Potential natural vegetation(PNV)is a valuable reference for ecosystem renovation and has garnered increasing attention ***,there is limited knowledge on the spatio-temporal distributions,transitional processes,and underlying mechanisms of global natural vegetation,particularly in the case of ongoing climate *** this study,we visualize the spatio-temporal pattern and inter-transition procedure of global PNV,analyse the shifting distances and directions of global PNV under the influence of climatic disturbance,and explore the mechanisms of global PNV in response to temperature and precipitation *** achieve this,we utilize meteorological data,mainly temperature and precipitation,from six phases:the Last Inter-Glacial(LIG),the Last Glacial Maximum(LGM),the Mid Holocene(MH),the Present Day(PD),2030(20212040)and 2090(2081–2100),and employ a widely-accepted comprehensive and sequential classification sy–stem(cscs)for global PNV *** find that the spatial patterns of five PNV groups(forest,shrubland,savanna,grassland and tundra)generally align with their respective ecotopes,although their distributions have shifted due to fluctuating temperature and ***,we observe an unexpected transition between tundra and savanna despite their geographical *** shifts in distance and direction of five PNV groups are mainly driven by temperature and precipitation,although there is heterogeneity among these shifts for each ***,the heterogeneity observed among different global PNV groups suggests that they may possess varying capacities to adjust to and withstand the impacts of changing *** spatio-temporal distributions,mutual transitions and shift tendencies of global PNV and its underlying mechanism in face of changing climate,as revealed in this study,can significantly contribute to the development of strategies for mitigating warming and promoting re-vegetation in degraded regions worldwide.

Irrespective of positive developments of cancer treatment,the mortality due to various cancers remains high and the mechanisms of cancer initiation and the development also remains *** we know that microRNAs are considered to be a short noncoding RNA molecules consisting of 21e25 nucleotides(nt)in length and they silence their target genes by inhibiting mRNA translation or degrading the mRNA molecules by binding to their 30-untranslated(UTR)region and play a very important role in cancer *** evidences indicate that miR-21 is over expressed in cancer stem cells and plays a vital role in cell proliferation,apoptosis,and *** though an increased expression level of miR-21 has been observed in cancer stem cells,studies related to the role of miR-21 in cancer stem cells are *** main aim of this mini review is to explain the potency of miR-21 in various cancer stem cells(cscs)and as a new target for therapeutic interventions of cancer progression.

基于cscs模型模拟的潜在自然植被可以很好地反映陆地生态系统对全球气候变化的响应特征,在草地植被类型演替、时空分布动态研究等方面具有重要的意义。本研究利用气象数据和cscs模型,模拟出当前中国潜在自然植被类型的分布状况,在此基础上,结合遥感数据,分析了中国现存自然植被的空间分布特征;研究了不同现存自然植被类型的归一化植被指数动态变化,并分析其与气候变化的响应关系;引入“人类占用强度”这一概念,研究人类活动对自然植被的影响状况;借助未来气候预测数据,分析不同情景下中国潜在自然植被分布特征。主要取得如下.研究结果：1)我国潜在自然植被类型包括冻原高山草地、：冷荒漠草地、半荒漠草地、斯太普草地、温带湿润草地、温带森林草地、亚热带森林草地、热带森林草地、热荒漠草地和萨王纳草地。在2001-2010年期间,现存自然植被面积总体呈上升趋势。2)在2001-2010年间,现存自然植被类型的月平均NDVI最大值一般出现在7-9月,处于极端自然环境(寒冷、炎热、干旱等)的现存自然植被的年平均NDVI呈现减少趋势,而偏向于亚热带、热带区域的现存自然植被类型的年平均NDVI呈现增加趋势,处于中间地带的现存自然植被类型的年平均NDVI变化平缓。3)冻原高山草地、半荒漠草地、斯太普草地温带湿润草地和温带森林草地NDVI值与温度和降水之间具有显著的正相关关系;萨王纳草地与温度和降水呈显著负相关关系;冷荒漠草地和热带森林草地与温度呈显著正相关关系,而与降水呈显著负相关关系：亚热带森林草地和热荒漠草地与温度呈显著性负相关关系,而与降水呈显著性正相关。4)在2001-2010年间,我国自然植被的人类占用面积呈先增加后递减的趋势。人类活动对潜在自然植被的影响表现为：冷荒漠草地的人类占用强度有略微升高的趋势;而斯太普草地、温带湿润草地、温带森林草地、亚热带森林草地和萨王纳草地的人类占用强度呈现缓慢减少的趋势;半荒漠草地、热带森林草地和热荒漠草地的人类占用强度变化不明显;冻原高山草地所受的影响程度较轻。5)在4种不同的未来气候情景下,潜在自然植被的分布变化也各不相同。冻原高山草地在不同情景下都表现为减少趋势;冷荒漠草地和亚热带森林草地面积在RCP2.6情景下呈先增加后减少的趋势,而在其他3种情景下表现为减少趋势;半荒漠草地和萨王纳草地面积在RCP2.6情景下表现为先增加后减少趋势,而在其他3种情景下表现为增加趋势;斯太普草地在RCP4.5情景下表现为增加趋势,而在其他3种情景下表现为先增加后减少趋势;温带湿润草地和温带森林草地在4种情景下变化趋势各不相同;热带森林草地和热荒漠草地在不同情景下都表现为增加趋势。6)在未来3个时期(2030s.2050s和2080s),在***6.0和RCP8.5情景下,冻原高山草地的中心点具有向西或向西北方向移动的趋势;冷荒漠草地有向东移动(RCP4.5和RCP6.0)、无明显移动(RCP2.6)及由东向南移动(RCP8.5)的规律;半荒漠草地和热荒漠草地在4种情境下的移动方向各不相同;斯太普草地在RCP2.6情景下移动方向依次为西南、西北和东北方向,在其他3种情景下都向西南方向移动;萨王纳草地表现为由东北向西南移动;在不同情景下,温带湿润草地均向西北方向移动;亚热带森林草地在RCP2.6和RCP4.5情景下由北向东北移动,RCP6.0情景下向西移动,RCP8.5情景下向东北方向移动;热带森林草地在RCP2.6情景下移动方向依次为西南、西北和东南方向,在其他情景下向东移动。

Cancer genes and cancer stem cells in tumorigenesis:Evolutionary deep homology and controversies

肿瘤干细胞(cscs)是肿瘤内具有自我更新、分化和致瘤潜力的细胞亚群,在引起肿瘤发生、促进肿瘤生长、维持肿瘤异质性方面起着重要作用,被认为是癌症治疗失败的主要原因。近年来,基于纳米药物靶向cscs的治疗策略取得了一定的进展,例如利用纳米药物化疗、免疫治疗、基因治疗、光热治疗、光动力治疗、声动力疗法、磁热疗法等,这些治疗策略主要是通过引起肿瘤细胞凋亡来实现治疗目的。然而,由于cscs具有自我更新和多向分化的特性,能够通过增强DNA修复、增加药物排出、提高免疫逃逸、形成乏氧微环境等多种手段抵抗凋亡,导致上述策略针对cscs的治疗效果不佳。因此,我们亟需寻找一种非凋亡死亡方式以杀死cscs。铁死亡(Ferroptosis)是一种铁依赖性的,区别于细胞凋亡、细胞坏死、细胞自噬的新型的细胞程序性死亡方式,其主要机制是,在二价铁或酯氧合酶的作用下,催化细胞膜上高表达的不饱和脂肪酸,发生脂质过氧化,从而诱导细胞死亡;此外,还表现为抗氧化体系(谷胱甘肽系统)的调控核心酶GPX4的降低。铁离子代谢和脂质过氧化被认为是铁死亡发生和进展的两个调控途径。研究表明,相对于凋亡,cscs对铁死亡更敏感。然而,cscs内有较多的抗氧化性物质如谷胱甘肽(GSH),这些还原性物质的存在能把生成的有毒性的过氧化脂肪酸还原成无毒性的脂肪酸,降低了铁死亡的效果;同时,当细胞受到铁死亡相关刺激时,cscs会高表达一种五次跨膜蛋白——Prominin2,生成的Prominin2会增加铁离子共外泌体排出体外,进一步增加铁死亡的抗性,导致铁死亡治疗肿瘤失败。因此,如何在增加细胞内铁含量的同时有效降低铁外排并清除抗氧化性物质,成为能否通过铁死亡方式有效杀死cscs的关键。基于上述分析,本文设计了具有较好生物相容性和靶向性的FeOOH/siPROM2@HA纳米颗粒用于基于cscs的肿瘤治疗。首先,我们通过水热法合成了FeOOH纳米颗粒,然后,该纳米颗粒通过静电吸附负载抑制Prominin2蛋白表达的小干扰RNA(siPROM2),最后,外面包覆上透明质酸用于靶向cscs。FeOOH/siPROM2@HA纳米粒子靶向进入cscs后,溶酶体的酸性环境使FeOOH以Fe形式释放,进而被细胞内GSH将其还原成Fe,实现了细胞内Fe的增加和GSH的消耗。同时,释放的siPROM2会抑制Prominin2蛋白表达,减少铁离子共外泌体外排增加了胞内铁含量,进一步增加了cscs对铁死亡的敏感性。另外,高浓度的Fe促进了芬顿反应,在cscs内产生了更多的羟基自由基。因此,FeOOH/siPROM2@HA通过增加Fe含量和消耗GSH“双管齐下”的方式刺激cscs内脂质过氧化物(LPO)的积累,从而高效诱发cscs铁死亡。该研究结果为开发靶向cscs的铁死亡治疗策略提供了依据。

自干细胞概念引入肿瘤研究后，肿瘤干细胞（cancer stem cells，cscs）学说开始逐渐形成。Ma等报道，肝细胞型肝癌细胞株中存在功能性的肝癌干细胞（liver cancer stem cells，Lcscs）；Sell用实验证实，肝干细胞在致癌基因和化学致癌剂的诱导下可以转化成癌前细胞并最终形成肝癌。cscs学说为人类认识肿瘤提供了一个崭新的视野，特异性靶向杀伤cscs将为肿瘤治疗带来新的希望。笔者对肝癌干细胞的靶向示踪及治疗进行综述。
