoint therapy response and immune cell infiltration within two ***: We have established a prognostic model related to tumor antigen presentation, which can well reflect the neoantigen status of the tumor, so as to make a good assessment of the responsiveness of the patient clinical immune checkpoint treatment.
